Members of the genus Dechloromonas are often abundant in EBPR plants worldwide and have long been considered putative PAOs, as intracellular poly-P has been identified with traditional staining methods. This study applied metagenomics to determine the metabolic potential and Raman microspectroscopy to verify and define the levels and dynamics of important storage polymers in FISH-defined Dechloromonas cells. A 99.2% complete metagenome-assembled genome, retrieved from Danish EBPR plants, showed the potential for poly-P, glycogen, and PHA accumulation. Dechloromonas cells exhibited in situ a Ca. Accumulibacter-like phenotype with dynamic levels of poly-P and PHA during feast-famine cycling. Quantitative Raman-FISH analysis of single cells from full-scale activated sludge plants confirmed Dechloromonas to be an important PAO
