Application of IR spectroscopy and Raman spectroscopy in the search for potential diagnostic and prognostic markers in the serum and bone marrow of pediatric patients with acute lymphoblastic leukemia (ALL)

UR - 199 s.Ostra białaczka limfoblastyczna (ALL) jest najczęściej diagnozowanym nowotworem u dzieci, a wysokie wskaźniki przeżywalności są efektem precyzyjnej diagnostyki, i optymalnego doboru terapii. Standardowe metody rozpoznania ALL są jednak czasochłonne, kosztowne i inwazyjne, co uzasadnia poszukiwanie nowych, szybkich i mniej obciążających narzędzi diagnostycznych. Celem pracy była ocena przydatności spektroskopii FTIR i Ramana w analizie surowicy krwi oraz lizatu komórkowego szpiku kostnego dzieci z ALL w kontekście identyfikacji potencjalnych markerów diagnostycznych i prognostycznych. Porównano widma 48 pacjentów z ALL, 43 pacjentów z innymi zaburzeniami hematologicznymi oraz 15 dzieci zdrowych. Spektroskopia FT-IR i Ramana wykazały ograniczoną przydatność w identyfikacji specyficznych zmian molekularnych w przebiegu ostrej białaczki limfoblastycznej u dzieci. Niemniej jednak ujawnione różnice w wartościach absorpcji pików w widmach FT-IR oraz w udziale struktur α-helisy i β-kartki białek mogą stanowić potencjalne markery diagnostyczne w różnicowaniu ALL jako przyczyny anemii i pancytopenii krwi obwodowej u dzieci oraz w ich wstępnej stratyfikacji do grup ryzyka. Z kolei różnice w intensywności pików w widmach Ramana surowicy krwi umożliwiały odróżnienie pancytopenii w przebiegu ALL od innych przyczyn.Acute lymphoblastic leukemia (ALL) is the most frequently diagnosed cancer in children, and the high survival rates are attributed to accurate diagnostics and the optimization of therapeutic strategies. However, standard diagnostic methods for ALL are timeconsuming, costly, and invasive, which justifies the search for new, rapid, and less burdensome diagnostic tools. The aim of this study was to evaluate the usefulness of Fourier-transform infrared (FT-IR) and Raman spectroscopy in the analysis of peripheral blood serum and bone marrow cell lysates from children with ALL, with respect to the identification of potential diagnostic and prognostic biomarkers. Spectra from 48 patients with ALL, 43 patients with other hematological disorders, and 15 healthy children were compared. FT-IR and Raman spectroscopy demonstrated limited utility in identifying specific molecular changes associated with the course of acute lymphoblastic leukemia in children. Nevertheless, the observed differences in peak absorbance values in FT-IR spectra and in the relative contribution of protein α-helix and β-sheet structures may serve as potential diagnostic markers for differentiating ALL as a cause of anemia and pancytopenia in children, as well as for their preliminary risk stratification.
