BABAM1: A Key Regulator for Serine Synthesis Pathway via PHDGH in Breast Cancer Cell Lines

Abstract

癌症已经成为威胁人类健康的最大隐患,而环境(含生活方式)与遗传因素的交互作用成为重要因素。在癌谱中,乳腺癌占据女性肿瘤的首位,利用生态毒理学技术揭示其细胞生物学通路具有重要科学价值。伴随着丝氨酸合成通路在癌症细胞中维持能量供应与提供生物大分子合成原料的重要性被揭示出来,肿瘤细胞对丝氨酸所特有的依赖性也呈现在人们面前,成为近年来的研究热点。而BABAM1基因在细胞中本身作为BRCA1基因的DNA链断裂修复复合体的组成部分,其在细胞代谢中的功能却并不明了。本文首先通过免疫共沉淀与蛋白组学手段,筛选出与BABAM1基因相互作用的基因PHGDH——这一在丝氨酸合成通路中一个重要的限速酶。随后通过分子生...Cancer has become the most serious threat towards human health over the past decades.
