Zusammenfassung Um Transmembranproteine, die mit Ubiquitin modifiziert wurden, von der endosomalen Membran in intraluminale Vesikel zu sortieren, wird die „Endosomal Sorting Complex Required for Transport“ (ESCRT)-Maschinerie benötigt (Ren and Hurley, 2010; Ziv et al., 2011; Katzmann et al., 2001). In Hefen und Tieren besteht sie aus vier Komplexen, die ESCRT-0 – III genannt werden (Wollert and Hurley, 2010). Die meisten der ESCRT-Komponenten sind in Pflanzen konserviert. Für die ESCRT-I-Komponente MVB12 und den gesamten ESCRT-0-Komplex konnten bislang keine homologen Sequenzen gefunden werden (Spitzer et al., 2006; Winter and Hauser, 2006; Leung et al., 2008).  In Arabidopsis thaliana wird  FYVE2 als Kandidat für die Funktion der ESCRT-0-Komponente Vps27 beschrieben (Shahriari, 2008).   Im Rahmen dieser Arbeit konnte ich bestätigen, dass FYVE2 mit der ESCRT-I-Komponente ELCH interagiert. Die Interaktion findet dabei über ein konserviertes PSAP-Motiv im N-Terminus von FYVE2 statt. Zudem konnte ich zeigen, dass FYVE2 über seine C-terminale DUF500-Domäne an Endosomen rekrutiert wird. Um den Einfluss von FYVE2 auf die Sortierung von monoubiquityliertem Membranprotein untersuchen zu können, wurde das Marker-Set PMA-EGFP/PMA-EGFP-Ub für die ubiquitinabhängige Sortierung von Membranproteinen entwickelt. Ich konnte zeigen, dass die translationale Fusion mit Monoubiquitin ausreichend ist, um in Pflanzen ein Membranprotein in das Lumen der Vakuole zu transportieren. Die Sortierung von PMA-EGFP-Ub erfolgt über den sogenannten „Hydrophobic Patch“ des Ubiquitins und kann durch Koexpression mit einer dominant-negativen Version der ESCRT-III assoziierten-ATPase AtSKD1 blockiert werden. Durch die Entwicklung zweier weiterer Marker-Sets (Lti6a-EGFP/Lti6a-EGFP-Ub und VRS1-EGFP/VRS1-EGFP-Ub) konnte zudem gezeigt werden, dass das translationale Anfügen eines Monoubiquitins prinzipiell zur Sortierung eines Membranproteins in das Lumen der Vakuole führt.  Durch Koexpression des Sortierungsmarkers PMA-EGFP-Ub mit FYVE2 konnte ich zeigen, dass die Sortierung von PMA-EGFP-Ub in die Vakuole funktionell ist, aber durch die Überexpression von FYVE2 verlangsamt abläuft.  Eine grundlegende Vorsausetzung für die ESCRT-0-Funktion von FYVE2 ist die Abhängigkeit der Lokalisation von ELCH, und somit von ESCRT-I, von der Lokalisation von FYVE2 an der Endosomenmembran. Da FYVE2 zwar mit ELCH kolokalisiert, aber ELCH auch an Endosomen zu finden ist, an denen FYVE2 nicht lokalisiert ist, wird diese Vorraussetzung nicht erfüllt. FYVE2 kann somit nicht die Funktion eines ESCRT-0-Komplexes in Arabidopsis thaliana übernehmen.  Um die Frage nach der Funktion von FYVE2 zu beantworten,  wurde die DUF500-Domäne analysiert. Ich konnte zeigen, dass sie Aktin (in Zusammenarbeit mit Dr. Ellen G. Allwood) und Phosphatidylinositol-3-Phosphat (PI(3)P) bindet. Da FYVE2 mit verschiedenen Proteinen, die an der Entstehung und der Fusion von Vesikeln beteiligt sind, interagiert, ist die Aktin- und PI(3)P-Bindung  ein Hinweis auf eine Funktion von FYVE2 als Regulator des Vesikeltransports.       Abstract Sorting of ubiquitylated transmembrane proteins from the endosomal membrane into the intralumenal vesicles requires the coordinated action of the Endosomal  Sorting Complex required for Transport (ESCRT) machinery (Ren and Hurley, 2010; Ziv et al., 2011; Katzmann et al., 2001). In yeast and animals the ESCRT consists of four complexes, which are called ESCRT-0-III (Wollert and Hurley, 2010). Most of ESCRT components are conserved in plants but no homologue sequence could be found neither for the ESCRT-I component MVB12 nor for one of the two components of ESCRT-0  (Spitzer et al., 2006; Winter and Hauser, 2006; Leung et al., 2008). FYVE2 was proposed to act as functional homolog of the ESCRT-0 component Vps27 in Arabidopsis thaliana (Shahriari, 2008).   During my thesis I confirmed the interaction between FYVE2 and the ESCRT-I component ELCH. The interaction between these two proteins takes place at a conserved PSAP-Motif in the N-terminal part of FYVE2.  The C-terminal DUF500-Domain recruits FYVE2 to the endosomal membrane  To analyze the effect of FYVE2 on the sorting of monoubiquitylated membrane proteins, the marker set PMA-EGFP/PMA-EGFP-Ub for sorting was developed. I could show that translational fusion of a single Ubiquitin is sufficient to sort membrane proteins to the vacuolar lumen. The sorting of PMA-EGFP/PMA-EGFP-Ub requires the hydrophobic patch of Ubiquitin and can be blocked by coexpression with the dominant-negative version of the ESCRT-III associated ATPase AtSKD1. With the development and analysis of two new sorting marker-sets (Lti6a-EGFP/Lti6a-EGFP-Ub and VRS1-EGFP/VRS1-EGFP-Ub) I was able to show that the translational fusion of an Ubiquitin in principle serves as a sorting signal. The coexpression of FYVE2 with PMA-EGFP-Ub showed that overexpression of FYVE2 slows down the sorting of the marker construct into the lumen of the vacuole. One crucial requirement to proof the ESCRT-0-Function of FYVE2 is the dependency of ESCRT-I on FYVE2 to localize on endosomes. Coexpression with the ESCRT-I component ELCH showed that FYVE2 cannot fulfill this requirement, because FYVE2 colocalized with ELCH, but ELCH can although be found on endosomes that show no FYVE2-signal. FYVE2 has no ESCRT-0 function in Arabidopsis thaliana. This finding raises the question of FYVE2’s role in the cell. To answer this question, I analyzed the function of the DUF500-Domain. I could show that the Arabidopsis thaliana DUF500 binds actin (in collaboration with Dr. Ellen G. Allwood) and Phosphatidylinositol-3-Phosphate (PI(3)P). This finding, together with the fact that FYVE2 interacts with proteins needed for vesicle fission and fusion, suggests that FYVE2 acts as a regulator of vesicle transport.
