Az anyagcsere homeostasist szabályozó hypothalamikus neuronrendszerek szerepe a reprodukció központi vezérlésében = The role of hypothalamic integrating centers of energy homeostasis in central regulation of reproduction

Abstract

A táplálkozást és a szaporodást szabályozó hypothalamikus központok működését a periféria felől érkező metabolikus szignálok és a jeleket feldolgozó és továbbító orexigén és anorexigén idegsejt csoportok befolyásolják. Morfológiai, molekuláris biológiai, farmakológiai és elektrofiziológiai módszerek együttes alkalmazásával tanulmányoztuk nevezett neuronális szabályozó rendszerek kapcsolatait és a jelátviteli mechanizmusokat. Figyelmünket az orexigén ghrelin, NPY, AGRP, valamint a CART és alfa-MSH tartalmú neuron rendszerekre és azoknak a reprodukciót szabályozó GnRH, valamint a metabolizmust és adaptációt reguláló TRH és CRH rendszerekkel kialakított kapcsolataikra terjesztettük ki. A perifériás hormonhatások közül az ösztrogén és pajzsmirigy hormonok agyi recepciójával és hatásmechanizmusaival foglalkoztunk. Kísérleti munkáinkról 31 tudományos közleményben adtunk számot, 117.71 összesített impakt értékkel. Eredményeink közül kiemeljük a hypothalamikus neuroszekretoros rendszerek glutamáterg fenotípusának felfedezését, a humán GnRH idegsejtek béta típusú ösztrogén receptoron keresztüli szabályozási lehetőségének felismerését, a metabolizmust szabályozó hypothalamikus rendszerek orexigén és anorexigén bemeneteinek jellemzését és eredtük feltárását, a humán agy táplálkozást szabályozó elsődleges hypothalamikus központjának kémiai és szerkezeti jellemzését, valamint a pajzsmirigy hormon aktiválásában résztvevő hypothalamikus szintű szabályozó mechanizmusok feltárását. | Peripheral metabolic signals are processed by orexigenic and anorexigenic neuronal networks of the basal hypothalamus that strongly influence the operation of neuroendocrine centers controlling reproduction and metabolism. By combination of structural functional tools, the networking and signal transduction mechanisms of the interacting neuronal systems have been studied. The scope of the work was focused on the orexigenic ghrelin, NPY, AGRP and the anorexigenic CART and alpha-MSH containing neuronal networks and their relationships established with GnRH, TRH and CRH systems, the known neuroendocrine regulators of reproduction, metabolism and adaptation. Regarding the hormonal effects, we have analyzed the receptors and processing of estrogen and thyroid hormone signals. The experimental results have been published in 31 papers with a cumulative impact value of 117.71. Results to be emphasized include the discovery of the glutamate-ergic phenotype of most hypothalamic neurosecretory systems, the structural proof of estradiol regulation of human GnRH neurons via estrogen receptor beta, the elucidation of the chemical nature and origin of the orexigenic and anorexigenic inputs onto hypothalamic neurosecretory systems regulating metabolism and reproduction, the characterization of the feeding centers of the human brain and finally, the cellular and biochemical mechanisms taking part in the activation and deactivation of thyroid hormones within the basal hypothalamus
