Het proefschrift van Henriëtte Rozeboom bespreekt kristalstructuren van zes verschillende enzymen. Deze zes enzymen behoren tot verschillende eiwitfamilies en maken gebruik van verschillende katalytische mechanismes. Begrip van hun 3D-structuren is essentieel voor het begrijpen van de moleculaire basis van hun functioneren en voor inzicht in hun evolutionaire ontstaansgeschiedenis. Het proefschrift bevat vijf delen: het eerste deel beschrijft de structuur en mutationele karakterisering van de katalytische A-module van het mannuronan C-5-epimerase AlgE4 uit Azotobacter vinelandii, het tweede deel bespreekt de kristalstruktuur van α-1,4-glucan lyase, een uniek glycoside hydrolase familielid met een ongebruikelijk katalytisch mechanisme. Het derde deel beschrijft de kristalstruktuur van een quinone-afhankelijk alcohol dehydrogenase uit Pseudogluconobacter saccharoketogenes, een veelzijdige dehydrogenase dat alcoholen en koolhydraten oxideert. Het vierde deel bespreekt de kristalstruktuur van de endo-xylogalacturonan hydrolase uit Aspergillus tubingensis, een enzym dat specifiek xylogalacturonan afbreekt. Tot slot worden de kristal strukturen van twee Bacillus carboxylesterases, naproxen esterase en CesB, met verschillende enantio-selectiviteit beschreven.Henriëtte Rozeboom (Appingedam, 1964) studeerde aan de Rijks Hogeschool Groningen. Het onderzoek werd uitgevoerd in de afdeling eiwitkristallografie van het Groningen Biomolecular Sciences and Biotechnology Institute (GBB).The thesis of Henriëtte Rozeboom discusses the crystal structures of six different enzymes.
