Abstract
U ovom diplomskom radu prikazan je postupak optimizacije raspršenja matice sedimenta kroz čvrstu fazu (MSPD) trinaest farmaceutika (azitromicin, tiamulin, imatinib, febantel, torasemid, omeprazol, linezolid, prazikvantel, etodolak, sulfametazin, sulfafurazol, albendazol i levamisol) koji zbog svojih različitih karakteristika pripadaju različitim skupinama lijekova. Ekstrakti dobiveni MSPD metodom snimani su na UHPLC uređaju. U svrhu određivanja optimalnih uvjeta korištena su različita otapala (čista otapala kao i njihove smjese) te smjese otapala s kiselinama. Utvrđeno je da je za optimalne uvjete ekstrakcije farmaceutika raspršenjem matice sedimenta kroz čvrstu fazu odabrana kombinacija otapala metanol:aceton=1:1, volumena 5 mL na sorbensu C18. Optimirana i validirana metoda uspješno je primjenjena na realnim uzorcima sedimenta što ukazuje na efikasnost ove metode za identifikaciju i kvantifikaciju farmaceutika iz različitih uzoraka sedimenta.This thesis shows the process optimization of matrix-solid phase dispersion of thirteen farmaceuticals (azithromycin, tiamulin, imatinib, febantel, torasemide, omeprazole, linezolid, praziquantel, etodolac, sulphamethazine, sulfafurazole, albendazole and levamisole) which has mutally different properties and belong to different groups of farmaceuticals. The obtained MSPD extracts were analyzed by UHPLC. To determine the optimal conditions, different solvents (pure solvent and various mixture of solvents) and mixture of solvents and acids were used. It was achieved that optimal conditions for extraction of farmaceuticals by MSPD is methanol:acetone 1:1 as elution solvent, volume of elution solvent is 5 mL and C18 sorbent. Optimized and validated method was successfully applied to real sediment samples suggestings efficiency of method for the identification and quantification of farmaceuticals from different sediment samples
