Abstract
Biološki uĉinak 12 ekstrakata iz cvjetova kamilice istražen je in vitro na staniĉnim linijama HuT-78, K562, HeLa, NCI-H358 i MDCK-I. Antiproliferativne sposobnosti ekstrakata i apigenina nakon 72 sata izlaganja stanica određene su MTT testom. Morfološke promjene analizirane su svjetlosnom mikroskopijom, a apoptoza je određena Annexin-V-FITC testom. Rast stanica znaĉajno je inhibiralo 3 ekstrakta u koncentracijama 0,2 i 0,5 mg/mL rezultirajući s više od 80 % inhibicije rasta HuT-78 i K562 stanica. Najmanje osjetljiva staniĉna linija na djelovanje ekstrakata je NCI-H358 kod koje je uoĉena inhibicija rasta od 60-65 %. Najizražajnije morfološke i apoptotiĉke promjene na HeLa stanicama izaziva ekstrakt 11 (IC50 = 0,098 mg/ml). Ispitivani izolat apigenina u odnosu na komercijalno dostupan apigenin pokazuje razliĉit obrazac antiproliferativnog djelovanja u odnosu na komercijalno dostupni uzorak što upućuje da apigenin nije jedina biološki aktivna tvar u ekstraktima. U zakljuĉku, ispitivani ekstrakti znaĉajno inhibiraju rast stanica i imaju sposobnost indukcije apoptoze. Nije uoĉena selektivnost u djelovanju izmeĊu tumorskih i normalnih stanica.The biological effect of 12 extracts from chamomile flowers was investigated in vitro on cell lines HuT-78, K562, HeLa, NCI-H358 and MDCK-I. Antiproliferative abilities of extracts and apigenin after 72 hours of cell exposure were examined by MTT assay. Morphological changes were analyzed by light microscopy, and apoptosis was determined by Annexin V-FITC assay. Cell growth was significantly inhibited by 3 extracts at concentrations 0.2 and 0.5 mg/mL resulting in a more than 80 % growth inhibition HuT-78 and K562 cells. The least sensitive cell line on the effect of extracts is NCI - H358 in which is observed inhibition of growth of 60-65 %. Pronounced morphological and apoptotic changes in HeLa cells were caused by extract 11 (IC 50 = 0.098 mg/mL). Isolate of apigenin compared to commercially available apigenin shows different patterns of antiproliferative activity compared to a commercially available sample, indicating that apigenin is not the only biologically active substance in the extract. In conclusion, the tested extracts significantly inhibited the growth of cells and have the ability to induce apoptosis. There was no selectivity toward normal and tumor cells
