101  PROGRESSÃO DA HIPERTENSÃO ARTERIAL PULMONAR INDUZIDA POR MONOCROTALINA: EFEITOS NO VD E NO VE Alexsandra Zimmer1, Rayane B. Teixeira1, Alan C.Bahr1, Patrick Türck1, Alexandre L. Carraro1, Guilherme Baldo1, Fernanda Visioli3,  Adriane Belló-Klein1 1Universidade Federal do Rio Grande do Sul 2 Faculdade de Odontologia, Universidade Federal do Rio Grande do Sul E-mail: zimmeralexsandra@gmail.com Introdução: Monocrotalina (MCT) é o modelo mais utilizado para estudar hipertensão arterial pulmonar (HAP), induzindo alterações na vasculatura pulmonar que levam à sobrecarga do ventrículo direito (VD), hipertrofia e insuficiência. Existe uma relação de interdependência ventricular, uma vez que os ventrículos compartilham o septo e competem por espaço dentro do pericárdio. Objetivo: analisar efeitos da progressão da HAP experimental no VD e VE. Materiais e métodos: Foram utilizados ratos machos Wistar, divididos em 4 grupos controles (C1s, C2s, C3s e C4s) e 4 grupos MCT (MCT1s, MCT2s, MCT3s e MCT4s). Animais receberam salina ou MCT (60 mg/kgi.p.) e, a cada sete dias, foram submetidos a análises ecocardiográficas e hemodinâmicas, seguidas de eutanásia para coleta de coração e pulmão. Lâminas de VD e VE foram obtidas para avaliar inflamação, área do cardiomiócito e colágeno. Resultados. Hipertrofia do VD foi encontrada a partir da segunda semana, nos animais MCT, com congestão pulmonar no grupo MCT3s. Redução da função diastólica e sistólica foi encontrada no VD após três semanas de MCT, sem alteração no VE. Houve aumento progressivo na pressão sistólica do VD a partir da primeira semana e da pressão média da artéria pulmonar a partir da segunda semana, enquanto que a pressão diastólica aumentou na terceira semana. Nenhuma alteração de pressão foi encontrada no VE. Foi encontrado aumento e diminuição da área do cardiomiócito nos animais MCT3s no VD e no VE, respectivamente. A inflamação ocorreu de modo leve, moderado e acentuado na primeira, segunda e terceira semana, respectivamente no VD, enquanto que no VE apareceu de forma leve nos animais MCT3S. Colágeno intersticial aumentou no VD e VE nos animais MCT3s. Conclusão: Os resultados permitem concluir que o modelo promove um remodelamento progressivo do VD, que culmina em alterações no VE em estágios avançados da doença. There is a relationship of ventricular interdependence, since the ventricles share the septum and compete for space within the pericardium. Objective: To analyze the effects of the progression of experimental PAH in the RV and LV. Materials and methods: Male Wistar rats, divided into 4 control groups (C1w, C2w, C3w and C4w) and 4 MCT groups (MCT1w, MCT2w, MCT3w and MCT4w) were used. Animals received saline or MCT (60 mg / kg i.p.). Every seven days, underwent echocardiographic and hemodynamic analysis, followed by euthanasia for tissue collection. RV and LV lamina were obtained to evaluate inflammation, cardiomyocyte area and collagen. Results: RV hypertrophy was found from the second week in MCT animals, with pulmonary congestion in the MCT3w group. Reduction of diastolic and systolic function was found in the RV after three weeks of MCT, with no change in the LV. There was a progressive increase in the RV systolic pressure from the first week and the mean pulmonary artery pressure from the second week, while the diastolic pressure increased in the third week. No change in pressure was found in the LV. It was found increase and decrease of the cardiomyocyte area in the MCT3w animals in the RV and in the LV, respectively. Inflammation occurred mildly, moderately and markedly in the first, second and third week, respectively in the RV, whereas in the LV it appeared mildly in the MCT3w animals. Collagen increased in RV and LV in MCT3w animals. Conclusion: The results allow concluding that the model promotes a progressive remodeling of the RV, which culminates in alterations in the LV in advanced stages of the disease.
