Principles and development of yeast two-hybrid system

Abstract

Međuproteinske interakcije igraju temeljnu ulogu u obnašanju različitih bioloških funkcija, pa je stoga njihova identifikacija i karakterizacija ključna za razumijevanje biologije stanice. Metoda dvostrukog kvaščevog hibrida domišljata je tehnika molekularne genetike koja omogućava jednostavnu detekciju velikog broja proteinskih interakcija in vivo. Ovaj rad daje osvrt na temeljne principe sustava dvostrukog kvaščevog hibrida, njegove prednosti i nedostatke, te neke od popularnih modifikacija i novih primjena ove metode.Protein-protein interactions play a major role in various biological mechanisms. Therefore, the identification and characterization of such interactions is the key to understanding cell biology. Yeast two-hybrid system is an inventive molecular genetic method which enables us to easily detect a great number of protein interactions in vivo. This paper offers an overview of basic principles of yeast two-hybrid system, its advantages and disadvantages, and some of the popular modifications and novel applications of the method
