Effects of High Flavanol Dark Chocolate on Cardiovascular Function and Platelet Aggregation.
Regular consumption of chocolate and cocoa products has been linked to reduced cardiovascular mortality. This study compared the effects of high flavanol dark chocolate (HFDC; 1064mg flavanols/day for 6 weeks) and low flavanol dark chocolate (LFDC; 88mg flavanols/day for 6 weeks) on blood pressure, heart rate, vascular function and platelet aggregation in men with pre-hypertension or mild hypertension. Vascular function was assessed by pulse wave analysis using radial artery applanation tonometry in combination with inhaled salbutamol (0.4 mg) to assess changes due to endothelium-dependent vasodilatation. HFDC did not significantly reduce blood pressure compared to baseline or LFDC. Heart rate was increased by LFDC compared to baseline, but not by HFDC. Vascular responses to salbutamol tended to be greater after HFDC. Platelet aggregation induced by collagen or the thromboxane analogue U46619 was unchanged after LFDC or HFDC, whereas both chocolates reduced responses to ADP and the thrombin receptor activator peptide, SFLLRNamide (TRAP6), relative to baseline. Pre-incubation of platelets with theobromine also attenuated platelet aggregation induced by ADP or TRAP6. We conclude that consumption of HFDC confers modest improvements in cardiovascular function. Sequence-specific protein aggregation generates defined protein knockdowns in plants
Protein aggregation is determined by short (5-15 amino acids) aggregation-prone regions (APRs) of the polypeptide sequence that self-associate in a specific manner to form beta-structured inclusions. Here, we demonstrate that the sequence specificity of APRs can be exploited to selectively knock down proteins with different localization and function in plants. Synthetic aggregation-prone peptides derived from the APRs of either the negative regulators of the brassinosteroid (BR) signaling, the glycogen synthase kinase 3/Arabidopsis SHAGGY-like kinases (GSK3/ASKs), or the starch-degrading enzyme alpha-glucan water dikinase were designed. Stable expression of the APRs in Arabidopsis (Arabidopsis thaliana) and maize (Zea mays) induced aggregation of the target proteins, giving rise to plants displaying constitutive BR responses and increased starch content, respectively. Overall, we show that the sequence specificity of APRs can be harnessed to generate aggregation-associated phenotypes in a targeted manner in different subcellular compartments. This study points toward the potential application of induced targeted aggregation as a useful tool to knock down protein functions in plants and, especially, to generate beneficial traits in crops
Aggregation Behavior And Chromonic Liquid Crystal Properties Of An Anionic Monoazo Dye
X-ray scattering and various optical techniques are utilized to study the aggregation process and chromonic liquid crystal phase of the anionic monoazo dye Sunset Yellow FCF. The x-ray results demonstrate that aggregation involves pi-pi stacking of the molecules into columns, with the columns undergoing a phase transition to an orientationally ordered chromonic liquid crystal phase at high dye concentration. Optical absorption measurements on dilute solutions reveal that the aggregation takes place at all concentrations, with the average aggregation number increasing with concentration. A simple theory based on the law of mass action and an isodesmic aggregation process is in excellent agreement with the experimental data and yields a value for the bond energy between molecules in an aggregate. Measurements of the birefringence and order parameter are also performed as a function of temperature in the chromonic liquid crystal phase. The agreement between these results and a more complicated theory of aggregation is quite reasonable. Overall, these results both confirm that the aggregation process for some dyes is isodesmic and provide a second example of a well-characterized chromonic system
The impossibility of unbiased judgment aggregation
Standard impossibility theorems on judgment aggregation over logically connected propositions either use a controversial systematicity condition or apply only to agendas of propositions with rich logical connections. Are there any serious impossibilities without these restrictions? We prove an impossibility theorem without systematicity that applies to most standard agendas: Every judgment aggregation function (with rational inputs and outputs) satisfying a condition called unbiasedness is dictatorial (or effectively dictatorial if we remove one of the agenda conditions). Our agenda conditions are tight. Applied illustratively to (strict) preference aggregation represented in our model, the result implies that every unbiased social welfare function with universal domain is effectively dictatorial.mathematical economics;
Aggregation functions with given super-additive and sub-additive transformations
Aggregation functions and their transformations have found numerous applications in various kinds of systems as well as in economics and social science. Every aggregation function is known to be bounded above and below by its super-additive and sub-additive transformations. We are interested in the “inverse” problem of whether or not every pair consisting of a super-additive function dominating a sub-additive function comes from some aggregation function in the above sense. Our main results provide a negative answer under mild extra conditions on the super- and sub-additive pair. We also show that our results are, in a sense, best possible
