The Multifaceted Role of Hypoxia‐Inducible Factor 1 (HIF1) in Lipid Metabolism

    Get PDF
Hypoxia‐inducible factor 1 (HIF1) is a master transcription factor and regulates expression of a large number of genes involving many aspects of biology. In addition to HIF1\u27s roles in glucose metabolism and angiogenesis, numerous studies have revealed an emerging role of HIF1 in controlling lipid homeostasis. In this chapter, we discuss that lipid accumulation is related to HIF1\u27s activity in several diseases and the growing evidence demonstrating the functional importance of HIF1 in controlling lipid metabolism. The functions include lipid uptake and trafficking, fatty acid metabolism, sterol metabolism, triacylglycerol synthesis, phospholipid metabolism, lipid droplet biogenesis, and lipid signaling. Defining the role of HIF1 in lipid metabolism is crucial to understand the pathophysiology of lipid in disease and may help us to identify additional target sites for drug development. This review would shed light on our understanding of the critical role of HIF1 in lipid metabolism

Oxidative Degradation of Fentanyl in Aqueous Solutions of Peroxides and Hypochlorites

Fentanyl widely used in clinics as practices is potentially utilised as an incapacitant in countering terrorism and its analogues would be used as an addictive drug, so the degradation of these compounds need to be investigated for protecting environment and human health. In this work, the degradation of fentanyl was examined in a series of oxidant aqueous solutions. Meanwhile, the degradation pathways of fentanyl in the oxidant solution were discussed according to all products identified by GC/MS and LC/MS.Defence Science Journal, 2011, 61(1), pp.30-35, DOI:http://dx.doi.org/10.14429/dsj.61.6

Structures of an intramembrane vitamin K epoxide reductase homolog reveal control mechanisms for electron transfer

    Get PDF
The intramembrane vitamin K epoxide reductase (VKOR) supports blood coagulation in humans and is the target of the anticoagulant warfarin. VKOR and its homologs generate disulfide bonds in organisms ranging from bacteria to humans. Here, to better understand the mechanism of VKOR catalysis, we report two crystal structures of a bacterial VKOR captured in different reaction states. These structures reveal a short helix at the hydrophobic active site of VKOR that alters between wound and unwound conformations. Motions of this “horizontal helix” promote electron transfer by regulating the positions of two cysteines in an adjacent loop. Winding of the helix separates these “loop cysteines” to prevent backward electron flow. Despite these motions, hydrophobicity at the active site is maintained to facilitate VKOR catalysis. Biochemical experiments suggest that several warfarin-resistant mutations act by changing the conformation of the horizontal helix. Taken together, these studies provide a comprehensive understanding of VKOR function

STUDY ON THE RELATION BETWEEN SELF CONSISTENCY AND CONGRUENCE AND MENTAL HEALTH OF POSTGRADUATES

Reveal the Relation between Self consistency and congruence and Mental Health of Postgraduates. Adopt self-consistency and congruence questionnaire, mental health questionnaire to carry out questionnaire investigation on 500 postgraduates of four Nanjing colleges and universities. Mental health of postgraduates has extremely negative notable relevance with the disharmony of oneself and experience and it has extremely negative notable relevance with self-flexibility. The degree of self consistency and congruence and self-flexibility of postgraduates with better psychological health situation is higher than that of postgraduates with poor mental health situation. Disharmony of oneself and experience, flexibility function of oneself forecast mentality symptom dissociation amounts to a certain extent. The degree of self-consistency and congruence has a significant influence on mental health

A diverse global fungal library for drug discovery

    Get PDF
Background: Secondary fungal metabolites are important sources for new drugs against infectious diseases and cancers. Methods: To obtain a library with enough diversity, we collected about 2,395 soil samples and 2,324 plant samples from 36 regions in Africa, Asia, and North America. The collection areas covered various climate zones in the world. We examined the usability of the global fungal extract library (GFEL) against parasitic malaria transmission, Gram-positive and negative bacterial pathogens, and leukemia cells. Results: Nearly ten thousand fungal strains were isolated. Sequences of nuclear ribosomal internal transcribed spacer (ITS) from 40 randomly selected strains showed that over 80% were unique. Screening GFEL, we found that the fungal extract from was able to block transmission to , and the fungal extract from was able to kill myelogenous leukemia cell line K562. We also identified a set of candidate fungal extracts against bacterial pathogens

A fast and accurate method to predict reliability of project completion time

    Get PDF
To meet the target completion time of a construction project is one of the most important performance indicators of project management. This paper proposes a fast and accurate method for evaluating the reliability of project completion time in large construction projects, using reliability theory. The proposed method is developed to overcome the limitations of existing methods, including the inaccuracy of the program evaluation and review technique and the long computational time of the narrow reliability bounds method. The proposed method is established in three main parts: (i) calculating the statistics of paths duration in the network; (ii) truncating insignificant paths of the network; and (iii) proposing an innovative solution to accurate estimate for reliability of project completion time. The effectiveness of the proposed method is evaluated using an example project. It is found that the results of the proposed method on the reliability of completion time are accurate. It is also found that the proposed method significantly reduces the number of analysed network paths and the computational effort. The method proposed here can serve as a fast and accurate tool for project managers and project planners in project planning, re-planning, and project control phases.
