    Construction of eukaryotic vector pcDNA3.1-flag-pygo2 and expression in C6 cell

    目的构建pcDNA3.1-flag-pygo2真核表达载体并在C6细胞中进行表达。方法从小鼠脑胶质细胞中提取总RNA,RT-PCR法反转录合成c DNA,设计引物,调取目的片段,与pcDNA3.1-flag载体连接后转化大肠杆菌DH5α,LB平板筛选菌落,提取质粒。重组质粒pcDNA 3.1-flag-pygo2经过酶切鉴定及测序后,阳离子脂质体法转染C6细胞并经免疫细胞化学染色及蛋白印迹检测重组体的表达。结果重构质粒pcDNA 3.1-flag-pygo2经限制性核酸内切酶EcoRⅠ,HindⅢ酶切分析及测序检查,表明真核表达载体构建正确;瞬时转染C6细胞后,免疫细胞荧光染色及蛋白印迹检测表明转染细胞能够表达外源Pygo2基因。结论成功构建了pcDNA3.1-flag-pygo2真核表达载体并能够在真核细胞中进行表达,这为今后研究pygo2基因在胶质瘤中的作用机制奠定了基础。Objective To construct the eukaryotic expression vector pcDNA3.1-flag-pygo2 and express the combined protein in C6 cell line.Methods To extract total RNA from primary glial cell of mouse and to synthesize c DNA by RT-PCR,then design primer and clone whole segment of pygo2 gene.After the targeted gene was inserted into vector pcDNA3.1-flag,the recombined plamid was transformed into E coli DH5α for LB agar plate screening and the recombined plasmid were extracted and purified.After verification by double enzyme digestion and sequencing.,the constructed eukaryotic expression plamid was transfected to C6 cell line by lipofectamin method,finally the protein expression was detected by immunocytochemical staining as well as western blot.Results The new constructed vector was confirmed by restricted enzyme Eco R I,HindIII digestion assay and correct Pygo2 was verified by sequenceing.finally,pcDNA3.1-flag-hpygo2 can express exogenous pygo2 gene in glioma C6 cell line after transient transfection by the determination of immunocytofluorescent staining and western blot.Conclusion The new plamid pcDNA3.1-flag-pygo2 was constructed successfully,and can express fused protein in eukaryotic cell,which establish the foundation for future research on pygo2 gene function in human glioma

    EFFECT OF UNIT CELL VOLUME OF RE(Ni, Co, Mn, Ti)_5 ALLOY (RE=La, Ce, Pr, Nd) ON ITS ELECTROCHEMICAL PROPERTIES

    制备了La(Ni,Co,Mn,Ti)5,Ce(Ni,Co,Mn,Ti)5,Pr(Ni,Co,Mn,Ti)5和Nd(Ni,Co,Mn,Ti)5四种单一稀土贮氢电极合金,分别测定了它们的单胞体积、氢化物生成焓和几个主要的电化学性能指标(包括活化循环次数、最大放电容量、高倍率放电率和容量衰退速率),以分析不同稀土元素对合金电化学性能的影响.结果表明,在四种合金中,单胞体积对合金的热力学性质和电化学性能起了决定性作用,它与合金的氢化物生成焓呈简单的线性关系,对电化学性能有双重影响,使四个主要指标随单胞体积的变化均出现极大或极小值.单胞体积本身的变化与稀土元素的周期性有关.Four rare earth based AB_5 typed alloys, La(Ni, Co, Mn, Ti)5, Ce(Ni, Co, Mn, Ti)5, Pr(Ni, Co, Mn, Ti)5 and Nd(Ni, Co, Mn, Ti)5, were prepared and then their unit cell volumes (Vcell), standard enthalpies of metal hydride formation at 298 K (△H298 K), and main electrochemical characteristics, such as activation cycle number (na), maximum discharge capacity at 50 mA/g charge/discharge rate (C50,max), high-rate-dischargeability K (includes K1 and K2 ), rate of decay (-dC / dn) were measured and analyzed to reveal the effects of the rare earth elements on the electrochemical properties. The results show that the unit cell volume is the key factor in determination of the alloys' thermodynamics and electrochemical properties. The standard enthalpies of metal hydride formation at 298 K (△H298 K) are proven to vary linearly with the unit cell volume, and the four electrochemical parameters, na, C50,max, -dC/dn and K,are dualistically affected by it.     通过对2013年7月渤海海域26个站点温度、盐度、营养盐及叶绿素a(Chl a)浓度的空间分布特征及其相关性进行了分析,发现:受渤海水深和夏季陆源河流输入影响,近岸水域表现出明显的高温、低盐与高营养盐特征,且垂直变化特征不显著;在水深较深的辽东湾湾口和渤海海峡,海水呈现明显的层化现象,表层水温高于中底层,而表层盐度与营养盐浓度则低于中底层。营养盐结构分析表明,渤海夏季磷酸盐浓度存在显著的绝对与相对限制,而受河流输入影响,硅酸盐的相对限制得到显著缓解。表层Chl a浓度的高值区位于滦河及复州河河口附近海区,中层与底层的高值区则出现在滦河与黄河口附近。Chl a浓度与环境因子的相关性分析表明,盐度..

Results showed that, excretion and metabolism were power functions of body mass. Excretion were positively linearly correlated with temperature.     研究了微量 Ti在 Ml(NiCoMnTi)5合金中的作用.结果表明,在铸态条件下,Ti几乎全部以 TiNi3第二相的形式在晶界析出,退火处理后 TiNi3相消失,但 SEM和 EDS表明 Ti取代了 AB5型化合物中 A侧的稀土 Ml;而不是 B侧的 Ni.Ti在 A侧的取代量以 5%为宜.此时合金在铸态和退火态的放电容量都在 310 mA·h/g以上.进一步提高取代量虽然会改善循环稳定性,但大大降低了放电容量.Substitutional effect of small amount of Ti in hydrogen storage electrode alloy Ml(NiCoMnTi)5 was systematically studied. The results show that Ti in the alloy is almost entirely separated out as a second phase of TiNi3 at boundary of the as-cast dendrite. After annealed, TiNi3 phase disappeared, but SEM and EDS analyses indicate that the partially substituted element by Ti is mishmetal Ml, not Ni in B side as expected, i.e. Ti is an A side substituent element. The optimal substitution amount is 5% (atomic fraction), with which the corresponding alloy, both as-cast and annealed, shows a high discharge capacity over 310 mA.h/g. If more Ti is added, a greatly decreased discharge capacity will be received with only a little increase of better cycle stability.国家自然科学基金资助项目59801008;; 国家863高新技术计划资助项目 715-004-006
