    Cell-free Embryonic Stem Cell Extract-mediated Derivation of Multi-potent Stem Cells from NIH3T3 Fibroblasts for Functional and Anatomical Ischemic Tissue Repair

    The oocyte-independent generation of multipotent stem cells is one of the goals in regenerative medicine. We report that upon exposure to mouse ES cell (ESC) extracts, reversibly permeabilized NIH3T3 cells undergo de-differentiation followed by stimulus-induced re-differentiation into multiple lineage cell types. Genome-wide expression profiling revealed significant differences between NIH3T3 and ESC-extract treated NIH3T3 cells including re-activation of ESC specific transcripts. Epigenetically, ESC extracts induced CpG de-methylation of Oct4 promoter, hyper-acetylation of histones 3 and 4 and decreased lysine 9 (K-9) dimethylation of histone 3. In mouse models of surgically-induced hind limb ischemia (HLI) or acute myocardial infarction (AMI) transplantation of reprogrammed NIH3T3 cells significantly improved post-injury physiological functions and showed antomical evidence of engraftment and trans-differentiation into skeletal muscle, endothelial cell and cardiomyocytes. These data provide evidence for the generation of functional multi-potent stem like cells from terminally differentiated somatic cells without the introduction of trans-genes or ESC fusion

    Revisiting the Lyman Continuum Escape Crisis: Predictions for z > 6 from Local Galaxies

    The intrinsic escape fraction of ionizing Lyman continuum photons (fescf_{esc}) is crucial to understand whether galaxies are capable of reionizing the neutral hydrogen in the early universe at z>6. Unfortunately, it is not possible to access fescf_{esc} at z>4 with direct observations and the handful of measurements from low redshift galaxies consistently find fescf_{esc} < 10%, while at least fescf_{esc} ~ 10% is necessary for galaxies dominate reionization. Here, we present the first empirical prediction of fescf_{esc} at z>6 by combining the (sparsely populated) relation between [OIII]/[OII] and fescf_{esc} with the redshift evolution of [OIII]/[OII] as predicted from local high-z analogs selected by their Hα\alpha equivalent-width. We find fescf_{esc} = 5.73.3+8.35.7_{-3.3}^{+8.3}% at z=6 and fescf_{esc} = 10.46.3+15.510.4_{-6.3}^{+15.5}% at z=9 for galaxies with log(M/Msun_{sun}) ~ 9.0 (errors given as 1σ\sigma). However, there is a negative correlation with stellar mass and we find up to 50% larger fescf_{esc} per 0.5 dex decrease in stellar mass. The population averaged escape fraction increases according to fescf_{esc} = fesc,0((1+z)/3)af_{esc,0} ((1+z)/3)^a, with fesc,0=2.3±0.05f_{esc,0} = 2.3 \pm 0.05% and a=1.17±0.02a=1.17 \pm 0.02 at z > 2 for log(M/Msun_{sun}) ~ 9.0. With our empirical prediction of fescf_{esc} (thus fixing an important previously unknown variable) and further reasonable assumption on clumping factor and the production efficiency of Lyman continuum photons, we conclude that the average population of galaxies is just capable to reionize the universe by z ~ 6.Comment: 10 pages, 4 figure, 1 table.     Constraining the Lyα escape fraction with far-infrared observations of Lyα emitters

    We study the far-infrared properties of 498 Lyα emitters (LAEs) at z = 2.8, 3.1, and 4.5 in the Extended Chandra Deep Field-South, using 250, 350, and 500μm data from the Herschel Multi-tiered Extragalactic Survey and 870μm data from the LABOCA ECDFS Submillimeter Survey. None of the 126, 280, or 92 LAEs at z = 2.8, 3.1, and 4.5, respectively, are individually detected in the far-infrared data. We use stacking to probe the average emission to deeper flux limits, reaching 1σ depths of ∼0.1 to 0.4 mJy. The LAEs are also undetected at ?3σ in the stacks, although a 2.5σ signal is observed at 870μm for the z = 2.8 sources. We consider a wide range of far-infrared spectral energy distributions (SEDs), including an M82 and an Sd galaxy template, to determine upper limits on the far-infrared luminosities and far-infrared-derived star formation rates of the LAEs. These star formation rates are then combined with those inferred from the Lyα and UV emission to determine lower limits on the LAEs’ Lyα escape fraction (f esc (Lyα)). For the Sd SED template, the inferred LAEs f esc (Lyα) are ?30% (1σ) at z = 2.8, 3.1, and 4.5, which are all significantly higher than the global f esc (Lyα) at these redshifts. Thus, if the LAEs f esc (Lyα) follows the global evolution, then they have warmer far-infrared SEDs than the Sd galaxy template. The average and M82 SEDs produce lower limits on the LAE f esc (Lyα) of ∼10%–20% (1σ), all of which are slightly higher than the global evolution of f esc (Lyα), but consistent with it at the 2σ–3σ level

    In vivo therapeutic efficacy of frog skin-derived peptides against Pseudomonas aeruginosa-induced pulmonary infection

    Pseudomonas aeruginosa is an opportunistic and frequently drug-resistant pulmonary pathogen especially in cystic fibrosis sufferers. Recently, the frog skin-derived antimicrobial peptide (AMP) Esc(1-21) and its diastereomer Esc(1-21)-1c were found to possess potent in vitro antipseudomonal activity. Here, they were first shown to preserve the barrier integrity of airway epithelial cells better than the human AMP LL-37. Furthermore, Esc(1-21)-1c was more efficacious than Esc(1-21) and LL-37 in protecting host from pulmonary bacterial infection after a single intra-tracheal instillation at a very low dosage of 0.1 mg/kg. The protection was evidenced by 2-log reduction of lung bacterial burden and was accompanied by less leukocytes recruitment and attenuated inflammatory response. In addition, the diastereomer was more efficient in reducing the systemic dissemination of bacterial cells. Importantly, in contrast to what reported for other AMPs, the peptide was administered at 2 hours after bacterial challenge to better reflect the real life infectious conditions. To the best of our knowledge, this is also the first study investigating the effect of AMPs on airway-epithelia associated genes upon administration to infected lungs. Overall, our data highly support advanced preclinical studies for the development of Esc(1-21)-1c as an efficacious therapeutic alternative against pulmonary P. aeruginosa infection

    Extracellular Matrix Aggregates from Differentiating Embryoid Bodies as a Scaffold to Support ESC Proliferation and Differentiation

    Embryonic stem cells (ESCs) have emerged as potential cell sources for tissue engineering and regeneration owing to its virtually unlimited replicative capacity and the potential to differentiate into a variety of cell types. Current differentiation strategies primarily involve various growth factor/inducer/repressor concoctions with less emphasis on the substrate. Developing biomaterials to promote stem cell proliferation and differentiation could aid in the realization of this goal. Extracellular matrix (ECM) components are important physiological regulators, and can provide cues to direct ESC expansion and differentiation. ECM undergoes constant remodeling with surrounding cells to accommodate specific developmental event. In this study, using ESC derived aggregates called embryoid bodies (EB) as a model, we characterized the biological nature of ECM in EB after exposure to different treatments: spontaneously differentiated and retinoic acid treated (denoted as SPT and RA, respectively). Next, we extracted this treatment-specific ECM by detergent decellularization methods (Triton X-100, DOC and SDS are compared). The resulting EB ECM scaffolds were seeded with undifferentiated ESCs using a novel cell seeding strategy, and the behavior of ESCs was studied. Our results showed that the optimized protocol efficiently removes cells while retaining crucial ECM and biochemical components. Decellularized ECM from SPT EB gave rise to a more favorable microenvironment for promoting ESC attachment, proliferation, and early differentiation, compared to native EB and decellularized ECM from RA EB. These findings suggest that various treatment conditions allow the formulation of unique ESC-ECM derived scaffolds to enhance ESC bioactivities, including proliferation and differentiation for tissue regeneration applications. © 2013 Goh et al

    El uso de estrategias de comunicación de aprendices de inglés como L2 con diferentes niveles de competencia en un contexto oral interactivo

    Indexación: Web of Science; Scielo.Abstract: This study aims to examine the different communication strategies (CSs) EFL learners employ when communicating orally, and determine the relationship between the learners’ proficiency level and their CS use. Spoken data from three conversations held by Spanish learners of English of different levels were analysed in order to determine the type of CSs they used when interacting with a native speaker (NS) in an informal environment outside the classroom. The identification of the CSs was carried out following Dörnyei and Körmos’ taxonomy (1998). Overall results show that there is an association between the learners’ proficiency level and their CS usage. Results from a detailed analysis confirmed this relationship and revealed that the learners’ linguistic competence is not only related to the frequency of the CSs used but mostly to the type of CS.Resumen: El objetivo de este estudio fue analizar las diferentes estrategias de comunicación (EsC) que utilizan aprendices de inglés como L2 al comunicarse oralmente, y determinar la relación entre la competencia lingüística de estos estudiantes y el uso de las EsC. Se analizó un corpus oral obtenido de tres conversaciones entre estudiantes de inglés de distintos niveles de competencia con el propósito de descubrir el tipo de EsC que éstos utilizan al interactuar con un hablante nativo en un ambiente informal fuera del aula. La identificación de las EsC utilizadas se realizó mediante la taxonomía propuesta por Dörnyei y Körmos (1998). Desde un punto de vista general los resultados muestran que existe una asociación entre el nivel de competencia lingüística de los estudiantes y las EsC que éstos utilizan. Esto fue corroborado mediante un análisis más detallado de los datos que demostró una relación no sólo entre el nivel de competencia y la frecuencia de uso de las EsC sino principalmente con el tipo de estrategias utilizadas por cada nivel.http://www.scielo.cl/scielo.php?pid=S0718-09342016000100004&script=sci_abstrac
