 Zusammenfassung  COMP und Matrilin-3 sind in der extrazellularen Matrix des Knorpelgewebes reichlich vorhandene Proteine. Sie fungieren als Adaptermoleküle und stabilisieren makromolekulare Netzwerke bestehend aus fibrillären Kollagenen und Proteoglykanen. Im Menschen führen Mutationen in den für COMP und Matrilin-3 kodierenden Genen zu Chondrodysplasien, während COMP- oder Matrilin-3-defiziente Mäuse keine ausgeprägten Skelettfehlbildungen aufweisen. Aufgrund ihrer ähnlichen Funktion beim Aufbau und der Stabilisierung der extrazellulären Matrix wird vermutet, dass der Verlust von COMP bzw. Matrilin-3 jeweils gegenseitig kompensiert werden kann. Um eine solche mögliche Redundanz zu untersuchen, wurden COMP/Matrilin-3 doppelt defiziente Mäuse generiert. Während in neugeborenen Mäusen keine Veränderung der Skelettentwicklung im Vergleich zum Wildtyp festgestellt werden konnte, wiesen ein Monate alte doppelt defiziente Mäuse verkürzte Röhrenknochen bei reduzierter Körpergröβe auf. Mittels peripherer quantitativer Computertomografie konnte in der Metaphyse der Femora von doppelt defizienten Mäusen eine erhöhte trabekuläre Knochendichte nachgewiesen werden. Darüber hinaus wurde neben einem verzögerten Abbau von Aggrekan im trabekulären Knochen der Metaphyse eine erhöhte Ablagerung von TIMP-3 im Knorpel der Wachstumsfuge nachgewiesen. Die Struktur und Morphologie der Wachstumsfuge war weitestgehend normal. Im Zentrum des Knochens wurde fokal ein Schlieβen der Wachstumsfuge, vergleichbar mit dem Phänotypen MMP13-defizienter Mäuse, beobachtet. Der Verlust von COMP und Matrilin-3 führt zu einer verstärkten Ablagerung von TIMP-3, die wiederum eine partielle Inaktivierung von MMPs verursachen könnte. So lieβe sich auch der mit MMP13-defizienten Mäusen vergleichbare Phänotyp erklären. Ein zweites Ziel war die Untersuchung der Prozessierung und Degradation von Matrilinen während der Knorpelumwandlung in der Wachstumsfuge des sich entwickelnden Knochens. Die beiden Aggrekanasen ADAMTS-4 und ADAMTS-5 spielen nachweislich eine Rolle bei der Entwicklung von Arthrose und sind am Umbau der extrazellulären Knorpelmatrix beteiligt. Desweiteren sind sie an der intrazellulären Prozessierung von Matrilinen involviert. Diese Prozessierung beeinflusst auch ihre Effektivität bei der Gewährleistung der Matrix-Kohäsion. Die Spaltung der Matriline resultiert in Formen mit verminderter Anzahl von Liganden-bindenden Untereinheiten. Sowohl das bei der Spaltung entstehende Neoepitop als auch ADAMTS-5 kolokalisieren in prähypertrophen und hypertrophen Chondrozyten der Wachstumsfuge von murinen Röhrenknochen, während sie in proliferierenden Chondrozyten nicht detektiert werden konnten. Die Analyse der subzellulären Verteilung ergab, dass ADAMTS-5 und die Spaltform von Matrilin-4 vorrangig in späten Golgi-Vesikeln vorliegen. In Abwesenheit von ADAMTS-5 findet keine Prozessierung von Matrilin-4 in der Wachstumsfuge statt. Dies impliziert, dass lediglich ADAMTS-5, jedoch nicht ADAMTS-4, eine physiologische Funktion in der intrazellulären Prozessierung von Matrilinen in der Wachstumsfuge einnimmt.     Abstract   Cartilage oligomeric matrix protein (COMP) and matrilin-3 are extracellular matrix proteins abundant in cartilage. As adaptor molecules, both COMP and matrilin-3 bridge and stabilize macromolecular networks consisting of fibrillar collagens and proteoglycans. Mutations in the genes coding for COMP and matrilin-3 have been linked to human chondrodysplasias, while in mice deficiency in COMP or matrilin-3 does not cause any pronounced skeletal abnormalities. Given the similar function of COMP and matrilin-3 in the assembly and stabilization of the extracellular matrix, it is possible that these proteins can functionally compensate for each other. To assess this putative redundancy we generated COMP/matrilin-3 double deficient mice. At the newborn stage their overall skeletal development was normal but at one month of age the long bones were shortened and the total body length reduced. Peripheral quantitative computed tomography measurements revealed an increased metaphyseal trabecular mineral density in the femora of double mutants. Moreover, the degradation of aggrecan in the cartilage remnants in the metaphyseal trabecular bone was delayed, paralleled by an increased deposition of TIMP-3 in the growth plate cartilage. The structure and morphology of the growth plate was grossly normal, but in the center focal closures were observed, a phenotype very similar to that reported in MMP-13 deficient mice. We propose that lack of COMP and matrilin-3 leads to an increased deposition of TIMP-3, which causes partial inactivation of MMPs, including MMP-13, a mechanism that would explain the similarities in phenotype between COMP/matrilin-3 and MMP-13 deficient mice.  The processing and degradation of matrilins during cartilage remodelling in the growth plate of developing long bones was studied in greater detail. The two aggrecanases ADAMTS-4 and ADAMTS-5 have been shown to play roles in the breakdown of cartilage extracellular matrix in osteoarthritis, but are also involved in intracellular processing of matrilins. The matrilins are adaptor proteins with a function in connecting fibrillar and network-like components in the cartilage extracellular matrix.  Cleavage resulting in matrilin forms with fewer ligand-binding subunits could make them less efficient in providing matrix cohesion. We show that ADAMTS-5 and a matrilin-4 neoepitope, revealed upon ADAMTS cleavage, colocalize in prehypertrophic/hypertrophic chondrocytes in the growth plate of maturing long bones of mouse while they are not detected in proliferating chondrocytes. The ADAMTS-5 and the cleaved matrilin-4 are preferentially detected in vesicles derived from late portions of the Golgi apparatus. In the absence of ADAMTS-5 no matrilin-4 processing takes place in the growth plate. We propose that in the growth plate ADAMTS-5, but not ADAMTS-4, have a physiological function in the intracellular processing of matrilins.
