刚地弓形虫表面抗原研究进展汪　琦 (综述 )　林宇光　洪凌仙 (审校 )作者单位:厦门大学寄生虫研究室 (厦门 , 361000)　　刚地弓形虫 ( Toxoplasma gondi i )是一种细胞内寄生原虫 ,可感染任何有核细胞 ,无论人或动物 ,感染率都很高。和其它细胞一样 ,弓形虫的表膜具有重要的作用 ,既是虫体与外界环境进行物质交换的界面 ,也是宿主免疫系统识别并杀伤虫体的主要部位。Handman, Goding和 Reming ton( 1980)首先对弓形虫表面抗原进行了分析 ,其后 Johnson ( 1983)、Dubremetz( 1985)、 Rodriguez( 1985) 、 Kasper( 1982,1984, 1987)亦对一些表面抗原进行了研究 ,虽然已发现的抗原种类很多 ,但由于技术方法差异 ,命名很混乱。 G. Couver( 1980)利用单克隆抗体技术对表面抗原进行了确认。随着分子生物学技术的发展 ,目前对表面抗原的研究已深入到基因水平 ,已能对编码P30、 P22和 P43的基因进行克隆和测序。 本文将对近年来弓形虫表面抗原的研究进展作如下综述。　　 1　速殖子的表面抗原1. 1　蛋白抗原　目前已知的速殖子表面蛋白抗原有五种 ,根据 SDS- PAGE中显示的分子量大小 ,分别命名为 P43、 P35、 P30、 P23和 P22〔1— 4〕。 编码 P30抗原的基因首先被克隆、测序 ,根据分子遗传学的传统命名方法 ,编码 P30的基因被称为 SAG〔S〕。随后 ,编码 P22和 P43的基因相继被克隆和测序 ,依次命名为 SAG2和 SAG3。至今编码 P23和 P35的基因仍无法克隆、测序。 SAG1、 SAG2和 SAG3的克隆均采用特异抗体和相关抗原对表达文库进行筛查的传统方法所得 ,三者具有一些明显特点:1. 1. 1　都具有一个暴露于外表 ,用于转运的 N- 末端信号肽 ;1. 1. 2　都具有一个疏水性 C端 ,这表明这些抗原被GPI部分固定 ;1. 1. 3　 SAG1和 SAG3具有 24% aa一致性 ,极有可能具相似的二级结构和三级结构。 PAGE的相对迁移率表明 , SAG3对应于 SAG1有一些插入组分 ,这导致两者大小差异。1. 1. 4　 SAG2具有一些独特的性质 ,它虽和 SAG1相似 ,具磷酸化多肽 ,但除此之外 ,和 SAG1、 SAG3再无共同之处 ;1. 1. 5　 SAG1序列显示了一个 N端相连糖基化位点 ,而 SAG2和 SAG3序列则无此位点。1. 2　 SAG多态性　弓形虫主要有三型 ,习惯上称为Ⅰ 、Ⅱ、Ⅲ 〔 6〕。对代表三型的 3株弓形虫 RH、 M49株和 CEP的 SAG1进行测序〔1, 5, 7〕 ,结果表明: 和 ME49(Ⅱ型 )的等位基因相比较 , RH株 (Ⅰ株 ) S AG1编码 8个 aa差异。 有趣的是 ,这种差异导致了 ME49株的SAG1中一个潜在 N - 相连位点的产生。 ME49和CEP核酸序列只具有一个非编码差异 ,两者的等位基因编码相同的蛋白。 Sibley等〔 6, 8〕先后对 26株和 106株的弓形虫 SAG1进行 RFLP分析 ,结果表明该位点具显著的双态性 , 95%以上的被检出株为仅有的两种RFLP类型之一。而且仅在Ⅰ 型中发现的等位基因 1和Ⅱ型、Ⅲ型中发现的等位基因 2之间具密切的联系。由于 RFLP分析仅涉及三种代表株 RH、 ME49和CEP中的一些限制性位点 ,该位点是否具高度保守性尚有争议。　　不少学者对 SAG2进行了同样的分析 ,结果表明SAG2亦具有有限的多态性 ,在 10株弓形虫中仅检测到 2个等位基因〔 9〕。 两种等位基因具 5个核苷酸差异 ,其中 4个核苷酸导致 aa变化。多余密码子的有无亦是两种等位基因的差异点 ,推测 ,等位基因的这些差异可能是 SAG2抗原性有别于其它抗原的原因。1. 3　 SRS家族　速殖子表面还存在着另一类抗原 ,即 SRS家族—— SAG1相关序列 ( SAG1 - related se-quence)。 SRS1是第四个被克隆的速殖子表面抗原 ,它是在研究 SAG1转录时发现的。当置于质粒上和转导入寄生虫时 , SAG1起动子表现出双重直接功能 ,这表明在 SAG,上游区有可能存在发散表达的基因 ,且为速殖子特异性〔 10〕。以后的研究终于发现 SAG1上游1～ 2kb处 ,存在着 SAG1类似物 ,它和 SAG1、 SAG3具 25%的同源性 ,并和 SAG1的转录方向相同 ,因此被称为 SRS1〔 11〕。 SRS1产生的融合蛋白免疫小鼠结果表明: SRS,亦是一种表面抗原 ,且仅在速殖子表面表达。 Westernblo t结果显示 SRS1的含量远远少于912000, 16 ( 4) 　　　　　　　　　　中 国 人 兽 共 患 病 杂 志Chinese Jour nal o f Zoonoses　SAG1和 SAG3 ,这可能是它在表面标记研究中被遗漏的原因。　　目前由弓形虫产生的 ESTs( expressed sequencetags )已逾 10000,从中又发现了 4种 SRS家族的基因。 Ests来源于 CDNA文库 ,能否表达尚属未知。为此 , Mange ID等进行了实验 ,选取 3种 SRS基因表达的融合蛋白免疫小鼠 ,产生抗血清 ,结果表明三者均在速殖子表面表达。 最近一个被命名为 SAG5的SRS基因被克隆、鉴定 ,该基因的表达亦在研究中〔12〕。1. 4　蛋白抗原的功能　目前对蛋白质抗原的功能知之甚少。 SAG1和 SAG3在吸附和入侵宿主细胞时起着一定的作用〔 13— 14〕 ,而 SRS家族的作用却完全未知 ,它可能是多余的受体 ,但亦可能在寄生虫和宿主免疫反应间的相互作用中起着重要的作用。有人认为 ,弓形虫宿主广泛 ,每个宿主都有不同的分子有待弓形虫识别 ,因此 ,弓形虫进化了一个受体家族和宿主的配体相接合 ;也有人认为 ,弓形虫具有入侵不同宿主的任何有核细胞的特殊能力 ,原因可能在于受体家族能处理所感染的宿主不同组织间的差异 ;当然 ,亦有可能 SRS家族确实是多余的受体 ,各种 SRS基因均为相同的受体 ,但对其基因序列分析结果表明 SRS基因仅显示 25%同源性 ,该结果并不支持这种观点。　　任何生物体的生存都不能依靠自身的无限度繁殖 ,速殖子表面抗原的最基本功能之一可能是对自身的抑制和破坏。速殖子的特点之一是迅速地分裂然后散布到身体各处 ,宿主受损 ,死亡越快 ,则形成的组织包囊数量越少 ,传播效率越低 ,这对种的生存并无益处 ,因此 ,速殖子表面抗原为了种的生存而对速殖子的无限增殖进行抑制和破坏 ,它刺激宿主引起抗速殖子的免疫反应 ,进而调节感染毒力。对 SAG1的研究已经证实了表面抗原的这种功能〔1, 4, 15, 16〕。 SAG1是高免疫原性 ,宿主对它产生的强烈免疫反应可使宿主不同杀灭缓殖子即可控制感染。速殖子表面抗原的这种特性可应用于寄生虫的防治。　　表面抗原的功能远不止上述几个方面。但目前的研究表明已知的表面蛋白质抗原与信号传递、营养吸收与转运等无关 ,而且 SRS家族均为 GPI锚住蛋白 ,不可能是转运或信号蛋白的组成部分 ,可能速殖子表面尚有其它的蛋白质抗原 ,因含量太低 ,目前尚缺乏敏感的检测手段。2　缓殖子表面抗原　　缓殖子表面抗原研究集中于蛋白质成份分析 ,已确定缓殖子表面不具有 SAG1和 SAG2 ,但却有SAG3存在。 缓殖子表面存在着许多特异性蛋白 ,Tomavo et al制备了针对缓殖子特异性抗 P36、 P21和 P18的单克隆抗体〔17〕。 随后 , Odberg Ferragut etal用单抗对编码 P18的基因进行了克隆和测序〔 18〕 ,由于该基因是弓形虫中第四个被克隆和测序的基因 ,故称为 SAG4。 P18抗原和已知抗原无任何相似之处 ,对其功能也是一个无所知。 最近从缓殖子ESTs中得到一个基因 ,该基因和 SAG4具 40%的同源性 ,该基因的表达情况及两者的功能正在研究之中。　　缓殖子的表面抗原中 , P36无疑是最重要的抗原之一 ,因为它是速殖子向缓殖子转变时最先出现的特异性蛋白 ,是研究期转变机理时重要的特征性蛋白。尤其令人兴奋的是 ,对编码该抗原的基因分析显示它是 SRS家族的一个成员〔19〕。据此可以推测 ,缓殖子表面的 P36是速殖子 SAG1的替代物 ,但是弓形虫为什么进化出这种类似蛋白期特异性表达的现象 ,仍是个谜。可能是由于不同的组织向性 ,亦有可能因为 SAG1是免疫反应的主要目标 ,缓殖子不具有这种高免疫原性蛋白是一种适应〔12〕。3　子孢子表面抗原　　由于很难得到大量的卵囊 ,至今对子孢子研究不多。子孢子表面亦具有许多特异性蛋白 ,其中最主要的两种约为 25kDa和 67kDa〔20〕。在子孢子表面未发现 SAG1和 SAG2 ,但存在着一种和 SAG3一起移动的蛋白质。尤其值得注意的是 ,子孢子入侵小肠细胞后很快就分化成可表达 SAG1的一种形式 ,推测该形式属速殖子期〔21〕。4　结　论　　弓形虫表面抗原的研究较多关注于速殖子 ,这主要因为速殖子引起急性感染 ,危害较大。从已有的研究可以看出 ,速殖子表面抗原既复杂又简单 ,抗原的种类远不止传统的表面标记。5　参考文献1.
