Abstract
Fruktani predstavljaju lako mobilizirajuće zalihe ugljikohidrata u biljci. Stabljika ječma zbog svoje morfološke građe pohranjuje znatnu količinu fruktana koji se mobiliziraju iz stabljike u zrno poslije cvatnje što značajno utječe na ukupni prinos zrna. U ovome radu istraživani su sadržaj fruktana i aktivnost gena uključenih u sintezu i remobilizaciju fruktana u stabljici jarog ječma (Hordeum vulgare L.) sorte Astor tijekom četiri razvojne faze: klasanje, cvatnja i nalijevanje zrna. Izmjeren je sadržaj fruktana i semikvatitatvno je izmjerena ekspresija gena saharoza: saharoza 1-fruktoziltransferaza (1-SST), saharoza: fruktan-6 fruktozil-transferaza (6-SFT) i fruktan-1-egzohidrolaza (1-FEH). Najveći sadržaj fruktana izmjeren je u fazi nalijevanja zrna, a najmanji u fazi zriobe. Ekspresija gena 1-SST konstanta na je tijekom razvoja, za razliku od gena 6-SFT koji ima najveću ekspresiju tijekom cvatnje. Ekspresija gena 1-FEH povećava se u fazi klasanja što ukazuje na početak mobilizacije fruktana iz stabljike u zrno.Fructans are easily mobile carbohydrate storage in the plant. Because of its morphology barley stem stores a considerable amount of fructans that can be mobilized in the grain after flowering, which significantly affects the overall yield. In this paper, the content of fructans and the activity of genes involved in the synthesis and remobilization of fructans in the stem of spring barley (Hordeum vulgare L.) cultivar Astor during for four growth stages: heading, flowering, grain filling and ripening was studied. Fructan content was measured and expression of genes sucrose:sucrose 1-fructosyltransferase (1-SST), sucrose:fructan-6 fructosyltransferase (6-SFT) and fructan 1-exohydrolase (1-FEH) was determined by semiquantitative analyses. The highest fructan content was measured in the grain filling stage, and the lowest in the ripening. Expression of gene 1-SST is constant during the development, in contrast to the 6-SFT gene having the highest expression during flowering. Expression of gene 1-FEH increased from heading what indicated beginning of the mobilization of fructans from the stem into the grain
