Production of soluble human Fas ligand by Escherichia coli and Dictyostelium discoideum
人类Fas配体(hFasL)是一种细胞凋亡因子,属于肿瘤坏死因子家族,是死亡蛋白Fas的天然受体。它是一种Ⅱ型跨膜糖蛋白,通过与细胞表面的Fas受体结合可引起Fas表达阳性的细胞凋亡。由于可以诱导肿瘤细胞凋亡,hFasL在肿瘤、AIDS、感染性疾病等的治疗中有着广阔的应用前景。 本文分别在原核表达系统大肠杆菌和真核表达系统盘基网柄菌中以可溶性形式实现了hFasL的高水平表达。从重组盘基网柄菌AX3-pLu8中提取重组质粒pMB74-hFasL作为模板,PCR成功扩增编码hFasL胞外区第141~281个氨基酸的基因序列。选用质粒pET32a(+)作为原核表达载体(其上的TrxA融合标签可以改...Human Fas ligand (hFasL) is a cell apoptosis factor belonging to the tumor necrosis factor (TNF) family. 肿瘤血管靶向治疗策略的新进展
肿瘤血管生成(tumor angiogenesis)对大多数实体瘤的生长和转移具有重要意义,他是一个多步骤多因素参与的复杂病理过程.抗肿瘤血管的治疗策略分为抑制肿瘤新生血管生成和阻断肿瘤血管血流两方面.抑制肿瘤新生血管生成旨在抑制肿瘤新生血管生成的过程,目前研究的成果比较多;而阻断肿瘤血管血流则是通过快速而选择性地破坏肿瘤血管功能,使肿瘤血供受阻,对于治疗较大体积的肿瘤有显著的疗效.肿瘤血管的靶向治疗特别是阻断肿瘤血管血流将成为恶性肿瘤的一种新的、重要的常规治疗手段.本文对近年来这两方面的研究进展进行综述
Microcomputer's Data Acquisition Based on Modbus Protocol

反相高效液相色谱法分析盘基网柄菌对氨基酸的利用
为观察盘基网柄菌(Dictyostelium discoideum)在SIH培养基上对氨基酸的利用情况,以2,4-二硝基氯苯为衍生化试剂,研究了用反相高效液相色谱(RP-HPLC)测定氨基酸的方法.在60min内16种氨基酸达到基线分离,峰面积与氨基酸浓度的线性相关系数为0.992~0.999.对发酵液样品的分析结果表明,盘基网柄菌对赖氨酸的利用最为彻底,发酵后期赖氨酸已被消耗完全.蛋氨酸、色氨酸、精氨酸、组氨酸也较易被盘基网柄菌利用,而对天冬氨酸、谷氨酸、甘氨酸、苏氨酸、脯氨酸、缬氨酸的需求不大.这一代谢特点为改进盘基网柄菌培养基组成提供依据

tfee181吸收式制冷机热力学性能研究
吸收式制冷机在工业余热回收利用中有非常广泛的应用前景,能有效改善因工业生产造成的热污染。从热力学定律和火积效率定义出发,介绍了吸收式制冷机的工作原理,并在质量和能量守恒定律基础上设计了吸收制冷机各部件的数学模型。在建立的热力学方程中,有5个假定:1)系统在稳态和热平衡条件下运行;2)各部件的热损和压降可以忽略不计;3)离开发生器和吸收器的溶液是饱和的;4)发生器出口蒸汽是纯工质,且离开发生器的稀溶液是平衡的;5)节流阀前后是等熵过程。研究了各个系统参数随火积效率(G*)的变化趋势。结果表明,当发生温度不变时,蒸发温度、制冷量、放气范围和系统COP随火积效率的增加而增加,而系统循环比、泵功和系统火用效率随之下降;如果火积效率一定,蒸发温度、制冷量、放气范围和系统COP随发生温度的增加而增加,系统循环比、泵功和系统火用效率则随之下降。因此,在吸收制冷机中,可以通过改变火积效率G+*来改善系统性能
Determination of Trace Atmospheric Mercury by Two-stage Gold Amalgamation Cold Vapor Atomic Fluorescence Spectrometry Method
在已有仪器的基础上进行改装,建立了二次金汞齐冷原子荧光光谱法(CVAfS)测定大气中痕量气态元素汞(gEM)和颗粒态总汞(TPM)的方法.该方法的检出限为2 Pg,二次金汞齐的转移效率为99%,在0--2 ng范围内做标准曲线,线性关系为0.999 5,检测精密度为1.89%.运用该方法于2009年7月对厦门地区大气环境中的gEM和TPM进行了采集检测,结果显示厦门地区未受明显汞污染.该分析方法也能用于其他环境样品痕量汞的检测.A new procedure to determine trace gaseous elemental mercury(GEM) and total particulate mercury(TPM) in the atmosphere by two-stage gold amalgamation cold vapor atomic fluorescence spectroscopy was set up.The detection limit of this method is 2 pg.The transfer efficiency from the sample trap to the analyze trap was 99% and precision of 1.89% was obtained.GEM and TPM are measured at two specific areas in Xiamen during July,2009,and the results showed that there was no significant mercury pollution in Xiamen area.This method can also be used to analyze trace mercury contents in other environmental samples.国家自然科学基金项目(20707018);福建省科技重点项目(2008I0024

Multi-parameter joint optimization for double-strip high-speed pantographs to improve pantograph-catenary interaction quality
The significant increase in speed of high-speed train will cause the dynamic contact force of the pantograph-catenary system to fluctuate more severely, which poses a challenge to the study of the pantograph-catenary relationship and the design of high-speed pantographs. Good pantograph-catenary coupling quality is the essential condition to ensure safe and efficient operation of high-speed train, stable and reliable current collection, and reduction in the wear of contact wires and pantograph contact strips. Among them, the dynamic parameters of high-speed pantographs are crucial to pantograph-catenary coupling quality. With the reduction of the standard deviation of the pantograph-catenary contact force as the optimization goal, multi-parameter joint optimization designs for the high-speed pantograph with two contact strips at multiple running speeds are proposed. Moreover, combining the sensitivity analysis at the optimal solutions, with the parameters and characteristics of in-service DSA380 high-speed pantograph, the optimization proposal of DSA380 was given
