Sep. 2021Microbellen zijn kleine gasbelletjes (~2.5 µm) die in eerste instantie zijn ontwikkeld als contrast middel voor de echocardiografie. Wanneer microbellen zich in een ultrageluid veld bevinden, gaan ze oscilleren. Ze zetten uit en krimpen in onder invloed van de negatieve en positieve drukverschillen van de geluidsgolf. Het zijn deze akoestische eigenschappen die hebben geleid tot de ontwikkeling van microbellen als een therapeutische modaliteit. Het blijkt namelijk dat cellen in de buurt van oscillerende of uit (in) elkaar klappende bellen een verhoogde opname van medicijnen of genetisch materiaal vertonen. Dit creëert de mogelijkheid om een zeer gerichte, lokale therapie te ontwikkelen met behulp van ultrageluid en microbellen, wat de specificiteit en effectiviteit van huidige medicatie kan verhogen. Hoewel het nu bekend is dat ultrageluid en microbellen voor een verhoogde opname van therapeutica kunnen zorgen, is het nog niet duidelijk via welke route(s) cellen deze moleculen internaliseren en welke biologische effecten worden geïnduceerd. Het onderzoek in dit proefschrift beschrijft de biologische effecten van ultrageluid en microbellen op het niveau van individuele cellen, als mede de mechanismen achter de verhoogde opname van moleculen. Microbubbles are tiny gas bubbles (~2.5 µm), originally developed as contrast agents for echocardiography. When microbubbles are exposed to ultrasound, they start oscillating. They expand and shrink according to the negative and positive pressure differences of an ultrasonic wave. These acoustic properties lead to the development of microbubbles as a therapeutic modality. As it appeared that cells surrounding oscillating or bursting microbubbles showed enhanced uptake of drugs or genes. This creates the opportunity to develop a targeted and local therapy using ultrasound and microbubbles, increasing efficacy of current medications. Although it is known that ultrasound and microbubbles cause increased uptake of therapeutics, it is still not fully understood how exactly these cells internalize the molecules, and which biological effects are induced. The research in this thesis describes the biological effects evoked by ultrasound and microbubbles at the level of individual cells, as well as the mechanisms behind increased uptake of extracellular molecules.
