    Studies of melatonin receptor in the mudskipper Boleophthalmus pectinirostris

    鱼类松果体是传达外界信息到体内的主要传感器之一,其分泌的褪黑激素与鱼类生殖活动密切相关.为了阐明大弹涂鱼松果体形态结构与其功能的关系,运用组织细胞学方法研究了大弹涂鱼松果体的形态结构.结果表明,大弹涂鱼松果体位于端脑和中脑之间,由松果体柄和终囊两部分组成,具有背囊结构.松果体柄细长,细胞排列紧密,柄中有空腔,开口与第三脑室相通;松果体终囊膨大成梨形,且有空腔,腔内有皱褶,终囊顶部表面有黑色素细胞;背囊位于终囊下方,形状不规则,其囊壁向囊腔内反复折叠,形成许多复杂、盘旋曲折的内褶,背囊内部可见血细胞.研究结果为将来揭示大弹涂鱼半月周期产卵的机制提供了基础资料.Pineal gland is a transducer that transmits environmental message to the brain in fishes.It produces melatonin to regulate fish spawning behaviors.In order to elucidate the relationship between the structure and functions of the pineal gland in the mudskipper(Boleophthalmus pectinirostris),the morphological structure of pineal organ of the mudskipper was studied using histocytology.Results showed that pineal organ of the mudskipper located between telencephalon and midbrain,was composed of pineal stalk and pineal vesicle,and had a dorsal sac.The pineal stalk was long and thin in shape,the cells arrangement was tight,and there was a cavum in the middle of the stalk.The opening of pineal organ was connected with the ventriculus tertius.Pineal vesicle was expanded as a pear in shape and had a cavum with rugas inside.The top surface of the vesicle was covered by melanophore.The dorsal sac lied under the pineal vesicle and was irregular in shape with a cavum.The inner wall of the sac cavum had many complicated,spiral and tortuous emboles.The blood cells were visible inside the dorsal sac.The data from the present study will be useful to future research on the mechanisms of spawning with semilunar periodicity in the mudskipper.国家自然科学基金项目(40976095);福建省科技计划重点项目(2008N0041

    Cloning and Expression Profile of Arylalkylamine N-acetyltranferase-2During the Spawning Season in Boleophthalmus pectinirostris

    克隆获得编码大弹涂鱼(bOlEOPHTHAlMuS PECTInIrOSTrIS)芳基烷基胺-n-乙酰转移酶亚型2(ArylAlkylAMInE nACETylTrAnSfErASE-2,AAnAT2)的CdnA序列,其核苷酸长度为630bP,可编码209个氨基酸.运用实时荧光定量聚合酶链式反应(PCr)技术,检测了生殖季节大弹涂鱼脑组织中芳基烷基胺-n-乙酰转移酶基因亚型2(AAnAT2)的月周期以及日周期的表达趋势.结果显示,AAnAT2基因具有明显的半月周期变化节律,即1个月有2个周期,每个周期均出现1个峰值.第1个峰值出现的时间在上弦月附近,第2次峰值则出现在下弦月附近,并且每个周期峰值出现的时间与大弹涂鱼产卵时间一致.此外,AAnAT2基因的表达在一天之内也呈周期性变化趋势,在12:00—15:00和03:00—06:00时,表达出现2段峰值;而在21:00和9:00时,表达量最低.本研究结果提示,AAnAT2基因通过影响褪黑素的合成和分泌,参与了大弹涂鱼半月周期产卵的调控.The mudskipper Boleophthalmus pectinirostris,a burrow-dwelling fish inhabiting intertidal mudflats,spawns around the first or last lunar quarter during the spawning season.To understand the involvement of arylalkylamine N-acetyltranferase-2(AANAT2)in this semilunar spawning rhythm,we first cloned cDNA of AANAT2from mudskipper in this study.The length of aanat2 cDNA was 630bp,encoding apolypeptide of 209amino acids.Quantitative Real-Time PCR was used to detect the semilunar and daily expression profile of aanat2 gene in the brain of mudskipper during the spawning season.The results showed that the transcriptional expression of aanat2 in the brain of B.pectinirostris appeared to have a semilunar periodic rhythm with two peaks.The two peaks coincided with the timing of spawning in this species,the first peak was observed around the first quarter moon,and the second one around the last quarter moon.The diurnal expression of aanat2in the brain of B.pectinirostris changed at a certain level,with the two higher concentrations observed during 12:00-15:00and 03:00-06:00,and the two lower ones at 21:00and 9:00.These results suggested that aanat2 may involve in the regulation of seminlunar spawning rhythm in B.pectinirostris through mediating melatonin synthesis and secretion.国家自然科学基金项目(40976095;41276129); 福建省科技重点项目(2008N0041); 厦门大学校长基金(2013121044
