    魁蚶(Scapharca broughtonii)热休克蛋白90(HSP90)基因的克隆及转录表达分析

    热休克蛋白90(HSP90)作为一种研究的常规免疫基因,在许多物种都有过报道。本研究从构建的魁蚶转录组文库中筛选得到的HSP90基因部分序列为基础,通过RACE技术获得其c DNA全长序列(命名为Sb HSP90),以期明确魁蚶HSP90基因的结构特征、组织分布及其对病原菌刺激的免疫变化规律。序列和结构分析表明,该c DNA全长2707bp,编码一个由728个氨基酸组成的多肽,该多肽含有HSP90家族共有的5个签名序列,C端高度保守的MEEVD短肽序列和ATPase结构域;预测蛋白的分子量(Mw)为83.72k Da,理论等电点(p I)为4.85;预测该蛋白无信号肽,具有4个糖基化位点。同源性及系统分析表明,Sb HSP90基因与软体动物的HSP90相似性达到83%以上,其中与长牡蛎(Crassostrea gigas)和海湾扇贝(Argopecten irradians)相似度最高达86%,与甲壳动物HSP90的相似度都在81%左右,与脊椎动物HSP90-α和HSP90-β的同源性都很接近。实时荧光定量PCR(q RT-PCR)结果表明:Sb HSP90 m RNA在魁蚶血细胞、斧足、鳃、外套膜、闭壳肌和肝胰腺和中均有表达,斧足中的表达量相对较高,而在肝胰腺中的表达量则相对较低;注射鳗弧菌后,相对于对照组,Sb HSP90基因在所检测的每个组织中m RNA水平上的表达量都显著上调(P<0.01),而且具有显著的时间依赖性和瞬时表达趋势。国家自然科学基金项目,31602142号;;中国水产科学研究院基本科研业务费项目,2017GH07号;;浙江省重中之重学科开放基金,KF2015007

    Acupoint application of Xiefu prescription in the prevention of functional constipation in long-term bedridden patients with pelvic fracture (泻腑贴方穴位贴敷预防长期卧床骨盆骨折患者功能性便秘的临床研究)

    Objective To explore the effect of acupoint application of Xiefu prescription on prevention of functional constipation in long-term bedridden patients with pelvic fracture. Methods A total of 60 patients with pelvic fracture without abnormal defecation function and constipation main symptom score ≤3 were selected and randomly divided into two groups: 30 patients in the control group received routine prevention and nursing measures, and 30 patients in the treatment group received acupoint application of Xiefu prescription based on the routine nursing interventions. The main symptoms of constipation were scored on the day of admission, the 7th and 14th day of bed rest, and the intervention of various assisted defecation measures within 14 days of bed rest was recorded. Results The total score of Main Constipation Symptoms Scale in the treatment group was lower than that in the control group on the 7th and 14th day of bed rest, respectively(P<0. 01); On the 14th day of bed rest, the scores of stool, difficulty in defecation, defecation duration, defecation frequency and abdominal distension in the treatment group were lower than those in the control group(P<0. 01). During the 14 days of bed rest, the number of assisted defecation interventions in the treatment group was less than that of the control group (P<0. 01) . Conclusion Acupoint application of Xiefu prescription and sticking therapy has a good preventive effect on functional constipation in long-term bedridden patients with pelvic fracture. It is an effective traditional Chinese medicine treatment technology, which is worthy of popularization and application. (目的 探讨泻腑贴方穴位贴敷对长期卧床单纯骨盆骨折患者功能性便秘的预防效果。方法 选取既往排便功能无异常且便秘主要症状评分≤3分的单纯骨盆骨折患者60例, 随机分为两组: 对照组(n=30)接受便秘常规预防护理, 治疗组(n=30)在对照组基础上增加泻腑贴方穴位贴敷。于入院当天、卧床第7、14天, 记录便秘主要症状评分, 并记录卧床14d内辅助排便措施介入情况。结果 卧床第7、14天治疗组便秘主要症状总分低于对照组, 差异有统计学意义(P<0. 01); 卧床第14天治疗组粪便性状、排便费力、排便时间、下坠/不尽/胀感、排便频率、腹胀条目得分低于对照组, 差异有统计学意义(P<0. 01)。治疗组辅助排便介入措施少于对照组差异有统计学意义(P< 0. 01)。结论 泻腑贴方对于长期卧床骨盆骨折患者功能性便秘有良好的预防作用, 是一种有效的中医治疗技术。

    Bias-voltage dependent ultraviolet photodetectors prepared by GaOx + ZnO mixture phase nanocrystalline thin films

    Ultraviolet (UV) photodetectors were prepared by using the GaOx + ZnO mixture phase thin films sputtered on sapphire as the photoresponse layer. The devices show good photoresponse in UV range. More interestingly, the device responsivity in the wavelength less than 280 nm range rapidly increases with increasing the applied voltage and becomes dominant for the bias &gt;= 3.0 V. X-ray diffraction, absorption and cathodoluminescence measurements firmly reveal the mixture phases in the thin films. Electric field dependent detrapping of photo-excited carriers in nanocrystals in the films shall be responsible for the observed bias-voltage dependent deep UV photoresponse of the devices
