Many studies point out that FTIR microspectroscopy can detect different spectral patterns between healthy and tumoral cells, but there are not vibrational studies carried out on living tumoral cells submitted to chemotherapy treatment.[2] At this regard, in order to obtain a detailed characterization at a molecular level of the biochemical processes induced by these toxic treatments on living cancer cells, we have exploited for the first time FTIR microspectroscopy on living tumoral oral cavity cells (OSCC_G3) treated with cisplatin and 5-fluorouracil, at different exposition times. By using a biocompatible IR-transparent microfluidic device, the spectral data of living tumoral cells in physiological solution have been acquired. By a preliminary analysis of the huge amount of spectral data, some modifications have been detected in the spectral profile, above all in the 1300-900 cm-1 IR region. According to cisplatin treatment, new bands at 1205, 1060, 1020 and 935 cm-1 have been detected at 48 hs, confirming some damages to DNA together with the increase of Z-DNA. In addition, a minor cellular activity is showed by the decrease of the band at 914 cm-1 (Fig. 1a). According to 5-fluorouracil treatment, the arise of the band at 1154 cm-1 could be related to an increase of hydrogen bond and, as further extent, to changes in DNA conformation (Fig. 1b).
