SalmoNet, an integrated network of ten Salmonella enterica strains reveals common and distinct pathways to host adaptation
Salmonella enterica is a prominent bacterial pathogen with implications on human and animal health. Salmonella serovars could be classified as gastro-intestinal or extra-intestinal. Genome-wide comparisons revealed that extra-intestinal strains are closer relatives of gastro-intestinal strains than to each other indicating a parallel evolution of this trait. Given the complexity of the differences, a systems-level comparison could reveal key mechanisms enabling extra-intestinal serovars to cause systemic infections. Accordingly, in this work, we introduce a unique resource, SalmoNet, which combines manual curation, high-throughput data and computational predictions to provide an integrated network for Salmonella at the metabolic, transcriptional regulatory and protein-protein interaction levels. SalmoNet provides the networks separately for five gastro-intestinal and five extra-intestinal strains. As a multi-layered, multi-strain database containing experimental data, SalmoNet is the first dedicated network resource for Salmonella. It comprehensively contains interactions between proteins encoded in Salmonella pathogenicity islands, as well as regulatory mechanisms of metabolic processes with the option to zoom-in and analyze the interactions at specific loci in more detail. Application of SalmoNet is not limited to strain comparisons as it also provides a Salmonella resource for biochemical network modeling, host-pathogen interaction studies, drug discovery, experimental validation of novel interactions, uncovering new pathological mechanisms from emergent properties and epidemiological studies. Gastrointestinal neuromuscular apparatus: An underestimated target of gut microbiota
Over the last few years, the importance of the resident
intestinal microbiota in the pathogenesis of several gastro-
intestinal diseases has been largely investigated. Growing
evidence suggest that microbiota can influence gastro-
intestinal motility. The current working hypothesis is that
dysbiosis-driven mucosal alterations induce the production
of several inflammatory/immune mediators which affect
gut neuro-muscular functions. Besides these indirect
mucosal-mediated effects, the present review highlights
that recent evidence suggests that microbiota can directly
affect enteric nerves and smooth muscle cells functions
through its metabolic products or bacterial molecular
components translocated from the intestinal lumen. Toll-
like receptors, the bacterial recognition receptors, are
expressed both on enteric nerves and smooth muscle and
are emerging as potential mediators between microbiota
and the enteric neuromuscular apparatus. Furthermore,
the ongoing studies on probiotics support the hypothesis
that the neuromuscular apparatus may represent a target
of intervention, thus opening new physiopathological and
therapeutic scenarios
Cereal non-starch polysaccharides in pig diets
This thesis is based on three different studies comprising the weaning and the growing period, aiming at monitoring the influence of cereal non-starch polysaccharides (NSP) and dietary enzyme supplementation on gastro-intestinal processes in pigs. The diets were based on cereals and cereal by-products, and composed to contain different amounts of total as well as soluble NSP. Results from these studies have shown that with increased NSP content, the reduction in digestibility of organic matter (OM) was twice as high in the small intestine than in the total tract, both in newly weaned piglets and growing pigs. An increased proportion of insoluble NSP decreased the digestibility of OM and fibre components in the small intestine of the newly weaned piglets and in the total tract of growing pigs. The gut environment, as described by content and proportions of organic acids (OA) and pH, as well as total microbial populations and coliform diversity, was altered by NSP content and solubility, whereas enzyme supplementation influenced the distribution of OA in the small intestine. PVTC-cannulation did not influence the coliform flora, and results obtained from PVTC-cannulated pigs were concluded to reflect true intestinal conditions. In conclusion, these results indicate that the dietary content of total and soluble NSP influence gastro-intestinal processes such as digestion site, gut environment and microbial populations in different ways in newly weaned piglets and in growing pigs. Therefore, NSP constitute an important tool with possibilities to influence gut health in pigs, and may therefore offer prospects to optimise the feed for pigs of different age
The gastro-intestinal tract as the major site of biological action of dietary melanoidins
Emerging evidence from laboratory researches has highlighted the bioactivity of food melanoidins and melanoproteins. Whilst such studies have been carried out with different in vitro systems, information about melanoidins absorption and bio-availability are scarce. However, they are generally considered as poorly absorbable and bio-available compounds. Therefore, we present a review in which the gastro-intestinal tract is hypothesized to be the main site of action of food melanoidins and melanoproteins biological activity. We described recent data supporting this hypothesis both in vitro model systems and in vivo. Importantly, we focused this review only on the effect of melanoidins and melanoproteins extracted from food. Most of the studies had been carried out using water-soluble carbohydrate-based melanoidins isolated from different food sources (beer, barley coffee, coffee). In bakery products, melanoidins are protein-based structure (melanoproteins) which are largely insoluble in water. Dietary melanoidins and melanoproteins have been demonstrated to exert in vitro antioxidant and metal chelating ability in the gastro-intestinal tract reducing the formation of lipid hydroperoxides and advanced lipid oxidation end products during the digestion of meat. The reduction in the formation of these pro-atherogenic compounds has been shown to be followed by a decrease in their absorption in human volunteers. Food melanoidins have also shown in vitro anti-caries and prebiotic activities. We conclude by underlining the possible role of food melanoidins in the prevention of gastro-intestinal tract cancers. We hope this review will stimulate further research on food melanoidins and their biological activities in the gastro-intestinal tract
Helminth parasites of sheep and goats in Eastern Ethiopia
A two-year epidemiology study of helminths of small ruminants involved the collection of viscera from 655 sheep and 632 goats from 4 abattoirs in eastern Ethiopia. A further more detailed epidemiology study of gastro-intestinal nematode infections used the Haramaya University (HU) flock of 60 Black Head Ogaden sheep. The parasitological data included numbers of nematode eggs per gram of faeces (EPG), faecal culture L3 larvae, packed red cell volume (PCV), adult worm and early L4 counts, and FAMACHA eye-colour score estimates, along with animal performance (body weight change). There were 13 species of nematodes and 4 species of flukes present in the sheep and goats, with Haemonchus contortus being the most prevalent (65–80%), followed by Trichostrongylus spp. The nematode infection levels of both sheep and goats followed the bi-modal annual rainfall pattern, with the highest worm burdens occurring during the two rain seasons (peaks in May and September). There were significant differences in worm burdens between the 4 geographic locations for both sheep and goats. Similar seasonal but not geographical variations occurred in the prevalence of flukes. There were significant correlations between EPG and PCV, EPG and FAMACHA scores, and PCV and FAMACHA scores. Moreover, H. contortus showed an increased propensity for arrested development during the dry seasons. Faecal egg count reduction tests (FECRT) conducted on the HU flocks, and flocks in surrounding small-holder communities, evaluated the efficacy of commonly used anthelmintics, including albendazole (ABZ), tetramisole (TET), a combination (ABZ + TET) and ivermectin (IVM). Initially, high levels of resistance to all of the anthelmintics were found in the HU goat flock but not in the sheep. In an attempt to restore the anthelmintic efficacy a new management system was applied to the HU goat flock, including: eliminating the existing parasite infections in the goats, exclusion from the traditional goat pastures, and initiation of communal grazing of the goats with the HU sheep and animals of the local small-holder farmers. Subsequent FECRTs revealed high levels of efficacy of all three drugs in the goat and sheep flocks, demonstrating that anthelmintic efficacy can be restored by exploiting refugia. Individual FECRTs were also conducted on 8 sheep and goat flocks owned by neighbouring small-holder farmers, who received breeding stock from the HU. In each FECRT, 50 local breed sheep and goats, 6–9 months old, were divided into 5 treatment groups: ABZ, TET, ABZ + TET, IVM and untreated control. There was no evidence of anthelmintic resistance in the nematodes, indicating that dilution of resistant parasites, which are likely to be imported with introduced breeding goats, and the low selection pressure imposed by the small-holder farmers, had prevented anthelmintic resistance from emerging
Prevalence of Helicobacter pylori in patients with gastro-oesophageal reflux disease : systematic review.
Objectives: To ascertain the prevalence of Helicobacter pylori in patients with gastro-oesophageal reflux disease and its association with the disease.
Design: Systematic review of studies reporting the prevalence of H pylori in patients with and without gastro-oesophageal reflux disease.
Data sources: Four electronic databases, searched to November 2001, experts, pharmaceutical companies, and journals.
Main outcome measure: Odds ratio for prevalence of H pylori in patients with gastro-oesophageal reflux disease.
Results: 20 studies were included. The pooled estimate of the odds ratio for prevalence of H pylori was 0.60 (95% confidence interval 0.47 to 0.78), indicating a lower prevalence in patients with gastro-oesophageal reflux disease. Substantial heterogeneity was observed between studies. Location seemed to be an important factor, with a much lower prevalence of H pylori in patients with gastro-oesophageal reflux disease in studies from the Far East, despite a higher overall prevalence of infection than western Europe and North America. Year of study was not a source of heterogeneity.
Conclusion: The prevalence of H pylori infection was significantly lower in patients with than without gastro-oesophageal reflux, with geographical location being a strong contributor to the heterogeneity between studies. Patients from the Far East with reflux disease had a lower prevalence of H pylori infection than patients from western Europe and North America, despite a higher prevalence in the general population
The effect of fructose and maltodextrin vs glucose and maltodextrin formulated sports beverages on mountain-bike race performance : a thesis presented in partial fulfilment of the requirements for the degree of Master of Science in Sport and Exercise Science at Massey University, Wellington, New Zealand
Background: Exogenous carbohydrate improves performance during prolonged high-intensity exercise. When ingested together, fructose and glucose polymers are oxidised at rates 1.5-1.7 higher than isocaloric glucose solutions. As fructose and glucose are transported across the intestine via different mechanisms, the capacity for exogenous-carbohydrate absorption is greater with composite carbohydrate mixtures. Therefore, since the effect of ingesting multi-transportable carbohydrate on field-based performance has to our knowledge not been investigated, we examined their effect on mountain bike race performance. Finishing time was expected to be substantially reduced when multi-transportable carbohydrates were ingested. Method: Ten; male (7) and female (3), mountain bikers aged 32.9 ± 8.7 years, weighing 68.8 ± 9.4 kg and training for at least 8 hours per week and racing regularly participated in a double-blind crossover study. Following a standardised training and diet regimen cyclists completed two Olympic-distance (target winning time of 2h 15m), cross-country mountain bike races during which they ingested either a 11.25% maltodextrin and fructose solution (MF) or an isocaloric, equi-volumetric, isosmotic control solution containing maltodextrin and glucose (MG). Performance times, ratings of perceived exertion, gastrointestinal discomfort and measurements of hydration status were recorded and compared. Data was analysed using appropriate mixed models in SAS. Results: Cyclists were 1.8% (2mins 31s) faster in MF compared to MG (90% confidence interval:±1.8%; 72% likelihood of a substantial benefit) The effect solution composition on the increase in time from the first the final lap (fatigue) was 9.7% (±2.8%) in MF and 10.7% (±2.8%) in MG; which corresponded to a 0.9% reduction (±3.5%; unclear) in the fatigue in MF. Abdominal cramps were reduced by 8.1% in MF relative to MG (±6.6%; likely benefit) and for every 1% change in abdominal cramp rating, lap time increased by 0.14% (±0.10%). There were no clear effects of MF on ratings of perceived exertion and hydration status compared with MG. Conclusion: Cross-country mountain bike race performance was substantially enhanced following ingestion of a maltodextrin and fructose solution. This outcome was related to reduced gastro-intestinal distress supporting the theory that solutions containing multiple-transportable carbohydrates increase the availability of carbohydrate for metabolism. Further investigation with a larger sample size is recommended to establish whether the performance effect is genuinely beneficial or trivial
Duodenal carcinoid tumour – a case report
Duodenal carcinoids are rare tumours of the small intestine with heterogenous clinical and pathological characteristics. The long-term prognosis is very good if discovered in the early stages. We present the case of a patient with a non-functional duodenal carcinoid tumour discovered incidentally during an upper gastrointestinal endoscopy. The diagnosis was confirmed through immunohistochemistry. Treatment consisted of the endoscopic resection of the tumour and the surveillance of the patient for the following 2 years, with no signs of recurrence. We have conducted a literature review regarding the clinical manifestations, diagnosis, treatment, and follow-up of patients with this type of tumours
