Ingrid Mecklenbräuker: The role of protein kinase Cdelta in Bcell immunity . 2001Proteinkinase Cd (PKCd) ist eine Serin/Threoninkinase, die in der Regulierung von Wachstum, aberauch für den Zelltod in verschiedenen Zelltypen, wie auch in den weißen Blutkörperchen, eineentscheidende Rolle spielt. Darüberhinaus wurde gezeigt, daß PKCd auch an der Antigenrezeptorvermittelten Signaltransduktion von B and T Zellen beteiligt ist. Die Beteiligung von PKCd in BZellentwicklung und -funktion und besonders die Rolle in der B Zellimmunität wurde in dieser Arbeitan Hand von PKCd-defizienten Mäusen untersucht. Das Fehlen von PKCd zeigte sich in einerZunahme von peripheren B Zellen und in der Produktion von polyreaktiven Antikörpern. Auch wurdegezeigt, daß PKCd eine wichtige Rolle in der negativen Selektion von autoreaktiven B Zellen spielt.Zusammengenommen hat PKCd eine wichtige Rolle in der B Zellantwort auf tolerante, aber nicht aufimmunogene Antigene, wobei PKCd nicht nur am B Zellrezeptor vermittelten Signalweg, sonderndarüber hinaus auch an anderen Rezeptorvermittelten Signalwegen, wie CD38 und RP105, beteiligtist. Protein kinase Cd (PKCd) is a serine-threonine kinase which was found to be involved in theregulation of growth as well as death in a number of cell types, particularly in lymphocytes. Inaddition, PKCd is likely to be involved in antigen-receptor mediated signaling in B and T cells.The possible involvement of PKCd in B cell development and function with a special focus on the roleof PKCd in B cell immunity has been studied in pkCd -/- mice. The lack of PKCd resulted in anincrease in peripheral B cell numbers. Furthermore, the increase in peripheral cells was accompaniedby an increase in the numbers of antibody producing cells and high levels of polyreactive antibodies.Moreover, PKCd plays an important role in the negative selection of autoreactive B cells which werein the absence of PKCd not anergized. Collectively, the results of my experiments show the essentialrole of PKCd in tolerogenic but not immunogenic B cell responses in mice, not only downstream ofthe BCR but also of other receptor proteins like RP105 and CD38.
