Wyniki badań ostatnich lat wskazują na to, że leptyna (LP) - hormon
wytwarzany głównie przez komórki tkanki tłuszczowej białej,
może wpływać na tkankę kostną. Przyjmuje się, że we wczesnym
okresie życia LP może bezpośrednio stymulować wzrost kości na
długość i grubość poprzez angiogeniczny i chondro-osteogeniczny
mechanizm. Natomiast w dojrzałym szkielecie LP może pośrednio
hamować tworzenie tkanki kostnej (działając na podwzgórze za pośrednictwem
układu nerwowego współczulnego) lub bezpośrednio
pobudzać kościotworzenie i/lub hamować resorpcję kości za pośrednictwem
systemu RANKL/RANK/OPG, zasadniczego szlaku sygnałowego,
przez który osteoblasty regulują wielkość puli aktywnych osteoklastów,
a tym samym resorpcję kości. Dominuje pogląd, że LP,
przez hamowanie ekspresji RANKL i/lub pobudzenie ekspresji OPG
w osteoblastach i komórkach zrębu szpiku, może powodować przesunięcie,
na korzyść OPG, stosunku OPG do RANKL w mikrośrodowisku
szpiku kostnego i kości, co prowadzi do zmniejszenia puli aktywnych
osteoklastów i w efekcie do ograniczenia resorpcji kości.
Aktualny rezultat działania LP jest najprawdopodobniej wypadkową
ośrodkowego (niekorzystnego) i obwodowego (korzystnego) oddziaływania
LP na tkankę kostną. Korzystny, obwodowy efekt działania
LP na tkankę kostną przeważa wówczas, gdy występuje ośrodkowa
oporność na LP, co stwierdza się w przypadku otyłości.Recent studies indicates that leptin (LP) - a hormone produced

of bone formation and/or to suppression bone resorption.
This last effect is most likely achieved through RANKL/RANK/OPG,
an essential signaling pathway through which osteoblasts regulating
rate of activated osteoclasts pool by the same token bone resorption.
Dominates the view that LP through suppression of RANKL
and/or stimulation of OPG in osteoblasts and marrow stroma cells
may to cause shift to the advantage of OPG relation OPG to RANKL
in microenvironment of bone marrow and bone. It leads to decrease
of activated osteoclasts pool and consequently to bone

resultant of central (negative) and peripheral (positive) influence
