C57BL/6موش هاي نر بالغ  مواد و روش ها:در  بعد از تائيد القاي ديابت، حيوانات  القا شد. استرپتوزوتوسين ديابت در حيوانات از طريق تغذيه با رژيم پرچرب و دوز پايين .به ترتيب به صورت گاواژ و تزريق زير پوستي درمان شدند بهمراه انسولين 3-، انسولين و امگا3-روزه با امگا 35يك دوره   .ندو كروماتين اسپرم مورد بررسي قرار گرفت DNAدرمان ساختار بيضه، پارامترهاي اسپرم و  ي بعد از اين دورهگروه شاهد  بهنسبت  كاهش قابل توجهي در حركت، حيات و مورفولوژي طبيعي اسپرم در گروههاي ديابتي ها: يافتهبه تنهايي و به همراه انسولين به طور قابل توجهي حركت و مورفولوژي اسپرم را  3-امگادرمان با  و حامل وجود داشت.درگروه ديابتي بهبود بخشيد. تعداد اسپرم در گروه ديابتي به طورغير قابل توجهي كاهش يافت و هيچ كدام از درمان ها رگروه ديابتي آسيب قابل توجه نسبت دوكروماتين اسپرم  DNAگروه هاي شاهد و حامل بازگرداند.  سطحنتوانست آن را به را به طور قابل توجهي بهبود بخشيد. اسپرم  وكرماتين  DNAو انسولين سالمت 3-درمان با امگا .ندبه بقيه گروه ها داشتبين گروههاي مختلف هيچ اختالف معني داري مشاهده نشد. ديابت نوع دو  كرومومايسين در حالي كه در نتايج آنيلين بلو وكه ساختار هاي بيضه شامل ضخامت اپيتليوم  ترون خون را كاهش داد. در حاليوسطح تستوس سلول هاي اليديگ تعداد  مانده اند.باقي ژرمينال وسطح لوله هاي اسپرم ساز بدون تغيير را   DNAكيفيت كروماتين و مثل نر مخصوصا پارامتر هاي اسپرم، عملكرد سيستم توليد 2ديابت نوع  نتيجه گيري:  .و انسولين درماني اين تغييرات ساختاري را بهبود مي بخشند 3-امگاتحت تاثير قرار مي دهد و مصرف  لين، بيضه، اسپرم، كيفيت كروماتين، انسو3-امگا، 2ديابت نوع  كلمات كليدي:Abstract Background and Objectives: The main goal of the present study was to evaluate the protective effects of omega3 and isulin on testicular structure, sperm parameters and chromatin integrity in type II diabetes-induced animals.  Material and methods: Adult male C57BL/6  mice were randomly divided into intact, vehicle and 3 diabetic (Dia) groups; diabetic animals received 8 weeks high fat diet followed by low dose streptotozocin injection. After diabetes confirmation, the animals received Omega3 (Dia+omg3 group), Insulin (Dia+Ins group) or combination (Dia+ Ins+Omg3 group). Following 35 consecutive days of treatment  the animals were sacrificed and different parameters in testis, sperm and sperm chromatin were assessed.  Results: There was a significant reduction in sperm motility, viability and normal morphology in Dia group versus intact and vehicle groups. Treatment with Omega3, alone or in combination with insulin  significantly improveed sperm motility and normal morphology compared with the Dia group. Induction of diabetes type II caused a non-significant reduction in sperm count, while none of treatments could restore it to those levels of intact  and vehicle groups.  Animal sperm in Dia group had significantly more damaged chromatin than other groups assessed by Acridin orange and Tolidine blue staining. Treatment with Omega3 and insulin,  significantly improved the sperm chromatin quality and DNA integrity. Neverthless, Chromomycin A3 and Aniline blue staining results did not differ among the groups.  Type II diabetes reduced Leydig cell number and testosterone level. However, testis structure including germinal epithelium thickness and seminiferous tubule area remained unchanged  among the different groups. Conclusion: We might conclude that type II diabetes interfere with the male reproductive function especially sperm quality and DNA integrity. These structural changes are reversed to some extent, by long term Omega3 and insulin replacement therapy.
