                                                                                                                                       AbstractAbstractThis study investigates the impact of environmental adaptations of reproductive biology ongeneflow and speciation processes in salamanders of the genera Salamandra andNeurergus.Chapter 1 describes the succesful cloning of polymorphic microsatellite loci forSalamandra salamandra by using a PCR based enrichment protocol. In contrast to classicalprotocols, repeated enrichments and subsequent hybridization with a (GATA)10 probe yielded8 polymorphic tetra nucleotide loci for S. salamandra.Chapter 2 focusses on habitat specific adaptive divergence of pond and streamreproducing populations of S. salamandra near Cologne. Field data on the abundance ofpotential food organisms and stomach content of the salamander larvae demonstrated adistinctly lower amount of food in the ponds than in the streams. Pond larvae were larger athatching and metamorphosed in the field at a smaller size than the larvae from the stream-dwelling populations. Common environment experiments of both types confirmed the smallersize and reduced body weight of pond larvae when food was limited, whereas stream larvaeneeded up to three times longer to finish metamorphosis. The predominant role ofenvironmental plasticity causing differences in life history traits such as metamorphosis ischallenged by significant differences of pond and stream breeding populations undercommon environmental conditions. The fact that pond and stream breeding are derivedpostglacially from an identical founder lineage suggests, that fastly evolved differences arerather selective advantageous habitat specific adaptations.By including several populations from adjacent areas monophyletic origin of pond andstream reproducing S. salamandra in the Kottenforst, a forest near Bonn, is suggested inchapter 3 on the basis of 11 microsatellite loci. Strong association of nuclear geneflow eitherwith pond or stream adaptations under fully sympatric conditions and weak support ofisolation by distance favors the view that fire salamanders of the Kottenforst are in theprocess of incipient speciation under sympatric conditions.Molecular analysis of stream and pond breeding Near East Neurergus species inchapter 4 stress the longtime influence of environmental adaptation on speciation, but alsoon external morphology and behaviour. Molecular analysis of mitochondrial DNA and nuclearcoded allozymes of all extant Neurergus species suggest that pond breeding of N. kaiseri isthe result of an intrageneric switch of reproductive biology of a monophyletic stream breedinglineage more than 5 million years ago. The evolution of ecotypes within Neurergus is anexample of how a habitat switch is paralleled by a dramatic change in larval and adultmorphology, while remaining in the original habitat results in morphological stasis.                                                                                                                     ZusammenfassungZusammenfassungDie vorliegende Studie untersucht den Einfluß lebensraumspezifischer Anpassungen derReproduktionsbiologie auf Genfluß und Artbildungsprozesse bei Salamandern der GattungenSalamandra und Neurergus.In Kapitel 1 wird die erfolgreiche Klonierung polymorpher Mikrosatelliten-Loci fürSalamandra salamandra anhand einer PCR-basierten Anreicherungsmethode beschrieben.Im Gegensatz zu bisherigen Methoden gelang es mit dieser Methode, durch wiederholtePCR-Amplifikationen und anschließende Hybridisierung mit einer (GATA)10 Sonde 8polymorphe Tetra-Nukleotid-Loci zu isolieren.Kapitel 2 befaßt sich mit der adaptativen Aufspaltung von sich in Stillgewässern undBächen fortpflanzenden S. salamandra Populationen bei Köln. Abundanzdaten möglicherFuttertiere im Freiland und Mageninhaltsuntersuchungen zeigten eine relativeFutterknappheit in bewohnten Stillgewässern im Vergleich zu den Bächen. DasGeburtsgewicht der Stillgewässer-Larven war größer als das der Bach-Larven, zudem wardie Metamorphosegröße der Stillgewässer-Larven geringer. Common EnvironmentExperimente bestätigten diese Trends: unter Futtermangel reduzierten die Stillgewässer-Larven drastisch ihre Metamorphose-Größe und -Gewicht. Die Bach-Larven brauchtendreimal solange für das Erreichen der Metamorphose. Der starke Einfluß phänotypischerPlastizität auf Life History Traits wie die Metamorphose wird durch die signifikantenUnterschiede der Stillgewässer- und Bach-Populationen unter einheitlichen ökologischenBedingungen in Frage gestellt. Da sich Stillgewässer- und Bach-Populationen auf einegemeinsame postglaziale Gründerlinie zurückführen lassen, handelt es sich eher umvorteilhafte genetisch fixierte lebensraumspezifische Anpassungen.In Kapitel 3 wird der monophyletische Ursprung sich in Stillgewässern oder Bächenfortpflanzender Feuersalamander aus dem Kottenforst nahe Bonn unter Einbezugangrenzender Populationen auf der Basis von 11 Mikrosatelliten-Loci nachgewiesen. Untersympatrischen Bedingungen ist der nukleare Genfluß stark mit der Anpassung anStillgewässer bzw. an Bäche korreliert, wobei geographische Entfernung keinen Einfluß aufden Genfluß hat. Dies legt nahe, daß es sich bei S. salamandra im Kottenforst um einenunmittelbaren Artbildungsprozess unter sympatrischen Bedingungen handelt.Die molekulare Analyse von sich in Bächen und Stillgewässern fortpflanzendenNeurergus im Nahen Osten in Kapitel 4 zeigt die Langzeitauswirkunglebensraumspezifischer Anpassungen auf Artbildung, Morphologie und Verhalten. DieReproduktion von N. kaiseri in Stillgewässern ist das Resultat einer gattungsinternenUmstellung einer monophyletischen Bach-Linie vor mehr als 5 Millionen Jahren und steht mitdrastischen morphologischen Anpassungen an den Lebensraum in Verbindung.
