A lesser degree of podocyte foot process effacement, basal membrane thickening, and abnormal mitochondria were found in EM. Conclusion: The findings provide evidence that melatonin restores renal alterations of LN in the pristane mice model, by modulating inflammation, oxidative stress and profibrotic markers. The dorsolateral prefrontal cortex (DLPFC) plays an important role in food intake regulation and may be an interesting target to transcranial direct current stimulation (tDCS), a new modality of treatment that has been associated with food craving reduction. Objective: To analyze the effect of repetitive tDCS over the right DLPFC (rDLPFC) associated with a hypocaloric diet on the quality of carbohydrate (CH) consumed by overweight or obese adults. Methods: In this randomized, placebo-controlled, double-blind study, overweight/obese adults, aged 20-50 years, completed a 4-week (20 sessions) protocol of fixe-dose tDCS (2mA,20 min). Subjects were randomized in 1:1 ratio to receive one of two types of intervention: (1)active tDCS+hypocaloric diet (AG), or (2)sham tDCS+hypocaloric diet (SG), both over the rDLPFC. The dietary behavior was assessed by 3-day food records (2week-days;1weekend-day) on 3 times: baseline (t 0 ), first fortnight (t 15 ) and final (t F ). The quality of CH consumed was assessed by the mean glycemic index (MGI) and mean glycemic load (MGL) per meal per day. Blood samples were collected at t 0 and t F in a 2hOGTT and for A1c determination. Changes over the time (t F -t 0 ) for MGI (ΔMGI) and MGL (ΔMGL) were analyzed with GEE and were adjusted for weight reduction %. Spearman&#39;s correlation between ΔMGI and ΔMGL versus 2-h OGTT and HbA 1 c was tested. Results are reported as means±SD, means [95%IC] or %. Results: Twenty subjects (male 50%, obese 75%, mean age 38.8±5.2, BMI 31.2±2.2 kg/m²) were randomized. Changes in MGI were not significant between groups over time (p=0.92). Despite the great reduction in ΔMGL from t 0 to t F in the AG (−23.5[−35.3, −11.6]) versus SG (−14.0[−19.6,−8.4]), these changes did not reach statistical differences (p=0.08). The relationship between ΔMGL and Δ2h plasma glucose (Δ2hPG) and ΔA1c was statistically significant for AG. While Δ2hPG and ΔA1c did not vary with changes in ΔMGL for SG (p=0.85 and p=0.17, respectively), the greater the ΔMGL reduction, the greater the Δ2hPG and ΔA1c reduction for AG (r=0.745, p=0.02; r=0.727, p=0.03). Conclusion: Preliminary analysis suggests that repetitive tDCS over the rDLPFC may help improving glycemic profile by reducing meal’s mean glycemic load. AO1819 Jejum intermitente como estratégia não-farmacológica na promoção de efeito ansiolítico  Lizia Nardi Menegassi, Afonso Kopzynski, Nathan Ryzewski Strogulski, Monia Sartor, Wanda Osório Dorneles, Marcelo Salimen Rodolphi, Randhall B Carteri, Luis Valmor Portela - UFRGS  Introdução: O Brasil é o país com a maior taxa de pessoas com transtornos de ansiedade no mundo, atingindo 9,3% da população. Drogas ansiolíticas reconhecidamente promovem uma melhora na função mitocondrial associada à diminuição do comportamento ansioso. Adicionalmente, o jejum intermitente (JI) modula positivamente a função mitocondrial em diferentes regiões cerebrais. Nesse contexto, embora exista uma relação robusta entre o prejuízo no metabolismo mitocondrial e os transtornos de ansiedade, estratégias não-farmacológicas de tratamento permanecem pouco exploradas. Objetivos: Investigar os efeitos do JI no comportamento tipo-ansioso e na neuroenergética mitocondrial. Métodos: Camundongos C57BL/6J foram alocados em dois grupos: Dieta Normal (DN;  n=10) com acesso irrestrito a ração padronizada e JI (n=10), submetidos a 24h de restrição alimentar seguidos de 24h de alimentação ad libitum, durante 20 dias. Foi avaliada a locomoção espontânea e o comportamento do tipo ansioso. Preparações de sinaptossomas foram utilizadas para investigar o metabolismo oxidativo mitocondrial através de respirometria de alta-resolução. Os resultados foram calculados e expressos como a média ± S.E.M., e as diferenças entre os grupos, usamos o teste t de Student, ou ANOVA de duas vias com post-hoc de Bonferroni. Todos os procedimentos foram realizados com GraphPad Prism 6.0. As diferenças foram consideradas estatisticamente significativas quando p<0.05. CEUA-UFRGS 22436. Resultados: O JI induziu aumento no consumo alimentar diário total nos dias de acesso ao alimento (p=0.003). Após a intervenção, não foram encontradas diferenças nos níveis sanguíneos de glicose (p=0.342) ou cetonas (p=0.539). O JI não alterou a atividade locomotora e exploratória (p=0.988) e diminuiu o comportamento ansioso (p=0.047).  Não foram encontradas diferenças no consumo de oxigênio mitocondrial, na eficiência de fosforilação oxidativa (p=0.545) e na capacidade respiratória de reserva (p=0.197). Conclusões: O JI promove alterações no padrão alimentar, além de exercer um efeito ansiolítico não relacionado com alterações na atividade exploratória espontânea. Além disso, o efeito ansiolítico do JI foi independente de adaptações bioenergéticas nos terminais sinápticos. Este projeto está em desenvolvimento, assim, a avaliação do imunoconteúdo de proteínas mitocondriais associadas ao comportamento ansioso como Bcl2, poderão auxiliar a elucidar os mecanismos envolvidos no efeito ansiolítico do JI.
