Phosphoenolpyruvate Carboxykinase Assayed at Physiological Concentrations of Metal Ions Has a High Affinity for CO2
The effect of Mn2+/Mg2+ concentration on the activity of intact, homogeneous phosphoenolpyruvate carboxykinase (PEPCK) from leaves of the C4 grass, Guinea grass (Panicum maximum), have been investigated. Assay conditions were optimized so that PEPCK activity could be measured at concentrations of Mn2+/Mg2+ similar to those found in the cytosol (low micromolar Mn2+ and millimolar Mg2+). PEPCK activity was totally dependent on Mn2+ and was activated at low micromolar concentrations of Mn2+ by millimolar concentrations of Mg2+. Therefore, at physiological concentrations of Mn2+, PEPCK has a requirement for Mg2+. Assay at physiological concentrations of Mn2+/Mg2+ led to a marked decrease in its affinity for ATP and a 13-fold increase in its affinity for CO2. The Km (CO2) was further decreased by assay at physiological ATP to ADP ratios, reaching values as low as 20 μM CO2, comparable with the Km (CO2) of ribulose 1,5-bisphosphate carboxylase-oxygenase. This means that PEPCK will catalyze a reversible reaction and that it could operate as a carboxylase in vivo, a feature that could be particularly important in algal CO2-concentrating systems

Responses of different panicum miliaceum l. Genotypes to saline and water stress in a marginal mediterranean environment
The aims of this study were to evaluate: (1) the effect of sodium chloride (NaCl) and mannitol at different osmotic pressures on the germination of three proso millet (Panicum miliaceum L.) genotypes (VIR 9181, Unikum, and Kinelskoje) under controlled laboratory conditions; and (2) the effects of irrigation water salinity, maximum crop evapotranspiration (ETm) restitution regimes, and arbuscular mycorrhizal fungi (AMF) inoculation on forage production in a marginal Mediterranean soil for the genotypes that showed the highest and lowest seed germination. In the laboratory experiment, the Unikum genotype showed the highest seed germination (95.1%), whereas the lowest was found for Kinelskoje (80.4%). Regardless of the osmoticum type, germination was significantly reduced by osmotic pressure increases. Unikum showed a higher fresh biomass yield (FBY) (620.4 ± 126.3 g m−2) than Kinelskoje (340.0 ± 73.5 g m−2). AMF inoculation did not influence FBY under salt conditions, while in the absence of salt conditions it significantly increased the Unikum FBY (+50.7%) as compared to the uninoculated treatment (552.5 ± 269 g m−2). The 25% ETm significantly reduced FBY in both genotypes (−86.2% and −84.1% for Unikum and Kinelskoje, respectively) sd compared to the 100% ETm treatments (1090.3 ± 49.7 g m−2 in Unikum and 587 ± 72.2 g m−2 in Kinelskoje). The obtained results give novel information about proso millet forage production in low-input agriculture in marginal semi-arid Mediterranean land
Macro-Climatic Distribution Limits Show Both Niche Expansion and Niche Specialization among C4 Panicoids
Grasses are ancestrally tropical understory species whose current dominance in warm open habitats is linked to the evolution of C4 photosynthesis. C4 grasses maintain high rates of photosynthesis in warm and water stressed environments, and the syndrome is considered to induce niche shifts into these habitats while adaptation to cold ones may be compromised. Global biogeographic analyses of C4 grasses have, however, concentrated on diversity patterns, while paying little attention to distributional limits. Using phylogenetic contrast analyses, we compared macro-climatic distribution limits among ~1300 grasses from the subfamily Panicoideae, which includes 4/5 of the known photosynthetic transitions in grasses. We explored whether evolution of C4 photosynthesis correlates with niche expansions, niche changes, or stasis at subfamily level and within the two tribes Paniceae and Paspaleae. We compared the climatic extremes of growing season temperatures, aridity, and mean temperatures of the coldest months. We found support for all the known biogeographic distribution patterns of C4 species, these patterns were, however, formed both by niche expansion and niche changes. The only ubiquitous response to a change in the photosynthetic pathway within Panicoideae was a niche expansion of the C4 species into regions with higher growing season temperatures, but without a withdrawal from the inherited climate niche. Other patterns varied among the tribes, as macro-climatic niche evolution in the American tribe Paspaleae differed from the pattern supported in the globally distributed tribe Paniceae and at family level.Fil: Aagesen, Lone. Note sur les modalités techniques d'installation des pâturages de Panicum
L'auteur met en évidence les qualités fourragères de l'espèce Panicum maximum. Quatre variétés proposées en vulgarisation en Côte d'Ivoire sont caractérisées. Les modalités de l'établissement du pâturage sont précisées au niveau de l'installation par semis ainsi que par boutures. Les résultats obtenus en matière d'utilisation du pâturage par le bétail sont également présentés ainsi que les normes observées sur la pérennité. Une attention particulière doit être portée sur les rythmes d'exploitation et la nécessité d'apporter une fertilisation annuelle d'entretien et de production. Avec l'espèce Panicum maximum et la mise au point d'une technique pour récolter des semences de qualité, l'on dispose d'un matériel végétal très performant pour améliorer et intensifier l'élevage en régions tropicales humide
