The newcompounds have been demonstrated to release the iron overloaddrug deferiprone on treatment with PBS and have also been shownto have antibacterial activity against H. pylori.Coordination network structures have attracted considerableattention in recent years.1,2 Although much of this interest hasarisen from the capacity of this type of material to store gaseswithin the pores, there has been an increasing interest in theinclusion of larger, more complex molecules including thosewith biological activity. Thus, metal–organic frameworks(MOFs) capable of storing drug molecules within pores anddelivering these to an organism have been developed.3 Analternative strategy makes use of the active ingredient as aligand bound to the metal within the framework.4–8 In thiscase, there is no requirement for the framework to be porous,and conceptually, a plasma-stable MOF could be fine-tunedfor oral delivery, as a suspension, releasing the bioactive cargounder conditions (e.g. pH) which digest the framework struc-ture into its constituent parts.Recently, we reported a series of zinc-based MOFs9 contain-ing deferiprone,10 an iron overload drug, as an anionic chelat-ing ligand. Immediate decomposition, and deferipronerelease, was eﬀected under mild conditions including in PBS(phosphate buﬀered saline) or dilute hydrochloric acid.Bismuth compounds such as bismuth subsalicylate (BSS)and ranitidine bismuth citrate (RBC) are generally poorlysoluble and structurally ill-defined, but despite this, they areeﬀective, orally-delivered drugs.11–15 When combined withantibiotics, (RBC also comprises a H2-antagonist, ranitidine),such drugs are extremely eﬀective against Helicobacter pylori,which is implicated in peptic ulcers, gastritis and gastriccancers.16 Studies have elucidated the role of bismuth in interalia disrupting enzymes such as urease (aﬀecting nickelhomeostasis and acid acclimation), mimicking iron depri-vation and inhibiting cellular ATP synthesis.17 Antibacterialbismuth compounds are now finding biomedical uses such asin dental implants and haemodialysis catheters.18,19The above uses and structural complexity of bismuth-basedmaterials has prompted us to extend our studies to includebismuth complexes as model compounds for potential drugdelivery and biomedical applications. It has been previouslydemonstrated that bismuth dicarboxylates can form coordi-nation network structures.20–22 We therefore reasoned that itmight be possible to include deferiprone into such structures,with the resultant compounds having a potential dual actionmode. To this end we have investigated the formation andstructures of bismuth–dicarboxylate–deferiprone complexes,then looked at their stability in terms of acting as deferipronesources and also their activity against H. pylori.The product from the reaction between bismuth(III) nitratepentahydrate, 1,4-benzenedicarboxylic acid (H2bdc) and deferi-prone (Hdfp) depends crucially on the Bi(NO3)3·5H2O : Hdfpratio employed, with a mixture of DMF and water as solventgiving the best crystals. When the reaction is carried out withthe Bi(NO3)3·5H2O : Hdfp ratio 1 : 1, the product consists ofmore than one phase, though pale yellow single crystals of[Bi2(bdc)2(dfp)2]·DMF 1 can readily be manually separatedfrom grey/cream powders. When the reaction is carried outwith the Bi(NO3)3·5H2O : Hdfp ratio 1 : 4, the reaction yieldssolely pale yellow crystals of [Bi2(bdc)(dfp)4(H2O)2]·2DMF 2.Both 1 and 2 were characterised by X-ray crystallography,with powder X-ray diﬀraction confirming phase purity and1H NMR spectroscopy of the acid-digested samples confirmingthat the bdc : dfp ratio in the bulk is the same as that in thecrystals.†Electronic supplementary information (ESI) available: Syntheses, crystallo-graphic studies, deferiprone release and antibacterial studies. In both cases, the reactions with a 1 : 1 ratio ofBi(NO3)3·5H2O : Hdfp gave mixed phases without singlecrystals analogous to those of 1, but reactions with a 1 : 4 ratioof Bi(NO3)3·5H2O : Hdfp gave [Bi2(bdc-NH2)(dfp)4(H2O)2]·2DMF3 and [Bi2{bdc-(OH)2}(dfp)4(H2O)2]·2DMF 4, both of which areisostructural to 2.The asymmetric unit of 1 consists of a bismuth centre, adfp ligand and a bdc ligand, with residual electron densityapproximating to half of a molecule of DMF per bismuthcentre. This gives rise to a formula for 1 of [Bi(bdc)-(dfp)]·0.5DMF. The bismuth centre forms six Bi–O contactsless than 2.65 Å, with coordination to a bidentate dfp ligandand two bidentate carboxylates. Three longer Bi–O contacts,between 2.85 Å and 2.95 Å, connect the bismuth centres intoBi-dfp-carboxylate chains (Fig. 1a). The dfp and carboxylategroups therefore adopt μ–κ1,κ2 coordination modes, andoverall the bismuth centre is 9-coordinate. The Bi-dfp-carboxy-late chains are linked together by the benzene rings of the bdcligands to form sheets (Fig. 1b). The dfp ligands projectapproximately perpendicular to these sheets, and those onneighbouring sheets interdigitate, though they are suﬃcientlyoﬀset to ensure there is no significant π-stacking. The dfpligand atoms, oxygens excepted, are evenly disordered over twosites.Compounds 2, 3 and 4 are isostructural, so only the struc-ture of 2 will be described in detail. The asymmetric unit of 2is composed of one bismuth centre, two dfp ligands, half of abdc ligand, one included water molecule and one includedDMF molecule. A centre of symmetry generates the remainderof the bdc ligand. The bismuth centre is coordinated to twobidentate dfp ligands and a bidentate carboxylate group,forming six Bi–O bond distances of less than 2.75 Å.Additional longer Bi–O contacts are present, with a bond to acarboxylate [Bi(1)–O(2)′ 3.046(6) Å] linking the bismuth centresinto carboxylate-bridged dimers and a bond to a water mole-cule [Bi(1)–O(7) 3.159(7) Å] meaning that the bismuth centresare best described as eight-coordinate (Fig. 2a). Overall, theformula for 2 is [Bi2(dfp)4(bdc)(H2O)2]·2DMF.The bdc ligands bridge between the Bi2(dfp)4(O2CR)2(H2O)2units, linking these into chains running along the crystallo-graphic a axis, as shown in Fig. 2b. There is also oﬀset π-stack-ing within the structure, with dfp⋯bdc⋯dfp interactionspresent between the ligands within each chain [3.3 Å]and dfp⋯dfp interactions present between the ligands inneighbouring chains [3.2 Å], serving to connect the chains intosheets. Hydrogen bonding is observed with the included watermolecule acting as a hydrogen bond donor to both a dfpligand and the included DMF molecule.Previous studies on the use of zinc-based MOFs as a poten-tial drug release platform for deferiprone showed immediaterelease in phosphate buﬀered saline (PBS) and HCl-PBS buﬀer(pH 5.5) as monitored by LCMS. This article is licensed under a Creative Commons Attribution 3.0 Unported Licence.View Article Onlineselected for analysis, and their stability studies were under-taken using 1H NMR spectroscopy and LCMS.[Bi2(bdc)(dfp)4(H2O)2]·2DMF 2 showed fast release of deferi-prone in D2O in NMR studies, which was mirrored in LCMSstudies in PBS, over a period of 5 h (Fig. 3, also see ESI†). Inter-estingly, the deferiprone was released so rapidly in eachrespective medium that the 1H NMR spectrum and LCMS tracetaken immediately after addition already showed significantrelease. Identical studies were undertaken with [Bi2{bdc-(OH)2}-(dfp)4(H2O)2]·2DMF 4, and these showed no detectable releaseof deferiprone from D2O over a period of 24 h, and signifi-cantly slower release than 2 in PBS as confirmed by LCMS(Fig. 3). To investigate the release of Hdfp from 4 using aciddigestion, the LCMS trace was initially taken in PBS, showinglittle release, followed by HCl-PBS (pH 2) where substantialrelease was observed.Compounds 2 and 4 were also tested against H. pylori(strain SS1) and compared with a positive control, bismuthsubsalicylate (BSS). Diﬀerent amounts of the compounds (0, 1,2, 4, 8, 10 20 μg ml−1) were used to test their bacteriostatic andbactericidal activities. This was determined by spreading alawn of an appropriately diluted suspension (post-exposure tothe compounds for 24 h in brain heart infusion broth sup-plemented with foetal calf serum (10%, v/v) under microaero-bic conditions) of the bacteria on Columbia blood agar platesfollowed by colony counting. Fig. 4 shows just the opticaldensity readings (600 nm) of the bacterial suspensions 24 hpost-treatment; as reduced readings relative to the controlcould represent either bactericidal or bacteriostatic activity theminimum inhibitory concentration (MIC) was determined.Both 2 and 4 showed MIC values of less than 10 μg ml−1. Incomparison, BSS showed a MIC value of less than 4.0 μg ml−1.This indicates that 2 and 4 have appreciable H. pylori inhibi-tory action, comparable to that of BSS.ConclusionsWe have demonstrated that deferiprone can be incorporated asa terminal bidentate ligand into one- and two-dimensionalbismuth coordination networks, and that it can be released ontreatment with acid or PBS. Notably, the substituents on the1,4-benzenedicarboxylate ligands play an important role in therate of deferiprone release, with 2,5-dihydroxy substituents sig-nificantly retarding this. The reasons for this are currentlyunder investigation. The coordination networks also showappreciable antibacterial activity, as demonstrated by inhi-bition of H. pylori growth. This potential dual mode of action,with both the ligand and metal centre playing diﬀerent bio-logical roles, has broad significance for other systems. Currentwork is focussing on removing the requirement for DMF in thesyntheses and further retarding the rate of dissolution of thecompounds in PBS as we move from the current modelsystems into compounds that are more suitable for in vivostudies.The University of Bath, the University of Sussex, theR.
