Uji Efektifitas Minyak Zaitun Sebagai Induser Dan MnSO4 Untuk Pembuatan Enzim Lipase Dari Aspergillus Niger Pada Substrat Ampas Kelapa

Abstract

Lipase is an enzyme that can catalyze the hydrolysis of triglycerol (oil or fat) into fatty acids and glycerol and the synthesis of esters. Lipase is one of the enzymes used on a large scale in the industry. Some of the industrial uses of lipase enzymes are in the processing of pharmaceuticals, detergents, cosmetics, the production of biodegradable polymers, and textiles. The use of enzymes in Indonesia reached 2,500 tons/year, and 99% was still imported, spending 187.5 billion/year. Lipase is one of the most widely used enzymes besides protease and xylanase. It shows that the need for enzymes in Indonesia is substantial. Therefore, efforts are needed to produce domestic enzymes to reduce the value of imported enzymes. This study aims to determine the optimum conditions for producing lipase enzyme activity using materials and media with relatively low prices and easy to obtain. The result of the study found that the addition of 8 mL of olive oil resulted in the highest lipase activity of 1 U/mL; too much olive oil would inhibit the growth of fungi. Meanwhile, the addition of 2 mmol MnSO4 produced the highest lipase activity, which was 0.867 U/mL
