Chemical profile and antioxidative activity of red wines obtained from autochthonous and international grape clone varieties
Ova tema je fokusirana na hemijsku i bioloÅ”ku karakterizaciju odabranih uzoraka crvenih vina. Akcenatje stavljen na hemijsku karakterizaciju klonskih sortnih vina vranac i merlo, kao i na procenu antioksidativneaktivnosti merlo vina u Äelijama kvasca. U radu su primenjene brojne analitiÄke tehnike zaodreÄivanje koncentracije fenolnih jedinjenja, elementalnog sastava i in vitro antioksidativnu aktivnostiispitivanih uzoraka (spektrofotometrija, cikliÄna voltametrija, klasiÄna polarografija, EPR spektroskopija,LC-MS/MS, ICP-OES i ICP-MS). Pored antioksidativne, ispitane su i citotoksiÄna i antibakterijska aktivnost odgovarajuÄihuzoraka vina. Eksperiment na bazi vinskog kvasca je obuhvatio odreÄivanje ukupnih āSHgrupa, kao i enzimskih aktivnosti glutation peroksidaze, glutation reduktaze i katalaze. Eksperimentalnodobijeni podaci za klonska sortna vina merlo ukazuju na poseban doprinos glutation peroksidaze preživljavanjuÄelija kvasca u uslovima oksidativnog stresa. KonaÄno, primenom veÅ”taÄke neuronske mreže razvijenaje brza i precizna metoda za predviÄanje uticaja malih promena u vinskom fenolnom profilu naaktivnosti istaknutih enzima antioksidativne zaÅ”tite.This topic focuses on the chemical and biological characterization of selected red wine samples. Emphasisis placed on the chemical characterisation of clonal wines of Vranac and Merlot varieties, as well ason the evaluation of the antioxidant activity of Merlot wines in yeast cells. Numerous analytical techniqueswere used to determine the concentration of phenolic compounds, elemental composition and invitro antioxidant activity of the tested samples (spectrophotometry, cyclic voltammetry, classical polarography,EPR spectroscopy, LC-MS/MS, ICP-OES and ICP-MS). In addition, total antioxidant activity, cytotoxicand antibacterial activity of the respective wine samples were determined. The wine yeast experimentinvolved determining the total groups āSH, as well as enzymatic activities of glutathione peroxidase, glutathionereductase and catalase. The experimentally obtained data for clonal Merlot wine varieties indicatea particular contribution of glutathione peroxidase to the survival of yeast cells under oxidative stress conditions.Finally, using an artificial neural network (ANN), a rapid and accurate method was developed to predictthe effect of small changes in the wine phenolic profile on the activities of prominent antioxidantenzymes
The effects of a selected methoxy substituted chalcone in human melanoma cells irradiated with Ī³-rays
Given the well-established potential of chalcones in modulating the response of cancer cells to therapeutic interventions, coupled with the growing imperative to enhance their biological attributes, the objective of this study was to synthesize a methoxy-substituted chalcone (OCH3) and assess its capacity to amplify the inhibitory effects of radiation in melanoma cells known for their resistance to radiotherapy. The A375 melanoma cells were subjected to a clinically relevant dose of 2 Gy gamma irradiation. OCH3 was employed either as a standalone treatment or in conjunction with irradiation. The obtained results unveiled the substantial radiosensitizing potential of OCH3 within this specific cell line. Our subsequent investigations will be designed to investigate the underlying mechanisms that contribute to the radiosensitizing properties of OCH3. Moreover, we intend to evaluate the efficacy of OCH3 against other types of radioresistant cancer cells. The presented data not only illuminates the enhanced therapeutic possibilities offered by OCH3 but also highlights its potential as a valuable agent in addressing a wider array of challenging malignancies.ICCBIKG 2023 : 2nd International Conference on Chemo and Bioinformatics, September 28-29, 2023; Kragujeva

In vitro Radioprotective Activity of the Bryozoan Hyalinella punctata
The objective of the present study was in vitro evaluation of radioprotective activity of the freshwater bryozoan Hyalinella punctata on cultured human peripheral blood lymphocytes after irradiation with 2 gy of Co-60 gamma-rays. Since its water extract at concentration 0.001 mg/mL reduced the incidence of radiation-induced micronuclei for almost 30 %, it could be considered as a promising source of new natural products with the aforementined activity. Both the content of sulphur (1.17%) determined by gravimetric method and infrared absorption frequences (76 % similarity with those of bacitracin) of the investigated extract indicate the presence of organic sulphur compound(s) such as cyclic peptides and polypeptides which might be responsible for the observed radioprotectio
Olanzapine intensifies lipid peroxidation and modulates catalase activity in liver of social isolated rats
Olanzapine is an antipsychotic proved to be effective in stress associated psychiatric diseases, but its effect on the liver, main site of drug metabolism,still remain unclear. We investigated the effects of chronic treatment of olanzapine (three-week) on the malondialdehyde (MDA) content and protein expression and activity of antioxidant enzyme catalase (CAT) in the liver of rats exposed to chronic social isolation (CSIS) (six-week), an animal model of depression. The increased cytosolic MDA content in both vehicle-and olanzapine-treated CSIS animals suggests oxidative stress. Increased CAT activityin vehicle-treated CSIS animals, which was not consistent with its protein expression, suggests induction of antioxidant defense mechanisms,while olanzapine significantly reduced CAT activity in CSIS group. Synthesis and cytotoxic activity of selected dual COX-2 and 5-LOX inhibitors in HeLa and MIA PaCa-2 human cancer cell lines
Among novel cancer chemotherapy approaches, the use of cyclooxygenases (COXs) and lipoxygenases (LOXs) inhibitors represents a promising mean for cancer treatment showing lesser toxicity comparing to the currently used cytotoxic drugs. This study detailed the synthesis of three novel compounds: 1ME, BHTK-AA, and IBU-Ac, each with the capability to concurrently inhibit both COX-2 and 5-LOX. Subsequently, we assessed their effectiveness in inhibiting the proliferation of HeLa cervical and MIA PaCa-2 pancreatic cancer cells. The IC50 values for both examined cell lines were approximately 40 Ī¼M, indicating the promising inhibitory potential of the IBU-Ac compound in both types of cancer cells. This finding is positioned to stimulate further investigation into the potential application of IBU-Ac against these particular types of cancers, while also advocating its use in combination with standard anti-cancer protocols, i.e., chemoterapeutics or radiation therapy.
