Abstract
Istraživači su nedavno razvili alat REPAIR koji se sastoji od spojene katalitičke domene
deaminaze ADAR i katalitički mrtvog Cas13b, iz obitelji proteina Cas13. Tako napravljen
enzim ima mogućnost specifičnog uređivanja molekule RNA od interesa u ljudskim stanicama.
Enzim ADAR, adenozin deaminaza djelujuća na RNA, pretvara adenozin u inozin - nukleotid
koji je funkcionalni ekvivalent gvanozinu. Alat je uspješno korišten za revertiranje dviju
mutacija G-u-A povezanih nastankom bolesti na mRNA – jedna mutacija uzrokuje X-vezani
nefrogeni dijabetes, a druga uzrokuje Fanconi anemiju.Researchers have created a fusion protein between the catalytic domain of deaminase
ADAR and a catalytically inert Cas13b, member of Cas13 protein family, with the ability to
target any RNA of interest in human cells creating REPAIR system. Enzyme ADAR, which is
adenosine deaminases acting on RNA converts adenosine into inosine - a nucleotide that is
functionally equivalent to guanosin. They used it to revert two disease-associated G-to-A
mutations, one that causes a form of diabetes, and another that causes Fanconi anemia, in two
separate messenger RNAs
