Two isolates were identified as “Burkholderia cepacia complex” (scores 1.96 and 1.98). These two isolates were re-evaluated after an extraction in tube with formic acid and acetonitrile and one isolate was identified as B. cenocepacia (score 2.27) and the other was confirmed as  “Burkholderia cepacia complex” (score 2.16). The relationship between the structure and the composition of the microbiome of the CF patient represents an important factor in his states of health. For microbiome analysis, the quality of DNA extracted has a pivotal impact in the sample representativeness. The aim of this study was to evaluate different protocols of DNA extractions in order to obtain the best method to be used in the microbiome analysis of sputum from Cystic Fibrosis (CF) patients. The extraction kit QIAamp DNA Mini Kit (QIAgen, Valencia CA) was used with three different pre-treatments: The first procedure (1A) followed the manufacturer protocol recommendation. The second procedure (1B) included a proteinase K treatment for 60 min at 56°C followed by bead-beating with zirconia/silica bead in a FastPrep 24 5G system (Qbiogene, CA), for  10 seconds at 6.0 m/sec (repeated 4 times). The third procedure (1C) was based in the sputum digestion with equal volume of N-acetyl-cysteine (100mg/mL). The last procedure (1D) used a TE buffer (10mM Tris-HCl [pH 8.0], 1mM EDTA) with the sputum and bead-beating as described above. The concentration of DNA was measured at Qubit 3.0 Fluorometer (Thermofisher) and the DNA integrity was verified with the 4200 Tape Station (Agilent). The amplicon library was prepared following the Illumina 16S Metagenomic Sequencing Library Preparation protocol and the high-throughput sequencing performed with MiSeq 600V3 kit. An average of 190,000 reads were obtained per sample. The total genus-level of taxonomic categories identified (TCI) was approximately 215 per sample. The total of species-level TCI was: 238 for 1A; 320 for 1B; 179 for 1C and 241 for 1D.  We selected a Pseudomonas aeruginosa and a Rothia mucilaginosa to evaluate the effectiveness of DNA extraction from gram-negative and gram-positive bacteria. The detection of P. aeruginosa and R. mucilaginosa, respectively, for each protocol extraction was: 1A, 22.08% and 0.85%; 1B 29.10% and 4.46%; 1C, 31.15% and 0.68% and 1D, 26.92% and 8.56%. In conclusion, the protocol 1B presented the best performance considering the total of species-level TCI. Noteworthy, both protocols 1B and 1D, which use the bead-beating strategy, increased the yield of Gram-positive bacterial DNA extraction.   eP2305 Avaliação da resposta de células de adenocarcinoma ductal pancreático PANC-1 à gemcitabina em perfil de tratamento semelhante ao protocolo clínico: foco no papel da autofagia Ronize Zeni da Silva; Solon Andrades da Rosa; Paula Ferst; Stefano Walter Agati; Mariana Lobo; Viviane Rosner de Almeida; Eduardo Cremonese Filippi Chiela; Patrícia Luciana da Costa Lopez UFRGS - Universidade Federal do Rio Grande do Sul  O câncer de pâncreas é a terceira causa de morte por câncer nos Estados Unidos e a sétima no Brasil. Menos de 10% dos pacientes sobrevivem 2 anos livres de doença. Dentre os diversos subtipos o mais comum é o Adenocarcinoma Ductal de Pâncreas (ADP). Apesar dos avanços observados na terapia antitumoral nas últimas décadas, quimioterápicos clássicos como a Gemcitabina (GEM) continuam sendo uma alternativa primária para terapia do ADP. Porém, a resistência e recorrência tumorais são frequentes, e são escassos os trabalhos que avaliam a resposta das células de ADP à GEM a longo prazo. Sendo assim, nós utilizamos um racional experimental de tratamento agudo (48h) seguido do crescimento das células tumorais em Meio Livre de Droga por 10 dias (período de recuperação), para mimetizar o período de recuperação dos pacientes. A partir deste modelo nós avaliamos o efeito da GEM na viabilidade fenotípica celular, na clonogenicidade das células tumorais e nos níveis de marcadores de autofagia tanto após o tratamento agudo (48h) quanto após 5 e 10 dias do tratamento. Por fim, avaliamos a relação entre níveis de autofagia e sobrevivência celular durante o período de recuperação. Nós observamos que o tratamento com GEM 10 e 30 mM por 48h causou uma redução do número de células com fenótipo viável a longo prazo. Para o tratamento com a dose GEM 1mM observamos redução desta população celular 5d após o tratamento, porém esta redução não se acentuou 10d após o tratamento. O tratamento com GEM também reduziu a clonogenicidade das células e aumentou indicadores de autofagia a longo prazo (medido pela marcação com laranja de acridina e complexidade intracelular, ambos por citometria de fluxo). Finalmente, observamos que células com fenótipo viável após 5d e 10d apresentaram níveis mais intensos de marcação com laranja de acridina, sugerindo que a autofagia atue favorecendo a sobrevivência das células de ADP em resposta à GEM. Concluímos, assim, que as células de ADP ativam autofagia possivelmente como mecanismo de citoproteção à GEM, e que o desenho experimental utilizado possui características que mimetizam, pelo menos parcialmente, o comportamento de recorrência tumoral observado clinicamente.
