Structure-based design of 2A protease inhibitors from Enterovirus 71

Abstract

手足口病(HFMD)是一种多发于5岁以下幼儿,由肠道病毒所引起的急性传染病,最常见病毒是柯萨奇病毒A16型(CVA16)和肠道病毒71型(EV71)。EV71的感染尤其可能导致严重的中枢神经系统疾病及相关并发症。近几年由EV71的感染呈现强传染性、高死亡率的趋势,而对于该疾病目前仍缺乏有效的对症治疗药物。 2A蛋白酶是EV71成熟和感染的重要因子,切割病毒多聚蛋白;参与病毒的抗干扰素通路;压制宿主的先天性免疫反应;促进病毒核酸的复制与蛋白质翻译;诱导细胞凋亡等作用。所以它是由EV71引起的手足口病的重要药物靶点,而至今还没有针对2A蛋白酶的特异抑制剂。 本课题中我们解析了EV712A(C1...Hand, foot and mouth disease (HFMD) is an acute infectious disease caused by enteroviruses mostly for children under 5 years old.
