Su, I-hsin: Requirement of polycomb group gene Enx-1 in lymphocyte development. 2001Die Gruppe der Polycomb (PcG) Proteine sind ein wichtiger Bestandteil des Chromatinremodulierenden Komplexes in der Fruchtfliege Drosophila. Kürzlich wurde durch gezielteGen-Mutagenese gezeigt, daß PcG Proteine eine essentielle Funktion für die Entwicklung derLymphozyten inMäusen haben. Eine konditionale Inaktivierung des Enx-1 Gens, ein neues Gen ausder PcG Genfamilie, hat die wichtige Rolle von Enx-1 für die Lymphopoiese bestätigt. DieEntwicklung der B und T Lymphozyten in Enx-1 defizienten Mäusen wurde in den Vorläuferzellen,nämlich in den pro B- und pro T-Zellstadien, blockiert. Eine genauere Untersuchung hat ergeben, daßsich diese Entwicklungsblockade auf eine defekte V(D)J Rekombination der VH J558 Familiezurückführen läßt, wobei die Rekombination der proximalen VH  Gene, wie zum Beispiel VH 7183,nicht beeinträchtigt ist. Durch das Einkreuzen eines pre-rearrangierten VH DH JH  Transgens in diesenMausstamm wurde die Entwicklungsblockade wieder aufgehoben. Zusammengenommen unterstützendiese Ergebnisse die Hypothese, daß Enx-1 die Zugänglichkeit des Rekombinasekomplexes imImmunoglobulin (Ig) Locus kontroliert.Polycomb group (PcG) proteins, which are important components of the chromatin remodelingcomplex in Drosophila, have recently been identified as essential regulators in mouse lymphocytedevelopment by gene targeting experiments. A conditional inactivation of Enx-1 gene, a novel memberof PcG genes, reveals the essential role of Enx-1 in lymphopoiesis. Both B and T cell development ofEnx-1 deficient mice is arrested in an early stage, where the B and T cells are rearranging their Ig andTCR loci, and assembling their B and T cell receptors. The developmental block of B cells is largelydue to a problem in VH  to DJH  recombination, which is supported by semi-quantitative PCR analysisof rearranged DNA. In addition, the B cell development is rescued by a pre-rearranged heavy chaintransgene. Moreover this recombination defect is restricted to VH J558 family genes, which are locatedat the 3¢ end of the VH  region, distal to the D element, whereas the recombination of the 5¢ VH  genesuch as VH  7183 remain unaffected. This leads to the suggestion that Enx-1 might control theaccessibility of the recombinase complex to the Ig locus in a position specific manner.
