    Studies on Growth Related Gene and Population Selective Breeding for the Fujian Oyster (Crassostrea angutala)

    福建牡蛎(Crassostreaangulata)旧称僧帽牡蛎,是福建的重要养殖贝类。《2011中国渔业统计年鉴》的数据显示,2010年福建省牡蛎养殖产量达1,456,106吨,占全国的40%,居全国首位,其中主要养殖品种为福建牡蛎。本论文对福建牡蛎生长相关基因和群体选育进行了研究,拟对福建牡蛎生长相关基因做深一步的了解,同时进行了福建牡蛎群体选育,有望为福建牡蛎良种选育提供理论支持。 本研究采用SSH、454/Roche转录组文库方法,建立牡蛎早期发育不同阶段cDNA文库。SSH文库随机调取672个克隆,送交上海生工测序。获得序列653条,测序成功率97.2%,将其数据进行拼接,得到Uni...Fujian oyster (Crassostrea angulata), formerly known as mitral oysters is the Fujian important cultured shellfish.     从患病锯缘青蟹组织中分离出一种无囊膜球状病毒,直径50~60 nm.通过病理切片观察,病蟹肝胰腺、肌肉、心脏、肠和鳃等多处组织器官受到感染,该病毒分布在细胞浆中,呈晶格状排列,不形成包涵体;这种病毒会导致细胞瓦解,细胞核肿大变形,核质聚缩,核膜破损,肌丝断裂,线粒体坏死等症状.The cause of an unknown disease broken out among the mud crab Scylla serrata was studied.Examination by electron microscope showed that there were a number of globose viral particles in the tissue of hepatopancreas,muscle,heart,intestine and gill.The virion was approximately 50~60 nm in size,without envelope and consisted only of a symmetrical globose capsid and an electric dense core.Usually,a mass of mature virus formed a crystal structure and distributed over the cytoplasm.That virus caused some histopathological changes such as cells broken and desquamation,karyolysis or karyopyknosis,mitochondria oedema and crista broken,fascicle rupture or in disorder and so on.We also isolated the virion from the tissue of unhealthy mud crab,they had the same size and structure with the virus observed in the cells

    扩增僧帽牡蛎(CrASSOSTrEA gIgAS AngulATA)的表皮生长因子受体(EPIdErMAl grOWTH fACTOr rECEPTOr,Egfr)基因,并与PET32A(+)载体连接,构建PET32A(+)/Egfr重组质粒.将重组质粒导入大肠杆菌(ESCHErICHIA COlI)bl21(dE3),成功地表达并纯化了一个50ku的重组蛋白.采用rT-PCr技术,检测了僧帽牡蛎幼体附着变态前后Egfr基因表达量的变化,结果表明,Egfr基因在附着变态前的浮游幼体阶段和附着变态24H后的稚贝阶段低表达,附着后1~6H强烈表达,这预示着Egfr基因在僧帽牡蛎幼体附着变态这一生物学过程中具有重要作用.Most benthic marine invertebrates have pelagic larvae that develop to competent stage and then settle on the bottom in response to physical,biological and chemical factors.Many previous investigators focused on exploring the inductive effects of chemical cues on larval settlement and metamorphosis.In this study,a ORF of epidermal growth factor receptor(EGFR) was amplified from sengmao oyster,Crassostrea gigas angulata and the target gene was cloned into prokaryotic expressing vector,pET-32a.After transformation,a soluble protein fused with a hexahistidine affinity ligand was expressed in E.coli BL21(DE3).The 50 ku recombinant protein was highly purified using affinity chromatography.As demonstrated by RT-PCR,EGFR was strongly expressed after 1 to 6 h of attachment of sengmao oyster larvae and weekly expressed before attachment and after 24 h of metamorphosis.And the result of RT-PCR indicated that EGFR played an important role during attachment and metamorphosis of sengmao oyster larvae.国家重点基础研究发展计划(973)项目(2010CB126403

    New records of two Nudibranchia species in coastal waters of China

    裸鳃类是重要的底栖生物类群.部分裸鳃类栖息在码头、养殖网箱、浮标、船底等人工设施上的污损生物群落中.该种生境的祼鳃类国内鲜有报道.2004年11月至2007年9月期间,作者多次在福建省近海污损生物中采集到两种裸鳃类标本.结合其外部形态和内部齿舌形态,鉴定它们为东方伪二列鳃(PSEudObOrnEllA OrIEnTAlIS)及透明脊突海牛(OkEnIA PElluCIdA).这两种裸鳃类在我国近海均属首次纪录.Nudibranchia is an important benthic group.Some nudibranchs are of marine fouling organisms attaching to manual marine facilities,such as wharfs,fishing gears,buoyages,ships and etc.Though nudibranchs are seldom reported in the fouling niche of coastal waters in China,two nudibranchs are often collected as fouling organisms in Fujian Province from November 2004 to September 2007.According to exterior morphology and interior radula morphology,they are identified as Pseudobornella orientalis Baba,1932 and Okenia pellucida Burn,1967 are new records in the coastal waters of China.国家自然科学基金资助项目(40676081
