However，　the　conditions　needed　to
sus ain　the　specific　autologous　tumor
responses　in　long－term　eultures　of　human
T工L　remain　to　be　determined．

Tu皿or－infiltrating　lymphocytes　（T工：L）were
isolated　by　enzynatic　digestion　and　gra－
dient　centrifugation　from　18　hurnan　ovari－
an　careinomas．　These　cells　were　cultured
in　a　complete　medium　supplemented　with
recombinant　interleukin　2　（工：L2）　a：lone　or
recombinant　IL2　plus　recombinant　tumor
necrosis　factor　a（TNF－a），　and　their
growth　and　antitumor　eytotoxicity　were
determined．　T工：L　cultured　in　the　pres－
ence　of　工1、2　plus　TNF一α（1000　units／m］．
each）　for　6　days　showed　significantly
higher　cytotoxicity　against　fresh　auto－
logous　tumor　targets　than　did　T工：L　cu：L－
tured　with　IL2　alone　（e．g．，　mean　lytic
units／lO7　cells　for　8　T工：L　preparations
were　290　versus　74；　p〈O．05）．　No　differ一一
ences　in　［3H］thymidihe　uptake　or　natural
killer　eell　activity　were　observed　among
these　T工L　cultures．　工n　titration　exper－
iments，　optimal　synergistic　concentra－
tions　of　工：L2　and　TNF一こ口ere　determined　as
lo2　and　lo3　units／ml，　respectively．
Using　these　coneentrations　for　culturing
the　TIL，　effector　cells　developed　which

positive）．　However，　the　autologous　tumor
cytotoxieity　mediated　by　these　cultured
T工：L　on　day　6　was　short　lived．　　By　day　l2，
it　was　replaced　by　non－major　histocompat－
ibility　complex－restricted，　lymphokine－
actived　kilter　cell－like　activity
mediated　by　CD3－CD56十　effeetor　cells．
Simultaneously，　the　production　of
y－interferon　and　interleukin　1　decreased
in　these　cultures．　ln　contrast　to　［［INF－a，
anti－CD3　antibody　synergized　with　工1、2　to
increase　2－3－fold　T工1・proliferation　l）ut
not　their　cytotoxic　activity　against
autologous　tumor　eell　targets．　These
data　suggest　that　TNF一α　and　工：L2　synergize
early　in　culture　to　induce　tumor－reactive
CD8十　effectors，　some　of　which　may　be　spe－
cific　for　autologous　ovarian　tumor　cells．
