Abstract

A cystás fibrosis (CF) a leggyakoribb autoszomális, recesszív öröklődés menetet mutató halálos kimenetelű megbetegedés a fehérbőrű populációban. Hazánkban átlagosan 2500-3000 élveszületésre jut egy CF beteg. A CF-et a "cystic fibrosis transmembrane conductance regulator" (CFTR) fehérjét kódoló génben bekövetkezett mutáció okozza, amely vagy a fehérje teljes hiányához, vagy elégtelen működéséhez vezet. Kutatásaink során olyan cAMP-től független anion csatornát szerettünk volna találni, amely in vivo is helyettesíthetné a CFTR Cl- csatornát. Eredményeink azt mutatják, hogy a P2X purinerg receptor csatornák stimulálása alkalmas lehet e cél elérésére. Enyhén alkalikus közegben (pH 7.8-7.9), ATP és cink jelenlétében sikerült in vitro tartós és reverzibilis kálcium jelet kiváltanunk, amely a szignál ideje alatt folyamatos klorid szekrécióhoz vezetett CF légúti hámsejtekben. Annak érdekében, hogy a sejten kívüli térben magas koncentrációban lévő Na+ ne akadályozza a Ca2+ beáramlást a kísérleteket nátrium-szegény környezetben végeztük. Az in vitro nem-polarizált sejteken elvégzett kísérletek mellett in vivo kísérletekkel is igazoltuk a módszer hatékonyságát. CF egerek orrüregébe juttatott ATP-t és cinket tartalmazó, Na+ mentes, alkalikus oldat tartósan, reprodukálhatóan, de egyúttal reverzibilisen serkentette a lumenbe történő Cl- kiválasztást. | The cystic fibrosis (CF) is the most common autosomal recessive hereditary disease among the Caucasians. CF affects one newborn upon 2500-3000 life birth in Hungary each year. CF is caused by mutations in the gene encoding the cystic fibrosis transmembrane conductance regulator (CFTR) protein, which functions as a cAMP-dependent Cl- channel. Mutations in CF gene leads to either a complete loss or impaired function of CFTR protein. Our goal was to identify cAMP-independent anion channels that could substitute for CFTR not only in vitro experiment but in vivo studies as well. Our results demonstrate the stimulation of P2X purinergic receptor channels are suitable for this purpose. In slightly alkaline environment (pH 7.8-7.9) ATP and zinc induced an intracellular calcium signal which was reversible and reproducible. During this calcium signal Ca2+-activated Cl- channels were activated in CF airway epithelial cells. We performed these experiments in Na+-free environment in order to enhance Ca2+ entry through P2X receptor channels. Importantly, in vivo data confirmed our in vitro observations.
