    Nucleic acids and protein synthesis in cancer cell mitochondria. I. Nucleic acids in rat hepatoma mitochondria

    The contents of nucleic acids in rat liver and hepatoma mitochondria and the physico-chemical properties on DNA's isolated from these mitochondria were comparatively investigated. The results are briefly summarized as follows. 1. The contents of DNA and RNA per mg protein of the hepatoma cell mitochondria were about 10 and 2 to 4 times higher than those of rat liver mitochondria, respectively. 2. The &#955; max. and &#955;min. values of DNA isolated from the hepatoma mitochondria were 257 m&#956; and 231 m&#956;, respectively and those of DNA isolated from the nuclei were 259 m&#956; and 233 m&#956;, respectively, in saline-citrate, pH 7.0. 3. Three fractions of mitochondrial DNA were obtained by the sucrose density gradient and these DNA fractions corresponded, probably, to about 30 S, and 20 S and 14 S DNA's. 4. There was little difference in base compositions between nuclear and mitochondrial DNA's of the hepatoma cells. 5. The degree of hybridization between the nuclear and mitochondrial DNA's of the hepatoma cells was almost the same as that between the nuclear and nuclear DNA's of the hepatoma cells, and somewhat higher than that between the nuclear DNA of rat liver and the nuclear DNA of hepatoma cells. 6. &#34;Highly twisted&#34; circular, &#34;open&#34; circular and linear forms were observed in the DNA preparations of the hepatoma mitochondria. The average values of contour lengths of rat liver and the hepatoma DNA's observed at high frequency were 5.3 &#956; and 4.5 &#956;. 7. A discussion was made on the relation between the genetic informations of mitochondrial DNA and the formation of a mitochondrion in rat liver and the hepatoma cells.</p

    Degassing history of water, sulfur, and carbon in submarine lavas from Kilauea volcano, Hawaii

    Major, minor, and dissolved volatile element concentrations were measured in tholeiitic glasses from the submarine portion (Puna Ridge) of the east rift zone of Kilauea Volcano, Hawaii. Dissolved H_(2)O and S concentrations display a wide range relative to nonvolatile incompatible elements at all depths. This range cannot be readily explained by fractional crystallization, degassing of H20 and S during eruption on the seafloor, or source region heterogeneities. Dissolved C0_2 concentrations, in contrast, show a positive correlation with eruption depth and typically agree within error with the solubility at that depth. We propose that most magmas along the Puna Ridge result from (I) mixing of a relatively volatile-rich, undegassed component with magmas that experienced low pressure (perhaps subaerial) degassing during which substantial H_(2)O, S, and C0_2 were lost, followed by (2) fractional crystallization of olivine, clinopyroxene, and plagioclase from this mixture to generate a residual liquid; and (3) further degassing, principally of C0_2 for samples erupted deeper than 1000 m, during eruption on the seafloor. The degassed end member may form at upper levels of the summit magma chamber (assuming less than lithostatic pressure gradients), during residence at shallow levels in the crust, or during sustained summit eruptions. The final phase of degassing during eruption on the seafloor occurs slowly enough to achieve melt/vapor equilibrium during exsolution of the typically CO_(2)-rich vapor phase. We predict that average Kilauean primary magmas with 16% MgO contain ~0.47 wt% H_(2)O, ~900 ppm S, and have δD values of ~-30 to -40‰. Our model predicts that submarine lavas from wholly submarine volcanoes (i.e., Loihi), for which there is no opportunity to generate the degassed end member by low pressure degassing, will be enriched in volatiles relative to those from volcanoes whose summits have breached the sea surface (i.e., Kilauea and Mauna Loa)

    Analytic philosophy, 1925-1969: emergence, management and nature

    This paper shows that during the first half of the 1960s The Journal of Philosophy quickly moved from publishing work in diverse philosophical traditions to, essentially, only publishing analytic philosophy. Further, the changes at the journal are shown, with the help of previous work on the journals Mind and The Philosophical Review, to be part of a pattern involving generalist philosophy journals in Britain and America during the period 1925-1969. The pattern is one in which journals controlled by analytic philosophers systematically promote a form of critical philosophy and marginalise rival approaches to philosophy. This pattern, it is argued, helps to explain the growing dominance of analytic philosophy during the twentieth century and allows characterising this form of philosophy as, at least during 1925-1969, a sectarian form of critical philosophy

    The author studied the hematopoietic disturbances of rabbit induced by saponin injection and drew the following conclusions: 1) By saponin injection, the structure of bone marrow is disintegrated and hematopoietic cells are released into the circulating blood forming extramedullary hematopietic foci mainly in liver and spleen. The main attacking point of saponin should be RES. Recovery of hematopoietic foci is associated with the recovery of RES. The most marked extramedullary hematopoiesis is found three days after the injection. Thereafter, bone-marrow hematopoiesis proceeds to recovery stage, during which hematopoietic foci in liver and spleen are preserved, especially those in spleen persist fairly for a long time. 2) Daily injections of India ink kept up over a long period of time after the treatment with saponin, prevent the recovery of anemia and bone-marrow hematopoiesis. The lymph nodes, whose RES escaped from the severe damage by India ink, keep the hematopoietic foci for a long time. 3) As far as hematopoiesis is concerned, there seems to be no functional differentiation among RE cells, though they seem to have a special function according to the organs to which they belong, e. g. antibody formation in lymph apparatus, hematopoiesis in bone marrow and red cell destruction in spleen.</p

    Female Cb mice weighing 20-23 g were exposed to 800 ppm (in average) of 1, I, 2, 2.tetrachloroethane for 3 hours. Both triglyceride and phospholipid in the liver and plasma were determined at varying times after the exposure. On the other hand, there were observed the ultraviolet absorption spectra of microwmallipids in the liver at 90 minutes after the 1, 1,2, 2-tetrachloroethane or the carbon tetrachloride exposure. The results thus obtained are summarized as follows: 1. The increase of hepatic triglyceride contents attained the maximum level in the period between 20 and 25 hours after the exposure and declined to the initial levels at 90 hours later. 2. The plasma triglyceride levels decreased until 25 hours after the exposure, then tended to increase significantly and were much higher than the control levels in the period between 70 and 90 hours later. 3. Both liver and plasma phospholipid levels decreased gradually up to 25 hours after the exposure, then slowly recovered with almost the same rate of increase. 4. It was suggested that the inhalation of the above vapors induced a little change in microsomal lipids in the liver.</p
