Functional analysis of the N-terminal domain of SPA1AbstractPlants  adapt  to  their  light  environment  by  regulating  several  key  developmental  processes  likeseedling  de-etiolation  and  the  transition  from  vegetative  to  reproductive  growth.  Arabidopsisthaliana is a facultative long day flowering plant. It has evolved mechanisms to time its floweringaccording to seasonal and environmental cues. There are several signaling pathways for floweringtime control that can be distinguished by the environmental or developmental cues they transduce.One important pathway, the photoperiodic pathway, allows plants to time flowering according toseasonal  changes.  The  current  understanding  is  that  plants  measure  day  length  through  theintegration  of  internal  signals  from  the  circadian  clock  and  external  light  cues.  The  centralintegrator of these signals CONSTANS (CO) is controlled at the transcriptional level by the plantcircadian clock and at the post-translational level by light through protein degradation in darkness.CO is targeted for degradation in darkness by the CONSTITUTIVE PHOTOMORPHOGENIC 1/SUPRESSOR OF PHYA-105 (COP1/SPA) E3 ubiquitin ligase complex. SPA1 is a member of asmall  gene  family  comprising  four  members.  Of  the  four  SPAs,  SPA1 has  the  most  importantfunction in flowering time control. SPA1, like COP1, contains a WD repeat domain and a coiled-coil domain. Unlike COP1 it contains a kinase-like domain. Complementation assays have shownthat the N-terminal kinase-like domain of SPA1 is essential for its function in flowering time controlbut not for seedling de-etiolation. To further investigate the function of the N-terminal domain ofSPA1, single amino acid exchanges in residues conserved in kinases were performed and used forcomplementation assays. These assays have revealed that some of these amino-acids are essentialfor SPA1 function in flowering time control and implicated in seedling de-etiolation specifically inred (R) light.The data presented here indicate that either the putative kinase function or the kinase-like structureof  SPA1 is  essential  for  its  function  in  photoperiodic  flowering  time  control  and  the  physicalinteraction of SPA1 with CO and COP1. Furthermore the SPA1 N-terminal domain is essential forseedling photomorphogenesis specifically in R light suggesting a function of that domain in a phyB-specific signaling pathway.ZusammenfassungPflanzen  passen  sich  den  Licht  Bedingungen  ihrer  Umwelt  an  indem  sie  verschiedeneEntwicklungsprozesse so wie Keimlings  De-Etiolierung und den Übergang von vegetativem zureproduktivem  Wachstum.  Arabidopsis  thaliana ist  eine  fakultative  lang  Tag  Pflanze.  Sie  hatMechanismen entwickelt um ihren Blühzeitpunkt der Jahreszeit und der Umwelt entsprechend zuregulieren. Es gibt mehrere Signalwege die den Blühzeitpunkt regulieren, diese unterscheiden sichnach den Umwelt- und Entwicklungssignale die sie transduzieren. Ein wichtiger Signalweg, derphotoperiodische  Signalweg,  ermöglicht  es  der  Pflanze  ihren  Blühzeitpunkt  jahreszeitbedingtenÄnderungen anzupassen. Pflanzen messen die Tageslänge indem sie interne Signale der ZirkadianenUhr und externe Lichtsignale integrieren. Der zentrale Integrator dieser Signale CONSTANS (CO)ist auf transkriptionaler Ebene von der Zirkadianen Uhr kontrolliert und auf post-transkriptionalerEbene von Licht durch Proteinabbau im Dunkeln.CO wird von dem CONSTITUTIVE PHOTOMORPHOGENESIS 1/ SUPPRESSOR OF PHYA-105 (COP1/SPA) E3 Ubiquitin Ligase Komplex zum Abbau im Dunklen bestimmt. SPA1 ist Teileiner kleinen Genfamilie die aus vier Genen besteht. Von den vier SPAs hat SPA1 die WichtigsteFunktion  in  der  Regulierung  des  Blühzeitpunktes.  SPA1,  wie  COP1,  besitzt  eine  WD-repeatDomäne  und  eine  coiled-coil  Domäne.  Anders  als  COP1  besitzt  es  eine  kinase-like  Domäne.Komplementationsanalysen haben gezeigt dass die SPA1 N-terminale Domäne essenziell für dieFunktion in der Regulierung des Blühzeitpunktes ist aber nicht für De-Etiolierung des Keimlings.Um  die  Funktion  der  SPA1  N-terminalen  Domäne  weiter  zu  untersuchen  wurden  einzelneAminosäuren  die  in  Kinasen  konserviert  sind  ausgetauscht  und  für  Komplementationsanalysenverwendet. Diese Analysen haben gezeigt dass manche dieser Aminosäuren essenziell sind für dieSPA1 Funktion in der Regulierung des Blühzeitpunktes und für die De-Etiolierung des Keimlingsspezifisch im rot (R) Licht.Die hier präsentierten Daten deuten an dass entweder die putative Kinase Funktion oder die Strukturder SPA1 kinase-like Domäne essenziell ist für die Funktion in der photoperiodischen Regulierungdes Blühzeitpunktes und der physischen Interaktion von SPA1 mit CO und COP1. Zudem ist dieSPA1 N-terminale Domäne essenziell für die De-Etiolierung des Keimlings spezifisch im R Lichtwas auf eine Funktion dieser Domäne in einem phyB-spezifischen Signalweg deutet.
