The estimation of the applicability of PCR-RFLP analysis method for detection of haplotypes among polyphagous species of aphids (on the example of Aphis craccivora Koch, Aphis gossypii Glover, Myzus persicae (Sulzer))

Abstract

The 636 nucleotide sequences of the COI gene from A. craccivora, A. gossypii, and M. persicae were analyzed in the work. As a result of the analysis, a high level of intraspecific genetic differences in the analyzed aphid species was revealed. On the basis of the obtained data the restriction maps were constructed and the PCR-RFLP keys were created to identify some haplotypes of COI genes among the polyphagous species of aphids A. craccivora, A. gossypii, and M. persicae.В работе было проанализировано 636 нуклеотидных последовательностей гена COI тлей A. craccivora, А. gossypii и M. persicae. В результате анализа выявлен высокий уровень внутривидовых генетических различий у анализируемых видов тлей. На основе полученных данных были построены рестрикционные карты и созданы ПЦР-ПДРФ ключи, позволяющие выявлять конкретные гаплотипы среди многоядных видов тлей A. craccivora, А. gossypii и M. persicae
