    采用苯酚-氯仿法提取基因组DNA,以相应引物经PCR扩增得到采自厦门港的刺尾纺锤水蚤(Acartia spinicau-da)和太平洋纺锤水蚤(Acartia pacifica)线粒体DNA细胞色素氧化酶I亚基基因(mtCOI)片段,经过连接、转化、扩大培养最后测序得到709 bp的有效片段.结果发现这两种纺锤水蚤的mtCOI序列差异为15.6%(共15个个体,刺尾纺锤水蚤7,太平洋纺锤水蚤8),达到种间的水平,证实了刺尾纺锤水蚤和太平洋纺锤水蚤为两个种,并确定了这两种过渡型的种类属性.依据本实验的结果,对传统的形态学分类方法进行改进,使其能够正确区分不同的过渡形态.Acartia spinicauda and A.pacifica are the sibling species in southeast coast of China.Taxonomic discrimination between the two species relies on subtle morphological characteristics.They are different with each other in seasonal distribution,but appear together at the end of autumn.The most interesting thing is that their female individuals have intermediate form.For this study,the molecular population genetic diversity of A.spinicauda and A.pacifica was described based on DNA sequence variation for a 709 base pair region of the mitochondrial COI gene.Samples of the two Acartia species were collected from Xiamen waters from May to October 2004.The average genetic distance of A.spinicauda and A.pacifica is 15.6% based on the 709 bps in mtCOI fragments,it would be concluded that they are different species for a certainty.We also found two kinds of intermediate forms between this two species,and defined their taxonomic status.According to the results,we bring forward a new morphological taxonomic method to distinguish the two kinds of Acartia species and their intermediate forms

    针对水下图像颜色失真与模糊问题,提出了基于背景光估计与透射率优化的水下图像复原算法。该算法将透射率定义为直接分量透射率和后向散射分量透射率,有效提升了成像模型的完整性。首先,对红色衰减分量进行补偿并重置直方图分布范围实现色彩均衡;其次,基于亮度、梯度判别及色调判断选出最佳背景光点;然后,通过红色暗通道先验得到后向散射分量透射率,利用无退化像素点求得直接分量透射率;最后,将两个透射率及所求背景光代入成像模型获得复原图像。实验结果表明,该算法能够有效平衡图像的色度、饱和度及清晰度,视觉效果接近自然场景下的图像。</p

    Extraction of Genomic DNA and Amplification of mtDNA COI from Calanus sinicus Stored in Formaldehyde Solution

    提取了保存于5%甲醛溶液中的单只中华哲水蚤基因组DNA.经凝胶电泳后虽无清晰条带,但PCR扩增后可得线粒体DNA细胞色素氧化酶I亚基基因(mtCOI)片段.测序分析,该片段长度为710bp,与GenBank中的序列(索引号AF332769)比对后确定为中华哲水蚤mtCOI序列的一部分.利用本实验方法所提取的基因组DNA可适用于系统发生、种群遗传结构等领域的分子水平研究,为保存在甲醛溶液中的小型海洋浮游动物的基因信息利用提供实用技术.The DNA extraction,in General,from samples stored in formaldehyde solution is inefficient,especially extracted from small individual.It is necessary extracting of DNA genome from formaldehyde samples which conserved in early days or fixed in formaldehyde solution which usually adopted in marine investigation. On the base of methods of DNA extraction used in other animals or tissues,genomic DNA is extracted from Calanus sinicus stored in 5% formaldehyde solution in this experiment.More attention was paid to the process of eliminating the formaldehyde solution at the very beginning and at the digestion period,we add DTT into the digestion reaction systemwhich are the most important steps in the whole experiment.The concentration of genomic DNA in the study is low relatively,but after efficient PCR amplification,mtDNA COI gene fragment (710 bp) was obtained.Then comparing with the sequence of Calanus sinicus recorded in Genbank (accession no.AF332769),it is sure of that what we get covers the recorded of GenBank,is Calanus sinicus mtDNA COI sequence.The study will serve other small pelagic crustacean for the extracting DNA genome.It can also apply to the research of phylogeny,the population genetics and so on at molecular level,providing the use of gene information of small marine zooplankton samples stored in formaldehyde solution with practical technique.国家重点基础研究发展规划 ( 973 )项目资助(G1999043708

    GPRS-based intelligent monitoring system of public lighting

    设计了公共照明远程智能监控系统。以单片机为主控制器,实现对现场终端的运行状态、电压/电流等开关量和模拟量的检测、控制与报警。通过无线通信gPrS与监控中心建立无线网络连接,接收监控中心的指令,同时将检测到的数据和报警信息上传至监控中心。监控终端包括强电主回路、开关电源、人机接口、通信模块、数据采集单元和开关量控制等部分。系统软件监控部分采用可视开发工具VISuAl 2005.nET开发,服务器采用WIndOWS 2000 SErVEr操作系统,数据库选用2000 SQl平台。软件系统包括:主控、数据服务、网络服务和地图服务等子系统。所设计的公共照明智能化监控系统能满足系统分散化、网络化及智能化的要求。An intelligent remote monitoring system of public lighting is designed,which takes singlechip as MCU to realize the detection,control and alarm of binary and analog variables,such as the operating condition,voltage and current of field terminals.The system receives command from monitoring center and uploads the detected data and alarm information to it via GPRS communication.The monitoring terminal consists of the high-voltage main circuit,switching power supply,man-machine interface,communication module,data acquisition unit and switching control unit.The software of monitoring module is developed with Visual 2005.net,Windows 2000 Server is employed as the operating system and 2000 SQL platform is used for the database management system.The software system includes the subsystems such as the master control,data service,network service,map service,etc.The designed system meets the requirements of decentralization,networkization and intelligentization
