Abstract  Nesprins are a recently characterized protein family of spectrin repeat containing proteins present in multiple sub-cellular locations including the outer and inner membranes of the nucleus, the nucleoplasm, in association with mitochondria, Golgi apparatus, and sarcoplasmic reticulum, in the muscle sarcomere including the Z-line, A/I junction, M-line and plasma membrane. Alternate initiation, termination and splicing give rise to a multitude of isoforms variously composed of a spectrin repeat containing rod domain linked in the giant isoforms to N-terminal paired calponin homology domains that bind actin and a C-terminal transmembrane KASH (Klarsicht, ANC-1, Syne homology) domain which mediates nuclear membrane anchorage and binding to SUN-domain proteins to form a bridging LINC complex that spans across the nuclear membranes and connects the nucleus to the cytoskeleton. Specific isoforms lacking the N-terminal ABDs are highly expressed in muscle and heart and bind to emerin and lamin A/C. Here I show that Nesprin-1 is broadly expressed in various human and mouse cell lines. Further, we provide evidence for the existence of Enaptin/Nesprin-1 Giant, the largest Nesprin-1 isoform with a predicted molecular mass of 1086 kDa, making it the largest non-titin protein known so far. I show that Nesprin-1 Giant is expressed in detectable levels in COS7, HeLa, human glioblastoma (A172) and differentiated mouse myoblasts (C2F3).  I show that Nesprin-1 variants that lack the KASH domain can also localize to the NE. Further, I provide evidence for a novel function of ABDs of Nesprin-1/-2 in their interaction with the N-terminus of Nesprin-3, possibly allowing the alignment of Nesprins along the ONM and forming a basket-like network on the cytoplasmic face of the ONM. I also show that the C-terminal construct of Nesprin-1 behaves in a dominant negative manner upon overexpression in HeLa cells and displaces endogenous lamin B1, emerin and LAP2 from the NE while the lamin A/C and Sun1 localization appears to be normal.  Finally, I characterized two new variants of Nesprin-1α1 that cause Emery Dreifuss muscular dystrophy (EDMD) - one carrying a missense mutation N323H and the other with a mutation in the 5’UTR (29A>G) and show that nuclei in the dermal fibroblasts from these patients show nuclear abnormalities typical of laminopathies along with mislocalization of Nesprins, emerin and lamins to folds, blebs and micronuclei. I also characterized a dermal fibroblast cell line from a patient suffering from Stiff Skin syndrome (SSS), a disease with unknown aetiology. I show that the nuclei in these fibroblasts also exhibit nuclear defects and slightly reduced levels of Nesprin-2 Giant compared to control. I provide evidence that nuclei from all three cell lines are hypersensitive to heat stress and present with extensive deformations upon heat shock, indicating a fragile scaffolding network that is susceptible to damage upon stress.                          Zusammenfassung Nesprine sind kürzlich beschriebene neuartige Mitglieder der Spektrinsuperfamilie. Sie sind riesige Proteine mit einer F-Aktin-Bindestelle am Aminoterminus (ABD) und einer Transmembrandomäne am Carboxylende, der KASH-Domäne (Klarsicht, ANC-1, Syne Homology), die eine Lokalisation in der Kernmembran ermöglicht und dort am Aufbau des sogenannten Linc-Komplexes beteiligt ist. Der Linc-Komplex stellt eine Verbindung zwischen dem Zellkern und dem Zytoplasma über die Kernmembran her, in der Sun-Proteine, die Proteine der inneren Kernmembran sind, mit den C-Termini von KASH-Domänen interagieren. Die Verbindung zwischen dem Zellkern und dem Zytoplasma wird als wichtige Komponente für die Verankerung des Zellkerns und seine Stabilität angesehen. Nesprine zeichnen sich durch sehr unterschiedliche subzelluläre Lokalisationen aus und wurden in der inneren und äußeren Kernmembran gefunden, im Zellkern, an den Mitochondrien, am Golgi, im Muskel im sarcoplasmatischen Retikulum, an der Z-Scheibe, der A/I-Bande, der M-Bande und an der Plasmamembran. Alternative Initiationsstellen, unterschiedliche Terminationsstellen und unterschiedliches Spleißen sind die Ursache für eine Vielzahl von Isoformen, die aus den unterschiedlichen Domänen der Nesprine aufgebaut sind.  In dieser Arbeit zeige ich, dass Nesprin-1 in verschiedenen menschlichen und murinen Zelllinien exprimiert wird und weise zum erstenmal die 1000 kDa Form des Proteins im Westernblot nach. Formen von Nesprin-1, die keine KASH-Domäne enthalten, lokalisieren ebenfalls an der Kernmembran. Ferner wird eine neuartige Interaktion der ABD des Nesprin-1 und Nesprin-2 mit dem N-Terminus von Nesprin-3 nachgewiesen. Nesprin-3 ist ein Nesprin, das zwar eine KASH-Domäne und Spektrindomänen besitzt, selbst aber keine ABD aufweist. Durch die Interaktion zwischen Nesprin-1 und Nesprin-2 könnte eine Netzwerk von Proteinen gebildet werden, das den Zellkern umhüllt. In einem weiteren Teil der Arbeit wurden Fibroblasten von Patienten untersucht, die Mutationen im Nesprin-1 Gen tragen und eine Emery-Dreifuss-Muskeldystrophie verursachen. Weiterhin wurden dermale Fibroblasten eines Patienten, der am Stiff-Skin-Syndrom leidet, analysiert. Alle diese Zellen zeichnen sich durch Veränderungen in der Kernform aus und durch veränderte Lokalisationen von Proteinen der Kernhülle. Funktionelle Tests zeigten ferner, dass diese Zellen gegenüber Stress eine veränderte Antwort aufweisen und weniger resistent sind als Wildtypfibroblasten.
