Isoenzyme analysis of genetic diversity in cultured Hapalogenys nitens stock

Abstract

应用聚丙烯酰胺凝胶电泳 (PAGE)技术 ,对取自广东饶平海水网箱养殖的斜带髭鲷 (Hapalogenysnitens)人工苗养殖群体同工酶的多态性进行分析 .结果表明 :在对 48尾鱼所检测出的 1 1种同工酶的 2 3个基因座位中 ,超氧化物歧化酶 (SOD)、苹果酸脱氢酶 (MDH)和过氧化物酶 (PER)等酶的基因座位上发现多态性 ,其平均多态位点比例 (P)为 1 7.39% ,基因座位有效等位基因的平均数 (Ne)为 1 .1 74,平均杂合度的观测值 (Ho)为 0 .0 51 ,预期值 (He)为 0 .0 39,Hardy Weinberg遗传偏离指数 (D)为 0 .2 91 .这说明该养殖群体具有较高的遗传变异水平 .Isoenzymes isolated from cultured Hapalogenys nitens stock collected from Raoping in Guangdong in January 2001 were investigated by polyacrylamid gel electrophoresis (PAGE) in this paper. Among the 23 loci in 11 enzymes examined, three loci polymorphic were found. The mean level of polymophism is 17.39% and heterozygosity is 0.051. Isoenzymes analysis values of H.nitens are mediate within the range of the values reported for other marine fish populations.
