Summary  Nesprin-2, a member of the spectrin repeat family of proteins, exists in many different isoforms. The largest one, termed Nesprin-2 Giant, acts as a linker between the nucleus and the actin cytoskeleton as well as the microtubule network. It is composed of an N-terminal actin-binding domain followed by a large number of spectrin repeats forming a central rod like domain and a C-terminal KASH domain that anchors the protein at the nuclear envelope. Additionally, several structural maintenance of chromosomes (SMC) like domains have been predicted in the central rod domain. One of these SMC like domains (NP_055995.4, aa 1464-1771) has been reported to interact with meckelin and MKS1, indicating that Nesprin-2 plays roles in centrosome/basal body positioning during cell polarization. However, the function of the SMC domains in Nesprin-2 remains largely elusive. In general, SMC domains can bind to DNA and are known as core components of condensin and cohesin complexes, that play a role in the packaging of DNA.  In order to study the functional relevance of the SMC domain in Nesprin-2, an antibody against this domain was generated. By using this novel Nesprin-2 SMC antibody in ChIP experiments we found interactions between Nesprin-2 and chromatin. Treating cells with shRNAs specifically targeted against the SMC domain of Nesprin-2 we found that during mitosis chromosomes failed to condensate properly and DNA appeared diffuse compared to the DNA of control shRNA treated cells.  To further characterize the interaction of Nesprin-2 with chromatin structures, Co-IP and pull-down experiments were performed to identify novel interaction partners of Nesprin-2. Ku70 was identified as novel interaction partner of the SMC domain in Nesprin-2. Ku70 binds to DNA and might mediate the interaction of Nesprin-2 to chromatin. Taken together, data in this study indicate that Nesprin-2 might play roles in DNA repair and organization and, importantly, in the condensation of chromosomes during mitosis.      Zusammenfassung  Nesprin-2, ein Mitglied der Familie der Proteine mit Spektrin Wiederholungen, existiert in verschiedenen Isoformen. Die größte Isoform, Nesprin-2 Giant, fungiert als ein Bindeglied zwischen dem Zellkern und dem Aktin und Mikrotubuli Zytoskelett. Es besteht aus einer N-terminalen Aktin Bindestelle gefolgt von einer großen Anzahl an Spektrin Wiederholungen, die eine zentrale Stab Domäne bilden, und einer C-terminalen KASH Domäne, welche die Proteine mit der Zellkernhülle verknüpft. Zusätzlich weisen Vorhersagen auf das Vorhandensein mehrerer „Structural maintenance of chromosomes“ (SMC) Domänen in der zentralen Stabdomäne hin. Für eine dieser SMC ähnlichen Domänen (NP_055995.4, aa 1464-1771) wurde eine Interaktion zu MKS1 gezeigt, die nahelegt, dass Nesprin-2 in der Zentrosomen/Basalkörper Ausrichtung während der Zellpolarisierung involviert ist. Dennoch bleibt die Rolle der SMC Domäne in Nesprin-2 weitestgehend ungeklärt. SMC Proteine im Allgemeinen binden an DNA und sind Kernbestandteile von Kondensin und Kohesin Komplexen, welche eine Rolle in der Verpackung von DNA übernehmen.   Um die funktionelle Bedeutung der SMC Domäne in Nesprin-2 zu untersuchen, wurde ein Antikörper gegen diese Domäne erstellt. Durch die Verwendung dieses neuen Nesprin-2-SMC Antikörpers wurde eine Interaktion zwischen Nesprin-2 und Chromatin gefunden. Durch die Behandlung von Zellen mit shRNAs, die speziell gegen die SMC Domäne in Nesprin-2 gerichtet waren, haben wir herausgefunden, dass mitotische Chromosomen nicht ordnungsgemäß kondensieren und die DNA infolgedessen diffuser erscheint im Vergleich zur DNA in Kontrollzellen.   Zur weiteren Charakterisierung der Interaktion zwischen Nesprin-2 und Chromatinstrukturen und zur Identifizierung neuer Interaktionspartner, wurden Coimmunpräzipitationsexperimente und Pull Down Experimente durchgeführt. Hierbei wurde Ku70 als neuer Interaktionspartner identifiziert. Ku70 interagiert mit DNA, was eine indirekte Interaktion von Nesprin-2 an Chromatin vermitteln könnten. Zusammenfassend weisen die in dieser Arbeit erzielten Ergebnisse auf eine Rolle von Nesprin-2 in der DNA Reperatur, der Organisation und besonders der Kondensation von Chromosomen während der Mitose hin.
