Computational localization microscopy with extended axial range
A new single-aperture 3D particle-localization and tracking technique is presented that demonstrates an increase in depth range by more than an order of magnitude without compromising optical resolution and throughput. We exploit the extended depth range and depth-dependent translation of an Airy-beam PSF for 3D localization over an extended volume in a single snapshot. The technique is applicable to all bright-field and fluorescence modalities for particle localization and tracking, ranging from super-resolution microscopy through to the tracking of fluorescent beads and endogenous particles within cells. We demonstrate and validate its application to real-time 3D velocity imaging of fluid flow in capillaries using fluorescent tracer beads. An axial localization precision of 50 nm was obtained over a depth range of 120μm using a 0.4NA, 20× microscope objective. We believe this to be the highest ratio of axial range-to-precision reported to date
Extended depth-of-field imaging and ranging in a snapshot
Traditional approaches to imaging require that an increase in depth of field is associated with a reduction in
numerical aperture, and hence with a reduction in resolution and optical throughput. In their seminal
work, Dowski and Cathey reported how the asymmetric point-spread function generated by a cubic-phase
aberration encodes the detected image such that digital recovery can yield images with an extended depth of
field without sacrificing resolution [Appl. Opt. 34, 1859 (1995)]. Unfortunately recovered images are
generally visibly degraded by artifacts arising from subtle variations in point-spread functions with defocus.
We report a technique that involves determination of the spatially variant translation of image components
that accompanies defocus to enable determination of spatially variant defocus. This in turn enables recovery
of artifact-free, extended depth-of-field images together with a two-dimensional defocus and range map
of the imaged scene. We demonstrate the technique for high-quality macroscopic and microscopic imaging
of scenes presenting an extended defocus of up to two waves, and for generation of defocus maps with an
uncertainty of 0.036 waves
Low-Cost Compressive Sensing for Color Video and Depth
A simple and inexpensive (low-power and low-bandwidth) modification is made
to a conventional off-the-shelf color video camera, from which we recover
{multiple} color frames for each of the original measured frames, and each of
the recovered frames can be focused at a different depth. The recovery of
multiple frames for each measured frame is made possible via high-speed coding,
manifested via translation of a single coded aperture; the inexpensive
translation is constituted by mounting the binary code on a piezoelectric
device. To simultaneously recover depth information, a {liquid} lens is
modulated at high speed, via a variable voltage. Consequently, during the
aforementioned coding process, the liquid lens allows the camera to sweep the
focus through multiple depths. In addition to designing and implementing the
camera, fast recovery is achieved by an anytime algorithm exploiting the
group-sparsity of wavelet/DCT coefficients.Comment: 8 pages, CVPR 201
Non-convex optimization for 3D point source localization using a rotating point spread function
We consider the high-resolution imaging problem of 3D point source image
recovery from 2D data using a method based on point spread function (PSF)
engineering. The method involves a new technique, recently proposed by
S.~Prasad, based on the use of a rotating PSF with a single lobe to obtain
depth from defocus. The amount of rotation of the PSF encodes the depth
position of the point source. Applications include high-resolution single
molecule localization microscopy as well as the problem addressed in this paper
on localization of space debris using a space-based telescope. The localization
problem is discretized on a cubical lattice where the coordinates of nonzero
entries represent the 3D locations and the values of these entries the fluxes
of the point sources. Finding the locations and fluxes of the point sources is
a large-scale sparse 3D inverse problem. A new nonconvex regularization method
with a data-fitting term based on Kullback-Leibler (KL) divergence is proposed
for 3D localization for the Poisson noise model. In addition, we propose a new
scheme of estimation of the source fluxes from the KL data-fitting term.
Numerical experiments illustrate the efficiency and stability of the algorithms
that are trained on a random subset of image data before being applied to other
images. Our 3D localization algorithms can be readily applied to other kinds of
depth-encoding PSFs as well.Comment: 28 page
Reinterpretable Imager: Towards Variable Post-Capture Space, Angle and Time Resolution in Photography
We describe a novel multiplexing approach to achieve tradeoffs in space, angle and time resolution in photography. We explore the problem of mapping useful subsets of time-varying 4D lightfields in a single snapshot. Our design is based on using a dynamic mask in the aperture and a static mask close to the sensor. The key idea is to exploit scene-specific redundancy along spatial, angular and temporal dimensions and to provide a programmable or variable resolution tradeoff among these dimensions. This allows a user to reinterpret the single captured photo as either a high spatial resolution image, a refocusable image stack or a video for different parts of the scene in post-processing.
A lightfield camera or a video camera forces a-priori choice in space-angle-time resolution. We demonstrate a single prototype which provides flexible post-capture abilities not possible using either a single-shot lightfield camera or a multi-frame video camera. We show several novel results including digital refocusing on objects moving in depth and capturing multiple facial expressions in a single photo
High-throughput intensity diffraction tomography with a computational microscope
We demonstrate a motion-free intensity diffraction tomography technique that enables direct inversion of 3D phase and absorption from intensity-only measurements for weakly scattering samples. We derive a novel linear forward model, featuring slice-wise phase and absorption transfer functions using angled illumination. This new framework facilitates flexible and efficient data acquisition, enabling arbitrary sampling of the illumination angles. The reconstruction algorithm performs 3D synthetic aperture using a robust, computation and memory efficient slice-wise deconvolution to achieve resolution up to the incoherent limit. We demonstrate our technique with thick biological samples having both sparse 3D structures and dense cell clusters. We further investigate the limitation of our technique when imaging strongly scattering samples. Imaging performance and the influence of multiple scattering is evaluated using a 3D sample consisting of stacked phase and absorption resolution targets. This computational microscopy system is directly built on a standard commercial microscope with a simple LED array source add-on, and promises broad applications by leveraging the ubiquitous microscopy platforms with minimal hardware modifications
Fourier optics approaches to enhanced depth-of-field applications in millimetre-wave imaging and microscopy
In the first part of this thesis millimetre-wave interferometric imagers are considered
for short-range applications such as concealed weapons detection. Compared to real
aperture systems, synthetic aperture imagers at these wavelengths can provide improvements
in terms of size, cost, depth-of-field (DoF) and imaging flexibility via digitalrefocusing.
Mechanical scanning between the scene and the array is investigated to
reduce the number of antennas and correlators which drive the cost of such imagers.
The tradeoffs associated with this hardware reduction are assessed before to jointly
optimise the array configuration and scanning motion. To that end, a novel metric is
proposed to quantify the uniformity of the Fourier domain coverage of the array and is
maximised with a genetic algorithm. The resulting array demonstrates clear improvements
in imaging performances compared to a conventional power-law Y-shaped array.
The DoF of antenna arrays, analysed via the Strehl ratio, is shown to be limited even
for infinitely small antennas, with the exception of circular arrays.
In the second part of this thesis increased DoF in optical systems with Wavefront
Coding (WC) is studied. Images obtained with WC are shown to exhibit artifacts
that limit the benefits of this technique. An image restoration procedure employing a
metric of defocus is proposed to remove these artifacts and therefore extend the DoF
beyond the limit of conventional WC systems. A transmission optical microscope was
designed and implemented to operate with WC. After suppression of partial coherence
effects, the proposed image restoration method was successfully applied and extended
DoF images are presented
