    Pentachlorophenol and spent engine oil degradation by Mucor ramosissimus

    Pentachlorophenol (PCP) has been widely used for many years and belongs to the most toxic pollutants. Spent engine oils enter environment every day in many ways. Both of them cause great environmental concern. In the present work we focused on identifying metabolites of PCP biodegradation formed in the cultures of Mucor ramosissimus IM 6203 and optimizing medium composition to enhance PCP removal in the presence of engine oil acting as a carbon source. Pentachlorophenol (PCP) to tetrachlorohydroquinone (TCHQ) transformation was the most interesting transformation conducted by the tested strain. TCHQ was further transformed to 2,3,5,6-TCP and 2,3,4,6- TCP. Strain IM 6203 is also capable of PCP transformation to corresponding anisoles – pentachloromethoxybenzene (PCMB) and pentachloroethoxybenzene (PCEB). Characterization of enzymatic background involved in PCP to TCHQ transformation showed that TCHQ formation is catalyzed by inductive and cytochrome P-450 dependent enzymatic system. Experiments conducted on mineral medium allowed defining the optimal quantitative and qualitative medium make-up for PCP to TCHQ transformation. Biodegradation of PCP on the optimized synthetic medium X was more efficient than on rich Sabouraud medium. The tested strain is capable of growing in the presence of spent engine oil therefore we checked the ability of PCP transformation on optimized synthetic medium containing oil as a carbon source. The obtained results showed that PCP removal and TCHQ formation occurred were found to be the most efficient on the oil containing medium (OX medium). PCP removal and TCHQ formation after 240 h of culturing reached 1.19 mg/l and 0.89 mg/l, respectively. Additionally, 55.5% of oil introduced to the medium was removed during 10 days of the experiment. PCP biodegradation mechanisms used by Mucor species have not been sufficiently explained. The presented results point to the tested strain as an interesting model for the research on fungal PCP biodegradation in the areas highly contaminated with engine oil and for its future application in PCP and oils removal

    Women’s use of Preventive Primary Care in the Late Postpartum Period

    The literature has limited data on how women access health care after the traditional postpartum period (postpartum). Modeled after a paper by Bryant (2016), this project assesses the prevalence of primary care visits in the late postpartum period (LPP)(60- 730 days postpartum). Study objectives included (1) Identify demographics of general delivering population at UVMMC compared to patients with UVM-affiliated primary care provider (UVMPCP). (2) Understand how the general delivering population uses the UVMHN LPP (3) Among women with a UVM-affiliated PCP, identify the prevalence of preventive care visits in the LPP. (4) Identify characteristics associated with LPP visit attendance. Hypothesis: Women with an established PCP prior to pregnancy are more likely to attend preventive PCP LPP visits. This was a retrospective cohort study for all women who delivered at UVMMC between 7/1/2015-6/30/2017. Data was extracted from Epic EMR. During the study period, 4169 women had one singleton pregnancy, 3413 (82%) had a known PCP, and 1279 (31%) had UVMPCP. 2535 (61%) of all delivering singleton women and 1112 (87%) of UVMPCP women had at least one clinical visit within UVMHN in the LPP. 959 (75%) of UVMPCP women had a LPP PCP visit, and 382 patients (30%) had preventative PCP LPP visits. Our hypothesis was rejected (OR 0.930), but attending any LPP PCP visit was associated with having a PCP established prior to pregnancy (OR 1.684). Attending preventive PCP visit was associated with having the same delivering provider as PCP (OR 1.742), a pre-pregnancy PCP visit (OR 1.460), a PCP visit during prenatal time (OR 1.459), ED visit early postpartum period (OR 0.402), a fetal or neonatal demise (OR 0.445), being single (0.601), and with public insurance (OR 0.489). Further work in understanding these associations will be important in developing improved transition of care models and increasing overall engagement in women’s preventive medicine

    Dishevelled genes mediate a conserved mammalian PCP pathway to regulate convergent extension during neurulation

    The planar cell polarity (PCP) pathway is conserved throughout evolution, but it mediates distinct developmental processes. In Drosophila, members of the PCP pathway localize in a polarized fashion to specify the cellular polarity within the plane of the epithelium, perpendicular to the apicobasal axis of the cell. In Xenopus and zebrafish, several homologs of the components of the fly PCP pathway control convergent extension. We have shown previously that mammalian PCP homologs regulate both cell polarity and polarized extension in the cochlea in the mouse. Here we show, using mice with null mutations in two mammalian Dishevelled homologs, Dvl1 and Dvl2, that during neurulation a homologous mammalian PCP pathway regulates concomitant lengthening and narrowing of the neural plate, a morphogenetic process defined as convergent extension. Dvl2 genetically interacts with Loop-tail, a point mutation in the mammalian PCP gene Vangl2, during neurulation. By generating Dvl2 BAC (bacterial artificial chromosome) transgenes and introducing different domain deletions and a point mutation identical to the dsh1 allele in fly, we further demonstrated a high degree of conservation between Dvl function in mammalian convergent extension and the PCP pathway in fly. In the neuroepithelium of neurulating embryos, Dvl2 shows DEP domain-dependent membrane localization, a pre-requisite for its involvement in convergent extension. Intriguing, the Loop-tail mutation that disrupts both convergent extension in the neuroepithelium and PCP in the cochlea does not disrupt Dvl2 membrane distribution in the neuroepithelium, in contrast to its drastic effect on Dvl2 localization in the cochlea. These results are discussed in light of recent models on PCP and convergent extension

    Interaction of the phencyclidine model of schizophrenia and nicotine on total and categorized ultrasonic vocalizations in rats

    Patients with schizophrenia smoke cigarettes at a higher rate than the general population. We hypothesized that a factor in this comorbidity is sensitivity to the reinforcing and reinforcement- enhancement effects of nicotine. Phencyclidine (PCP) was used to model behavioral changes resembling negative symptoms of schizophrenia in rats. USVs in rats have been used to measure emotional states, with 50 kHz USVs indicating positive states and 22 kHz indicating negative. Total and categorized numbers of 22 and 50 kHz ultrasonic vocalizations (USVs) and USVs during a visual stimulus (e.g. a potential measure of reinforcement-enhancement) were examined in rats following .injection ofh PCP (2.0 mg/kg), and/or nicotine (0.2 or 0.4 mg/kg) daily for 7 days. PCP was then discontinued and all rats received nicotine (0.2 mg/kg and 0.4 mg/kg) and PCP (2.0 mg/kg) on 3 challenge days. PCP acutely decreased 50 kHz vocalizations while repeated nicotine potentiated rates of vocalizations, with similar patterns during light presentations. Rats in the PCP and nicotine combination groups made more 50 kHz vocalizations compared to control groups on challenge days. We conclude that PCP may produce a reward deficit that is shown by decreased 50 kHz USVs, and behaviors post-PCP exposure may best model the comorbidity between schizophrenia and nicotine

    Prediction of Thermal Behavior of Pervious Concrete Pavements in Winter

    Because application of pervious concrete pavement (PCPs) has extended to cold-climate regions of the United States, the safety and mobility of PCP installations during the winter season need to be maintained. Timely application of salt, anti-icing, and deicing agents for ice/snow control is most effective in providing sufficient surface friction when done at a suitable pavement surface temperature. The aim of this project was to determine the thermal properties of PCP during the winter season, and to develop a theoretical model to predict PCP surface temperature. The project included a laboratory and a field component. In the laboratory, thermal conductivity of pervious concrete was determined. A linear relationship was established between thermal conductivity and porosity for pervious concrete specimens. In the field, the pavement temperature in a PCP sidewalk installation at Washington State University was monitored via in-pavement instrumentation. Based on the field data, the Enhanced Integrated Climatic Model (EICM) was developed and validated for the site, using PCP thermal properties and local climatic data. The EICM-predicted PCP surface temperature during the winter season agreed well with the field temperature. Overall, the predicted number of days that the pavement surface fell below 32°F agreed well with the number based on field data for 85% of the days. Therefore, the developed model is useful in identifying those days to apply deicer agents. Finally, a regression model using climatic indices was developed for PCP surface temperature prediction in the absence of a more advanced temperature model

    The release of contaminants into the environment is inevitable. Contaminants are released through manufacture and use of products and as a result of treatment and disposal of wastes. Upon release to the environment, contaminants move and respond to a number of interrelated natural and man made factors. Penta-chloro-phenol (PCP) is one such contaminant that has been released into the environment and is known to have serious long term environmental effects. The objective of this study was to determine the effectiveness of biological processes to remediate soil contaminated with Penta-chloro-phenol (PCP). This thesis reviews mechanisms by which soil is contaminated, processes available to remediate soils, and in particular, process requirements for successful bioremediation. The abilities of bacteria to degrade PCP from soil contaminated with PCP was evaluated. Solid phase and slurry phase experiments were examined for their effect on PCP concentration over a four month period at the Department of Technology. Massey University. The objectives of this study were (1) To determine if aeration and inoculation of soil in-situ could produce significant removal of PCP. (2) Determine the effect of concentration on bioremediation rates. (3) Compare in-situ treatment with bio-slurry treatments. The experiments showed that it is possible to remove up to 95% of PCP from contaminated soil by inoculation with bacteria. Inoculum size and aeration were shown to be critical factors in affecting the rate of degradation. The larger the initial inoculum the greater the rate of degradation. Without aeration the inoculum was unable to significantly degrade PCP. The bio-slurry confirmed that PCP could be removed readily from soil to an aqueous state. In an aqueous state PCP is degraded at a faster rate than when it is incorporated into the soil matrix. The results of this work is to show that soil rehabilitation by way of biodegradation is a feasible and attractive process

    Derandomized Parallel Repetition via Structured PCPs

    A PCP is a proof system for NP in which the proof can be checked by a probabilistic verifier. The verifier is only allowed to read a very small portion of the proof, and in return is allowed to err with some bounded probability. The probability that the verifier accepts a false proof is called the soundness error, and is an important parameter of a PCP system that one seeks to minimize. Constructing PCPs with sub-constant soundness error and, at the same time, a minimal number of queries into the proof (namely two) is especially important due to applications for inapproximability. In this work we construct such PCP verifiers, i.e., PCPs that make only two queries and have sub-constant soundness error. Our construction can be viewed as a combinatorial alternative to the "manifold vs. point" construction, which is the only construction in the literature for this parameter range. The "manifold vs. point" PCP is based on a low degree test, while our construction is based on a direct product test. We also extend our construction to yield a decodable PCP (dPCP) with the same parameters. By plugging in this dPCP into the scheme of Dinur and Harsha (FOCS 2009) one gets an alternative construction of the result of Moshkovitz and Raz (FOCS 2008), namely: a construction of two-query PCPs with small soundness error and small alphabet size. Our construction of a PCP is based on extending the derandomized direct product test of Impagliazzo, Kabanets and Wigderson (STOC 09) to a derandomized parallel repetition theorem. More accurately, our PCP construction is obtained in two steps. We first prove a derandomized parallel repetition theorem for specially structured PCPs. Then, we show that any PCP can be transformed into one that has the required structure, by embedding it on a de-Bruijn graph

    Pneumatization of the sphenoid sinus, dorsum sellae and posterior clinoid processes in computed tomography

    Purpose: Assessment of sphenoid sinus (SS), posterior clinoid process (PCP), and dorsum sellae (DS) pneumatisation is significant when planning surgical approaches both to intra- and extrasellar pathologies. The authors analysed pneumatisation of the SS, with particular attention paid to pneumatisation of the PCP and DS. Material and methods: The study was based on 100 computed tomography angiography examinations. SS, PCP, and DS pneumatisation grades were assessed using the classification system proposed by Hardy. The analyses were conducted in sagittal planes: midline plane (MP), sagittal posterior clinoid plane right (SPCP-R), and sagittal posterior clinoid plane left (SPCP-L). The occurrence of the highest pneumatisation grade (IVB according to Hardy), which encompasses DS and PCP, prompted the authors to conduct a further analysis in the coronal plane. In this way, seven DS and PCP pneumatisation symmetry types were identified. Results: In the MP and SPCP-R, the most frequent pneumatisation grade was grade III (41% and 38%, respectively). In the SPCP-L, grade IVA prevailed (41%). Grade IVB was found in 12% of the SPCP-R images, 10% of the SPCP-L images, and in 12% of the MP images. Consistent pneumatisation grades in all analysed planes were found for 64% of cases. This was usually grade III noted in 28% of cases. Conclusions: SS pneumatisation is characterised by considerable individual variability. There are a number of SS pneumatisation classification systems, but the system proposed by Hardy is the most useful for assessment of DS and PCP pneumatisation. Grade III of SS pneumatisation is the most common. Pneumatisation encompassing DS and PCP was found in 10-12% of cases
