Juraj Velcicky: Transition metal mediated, enantioselective synthesis of novel carbocyclicnucleoside analogs with strong apoptosis-inducing properties. 2003Die vorliegende Arbeit beschreibt die enantioselektive Synthese von zwei neuartigen Typen3´-substituierten-2´,3´-ungesättigten carbozyklischen Nukleosidanaloga (CNAs). In dieser Dissertationwurden 19 neuen CNAs hergestellt, von denen 14 in beiden enantiomeren Formen synthetisiertwurden. Bei diesen insgesamt 33 Derivaten zeigten 13 eine starke Apoptose- induktion in Tumorzellenmit einem Wert von LD50 < 50 M.Die neu entwickelte Synthese ist kurz (nur 11 bis 13 Stufen vom Propargylalkohol), hoch effizient (dieEndprodukte sind in bis zu 34% Ausbeute ohne kinetische Racematspaltung erhalten) und hochstereoselektiv. Zwei Schlüsselreaktionen sind Übergangsmetall (Kobalt und Palladium) vermittelt. Dieerste Kobalt-vermittelte Pauson-Khand (PK) Reaktion wurde für die Konstruktion eineshochsubstituierten bizyklischen Intermediates von einem einfachen und leicht zugänglichenAusgangsmaterial benutzt. Das racemische PK-Produkt wurde effizient mit Hilfe vonCorey-Bakshi-Shibata (CBS) Reduktion vermittelter kinetische Racematspaltung gespalten. Derbeobachtete Selektivitätsfaktor (s = 25-40) deutet eine hohe Effizienz des Prozesses an, da daslangsamer reagierende Enantiomer in einem hohen e.e. (>99%) nach nur 56-61% Umsatz gewonnenwurde. Die absolute Konfiguration der Produkte der kinetischen Racematspaltung wurde zuerst vomCorey-Model für CBS-Reduktion vorausgesagt und später mit Hilfe von drei unabhängigen Methoden(CD-Spektroskopie, Mosher Ester Analyse sowie Vergleich der optischen Drehungen mitliteraturbekannten Verbindungen) bestimmt.Die zweite Schlüsselreaktion ist die Pd-katalysierte allylische Substitution, die die Einführung vonfünf natürlichen und zwei modifizierten Nukleobasen ermöglicht. Die Kompatibilität dieseskonvergenten Prozesses und die spätere Einführung verschiedener Substituenten in die 4´-Position deszentralen Bausteins bietet einen Zugang zu verschiedenen CNAs in späten Stufen der Synthese.In der Synthese der eisenenthaltenen CNAs wurde eine diastereoselektive Komplexierung benutzt,wobei eine signifikante Diastereoselektivität (2.2 bis 4.1:1) als Folge der sterischen Hinderung voneiner der ?-Seiten des Diens beobachtet wurde. Die relative Konfiguration der gewonnenen Komplexewurde mit Hilfe von drei Methoden (Dipolmoment Model, CD- und NMR- Spektroskopie) bestimmt.Zusammenfassend konnten in dieser Arbeit zwei neue Typen enantiomerreiner, stereochemischvolldefinierter und stark apoptose-induzierender CNAs effektiv synthetisiert werden. This work describes the enantioselective synthesis of two novel types of 3´-substituted-2´,3´-unsaturated carbocyclic nucleoside analogs (CNAs). During this thesis 19 new CNAswere prepared, 14 of which were synthesized in both enantiomerical forms. Among the 33 derivatives,13 exhibit a strong apoptosis induction with LD50 < 50 M in cancer cells.The newly developed synthesis is short (only 11 to 13 steps starting from propargyl alcohol), highlyefficient (the final CNAs are obtained in up to 34% overall yield - excluding the resolution step), andhighly stereoselective.Two key-steps of the synthesis are mediated by transition metals (Cobalt and Palladium). First, aCobalt-mediated Pauson-Khand (PK) reaction was applied for the construction of the highlysubstituted key-bicyclic intermediate starting from a simple and easily available precursor. Theracemic PK-product was efficiently resolved, employing a kinetic resolution by means of aCorey-Bakshi-Shibata (CBS) reduction. The observed selectivity factors (s = 25-40) indicate the highefficiency that allows to obtain the slower reacting enantiomer in a very high e.e. (>99%) after "only"56-61% conversion. The absolute configuration of the products of the kinetic resolution was firstassigned from the Corey-model for CBS- reduction and later confirmed by 3 independent methods(CD-spectroscopy, Mosher´s ester analysis and by comparison of optical rotations with known compounds).The second key-reaction is a Pd-catalyzed allylic substitution allowing the attachment of all 5 naturalplus 2 modified nucleobases. The compatibility of this convergent process for the further introductionof different substituents into the 4´-position the central building block offers access to a large numberof diverse CNAs in a late stage of the synthesis.In the synthesis of iron-containing CNAs, the diastereoselective ?-facial complexation of the dienes byirontricarbonyl-group proceeded with a significant diastereoselectivity (2.2 to 4.1:1) caused by asterical hindrance of the substituents close to diene. The relative configuration of the producedcomplexes was confirmed by three methods (dipole-moment model, CD- and NMR-spectroscopy).In conclusion, 2 types of new, enantiomerically pure and stereochemically well defined CNAs wereprepared, many of which showed a strong apoptosis- inducing activity.
