Pengaruh pemberian vitamin E (a-Tocoferol) dalam media DMEM (Dulbeccos modified eagles medium) terhadap proliferasi sel paru-paru fetus hamster kultur primer

Abstract

INDONESIA: Vitamin E mempunyai rantai Phythyl yang terdiri dari 3 isoprenoid dan bersifat hidrofobik, sehingga vitamin E dapat masuk ke dalam membran sel dan bekerjasama dengan molekul transduktor sinyal untuk mengaktifkan enzim protein kinase. Enzim protein kinase yang teraktifasi akan mengaktifkan protein faktor transkripsi. Protein faktor transkripsi akan berikatan dengan segmen promoter yang akan memicu jalannya transkripsi sehingga siklus sel menjadi lebih cepat. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian vitamin E (α-tocoferol) dalam media DMEM (Dulbeccos Modified Eagles Medium) terhadap proliferasi sel kultur primer paru-paru fetus hamster. Penelitian ini merupakan penelitian eksperimental, menggunakan RAL (Rancangan Acak Lengkap) dengan 6 perlakuan dan 4 kali ulangan. Analisis data menggunakan ANAVA tunggal 1%, apabila terdapat perbedaan sangat nyata maka dilanjutkan dengan uji lanjut. Perlakuan yang digunakan adalah pemberian vitamin E (α-tocoferol) dengan berbagai konsentrasi P0 (kontrol), P1 (25 µM), P2 (50 µM), P3 (75 µM), P4 (100 µM), dan P5 (125 µM). Sampel yang digunakan adalah sel paru-paru fetus hamster berumur 2 hari yang dikultur dalam media DMEM + FBS 20% . Sel paru-paru fetus hamster diinkubasi dalam inkubator CO2 dengan suhu 37oc selama 96 jam, kemudian diamati konfluen, viabilitas dan abnormalitas sel paru-paru fetus hamster. Hasil penelitian menunjukkan bahwa ada pengaruh pemberian vitamin E (α- tocoferol) dalam media DMEM (Dulbeccos Modified Eagles Medium) terhadap proliferasi (konfluen, viabilitas, dan abnormalitas) sel paru-paru fetus hamster. Konsentrasi vitamin E (α-tocoferol) dalam media DMEM (Dulbeccos Modified Eagles Medium) yang berpengaruh terhadap peningkatan konfluen sel paru-paru fetus hamster ditemukan pada konsentrasi P1 (25 µM), tetapi pada konsentrasi tersebut dapat menurunkan abnormalitas sel paru-paru ferus hamster. Peningkatan viabilitas sel paru-paru fetus hamster terjadi pada konsentrasi P2 (50 µM. ENGLISH: Vitamin E is a vitamin that has Phythyl chain consisting of three isoprenoid, and are hydrophobic. Vitamin E can get into the cell membrane, and then work with transduktor to activate the receptor molecule that binds to the ligand in the form of the enzyme protein kinase. The enzyme protein kinase that is activated boom will activate the transcription factor protein. Protein transcription factor binds to the promoter segman will trigger the course of transcription so that the cell cycle becomes faster and be quicker confluent cells. This study aims to determine the effect of vitamin E (α-Tocoferol) in DMEM medium (Dulbeccos Modified Eagles Medium) on cell proliferation of primary cultured fetal hamster lung. This study is an experimental study using the (Completely Randomized Design) with 6 treatments and 4 replications. Analysis of data using a single ANAVA 1%, if there is a very real difference then proceed with further testing LSD (Smallest Real Difference), BNJ (Beda Real Honest), and UJD (Duncan Test Distance) 1%. The treatments that are used are vitamin E (α-Tocoferol) with a concentration of P0 (control), P1 (25 µM), P2 (50 µM), P3 (75 µM), P4 (100 µM), and P5 (125 µM). The samples used are fetal lung cells 2-day-old hamsters that were cultured in DMEM medium + 20% FBS. Lung cells were then incubated in fetal hamster CO2 incubator with 37 ° C for 96 hours, then observed confluent, and abnormalities of cell viability of primary cultured fetal hamster lung. The results showed that there is the effect of giving vitamin E (α-Tocoferol) in DMEM medium (Dulbeccos Modified Eagles Medium) on cell proliferation of primary cultured fetal hamster lung. In this research shows that vitamin E (α-Tocoferol) in medium (Dulbeccos Modified Eagles Medium) effect on cell proliferation of primary cultured fetal lung hamster.ditemukan at treatment P1 (25 µM) to confluent, viability P2 (50 µM), and abnormality of P1 (25 µM)
